"Wu zhu yu", which is obtained from the dried unripe fruits of Tetradium ruticarpum(A. Jussieu) T. G. Hartley, has been used as a traditional Chinese medicine for treatment of headaches, abdominal colic, and hypertension for thousands of years. The present study was designed to assess the molecular genetic diversity among 25 collected accessions of T. ruticarpum(Wu zhu yu in Chinese) from different areas of China, based on inter-primer binding site(i PBS) markers and inter-simple sequence repeat(ISSR) markers. Thirteen ISSR primers generated 151 amplification bands, of which 130 were polymorphic. Out of 165 bands that were amplified using 10 iPBS primers, 152 were polymorphic. The iPBS markers displayed a higher proportion of polymorphic loci(PPL = 92.5%) than the ISSR markers(PPL = 84.9%). The results showed that T. ruticarpum possessed high loci polymorphism and genetic differentiation occurred in this plant. The combined data of iPBS and ISSR markers scored on 25 accessions produced five clusters that approximately matched the geographic distribution of the species. The results indicated that both iPBS and ISSR markers were reliable and effective tools for analyzing the genetic diversity in T. ruticarpum.

Multi-components in herbal formulae exert holistic effects in synergistic or additive manners. However, appropriate strategies and supportive evidences are still lacking to uncover the synergistic or additive combinations. The present investigation aimed at seeking a screening strategy to identify the targeted combinations in GuG e FengT ong Tablet(GGFTT), an herbal formula. Two compounds, belonging to different chemical classes, were combined with different concentration ratios and their anti-inflammation effects were investigated. The most significant anti-inflammatory combinations were evaluated by combination index(CI) method(additive effect, CI = 1; synergism, CI 1). The modulating effects of candidate combinations on pro-inflammatory cytokines and MAPKs signaling pathway were also detected. Two combinations, "biochanin A + 6-gingerol"(Bio-6 G) and "genistein + 6-gingerol"(Gen-6 G), showed synergistic effects(CI herbal formulae.

Functional annotation map of natural compounds in traditional Chinese medicines library: TCMs with myocardial protection as a case

The chemical complexity of traditional Chinese medicines(TCMs) makes the active and functional annotation of natural compounds challenging. Herein, we developed the TCMs-Compounds Functional Annotation platform(TCMs-CFA) for large-scale predicting active compounds with potential mechanisms from TCM complex system, without isolating and activity testing every single compound one by one. The platform was established based on the integration of TCMs knowledge base, chemome profiling, and high-content imaging. It mainly included:(1) selection of herbal drugs of target based on TCMs knowledge base;(2) chemome profiling of TCMs extract library by LC-MS;(3) cytological profiling of TCMs extract library by high-content cell-based imaging;(4) active compounds discovery by combining each mass signal and multi-parametric cell phenotypes;(5) construction of functional annotation map for predicting the potential mechanisms of lead compounds. In this stud TCMs with myocardial protection were applied as a case study, and validated for the feasibility and utility of the platform. Seven frequently used herbal drugs(Ginseng, etc.) were screened from 100,000 TCMs formulas for myocardial protection and subsequently prepared as a library of 700 extracts. By using TCMs-CFA platform, 81 lead compounds, including 10 novel bioactive ones, were quickly identified by correlating 8089mass signals with 170,100 cytological parameters from an extract library. The TCMs-CFA platform described a new evidence-led tool for the rapid discovery process by data mining strategies, which is valuable for novel lead compounds from TCMs. All computations are done through Python and are publicly available on GitHub.

Objective:Phellodendron chinense(PC)bark and Phellodendron amurense(PA)bark are two herbal medicines recorded in the Chinese Pharmacopoeia(2020 edition)that are easily mistaken for one *** this study,the chemical constituents of PC and PA were compared using chromatographic fingerprints,and the potential synergistic acetylcholinesterase(ACh E)inhibitor components were screened based on the correlation of fingerprint *** and Methods:Chromatographic fingerprints based on high-performance liquid chromatography were developed for the analysis and comparison of chemical compounds in PC and PA *** ACh E inhibitory activity of PC and PA was determined using the Ellman ***,the contribution of the characterized alkaloids in PC and PA to the overall ACh E inhibition was modeled using partial least squares regression(PLSR).Results:The total alkaloid content in PC was higher than that in PA,which causes PC to have stronger anti-ACh E ***,13 and 20 common peaks were identified in the PC and PA samples,*** them,berberine(BER)was the dominant alkaloid in PC,which covered more than 65%of the total peak area in PC,but only approximately 25%of that in PA,indicating that the chemical composition is different between PC and *** spectrum–effect analysis based on PLSR and the correlation analysis showed that the BER-palmatine(PAL)and BER-jatrorrhizine(JAT)pairs have a synergistic inhibitory effect on ACh E ***:A high-performance chromatographic fingerprint was established to distinguish PC and *** efficacy-associated markers were screened,including the pairs of BER-PAL and BER-JAT with anti-ACh E activity,and the findings may assist with the quality control of PA and PC.

Irinotecan (CPT11) chemotherapy-induced diarrhea affects a substantial cancer population due to β-glucuronidase (Gus) converting 10-O-glucuronyl-7-ethyl-10-hydroxycamptothecin (SN38G) to toxic 7-ethyl-10-hydroxycamptothecin (SN38). Existing interventions primarily address inflammation and Gus enzyme inhibition, neglecting epithelial repair and Gus-expressing bacteria. Herein, we discovered that dehydrodiisoeugenol (DDIE), isolated from nutmeg, alleviates CPT11-induced intestinal mucositis alongside a synergistic antitumor effect with CPT11 by improving weight loss, colon shortening, epithelial barrier dysfunction, goblet cells and intestinal stem cells (ISCs) loss, and wound-healing. The anti-mucositis effect of DDIE is gut microbiota-dependent. Analysis of microbiome profiling data from clinical patients and CPT11-induced mucositis mice reveals a strong correlation between CPT11 chemotoxicity and Gus-expressing bacteria, particularly Enterococcus faecalis (E. faecalis). DDIE counters CPT11-induced augmentation of E. faecalis, leading to decreased intestinal Gus and SN38 levels. The Partial Least Squares Path Model (PLS-PM) algorithm initially links E. faecalis to dysregulated epithelial renovation. This is further validated in a 3D intestinal organoid model, in which both SN38 and E. faecalis hinder the formation and differentiation of organoids. Interestingly, colonization of E. faecalis exacerbates CPT11-induced mucositis and disturbs epithelial differentiation. Our study unveils a microbiota-driven, epithelial reconstruction-mediated action of DDIE against mucositis, proposing the ‘Gus bacteria–host–irinotecan axis’ as a promising target for mitigating CPT11 chemotoxicity. © 2024 The Authors

The discrimination of liquors plays a vital role in our daily life as a large amount of cheap alcoholic beverages and low-quality counterfeits greatly damage the health of ***-based optical sensors have been developed as a fast and effective tool to discriminate multi-analytes or complex mixtures with similar structures,such as acids,esters,aldehydes and *** the past decades,optical sensor arrays have demonstrated remarkable capabilities in fast and accurate detection of diverse types,qualities,and authentic *** identification strategy mainly focuses on the specific/non-specific interactions of flavor compounds in liquors with sensor *** cross-reactive optical signal readouts from the combination of sensor elements with liquors and multiple machine learning algorithms are essential to ensure the discriminatory capability of sensing *** this review,we discuss the advantages and recent advances in pattern recognition of liquors in terms of the design of various types of sensing materials and the construction strategies of sensor arrays.
