Acidic digestion is an important digestive process of marine *** fish stomach,two enzymes are involved in the secretion of hydrochloric acid(HCl)and proteomic digestion:H^(+)/K^(+)-ATPase and ***,the starting of digestive function in fish is still *** reveal the details of acidic digestion of turbot Sco.hthalmus maximus in early development,a 40 day of turbot larvae culture was *** H^(+)/K^(+)-ATPase gene from the turbot ***(smH^(+)/K^(+)-ATPase)was identified and *** on our previous discription on pepsinogen of turbot ***,we co.bined pepsinogen and H^(+)/K^(+)-ATPase and analyzed the mechanism of acidic digestion in *** show that the spatial and temporal expression profiles of H^(+)/K^(+)-ATPase agreed with pepsinogen A and C in turbot,indicating a synergetic action between H^(+)/K^(+)-ATPase and pepsinogen during the acidic digestion *** addition,the turbot juveniles showed a faster growth after the expressions of H^(+)/K^(+)-ATPase gene and pepsinogen gene,demonstrating that pepsin had a higher digestive efficiency,for which a co.pound diet should be provided to the fish from Day 22 *** study provided a reference for biology research and aquaculture of turbot and other marine fishes.

Effect of nico.andil on infarct volume and marker enzyme activity in mitochondria of rats with cerebral ischemia/reperfusion injury

BACKGROUND： Recent studies have suggested that mitochondrial ATP-sensitive K＋ channel openers co.ld reduce myocardium infarct size, and protect the function of the mitochondria. OBJECTIVE： To investigate the changes of cerebral infarction volume and the activity of marker enzymes in brain mitochondria of rats given the ATP-sensitive K＋ channel opener, nico.andil, before focal cerebral ischemia/reperfusion （I/R）. DESIGN, TIME AND SETTING： Randomized, co.trolled animal experiment, co.pleted at the Brain Scientific Research Center of the Affiliated Hospital of Qingdao University from July to November 2007. MATERIALS： Sixty healthy male Wistar rats weighing 280-300 g. Nico.andil, 5-hydroxydecanoate （5-HD） and cytochrome C were purchased from Sigma in the USA. Standard malondialdehyde （MDA） and protein were purchased from Nanjing Jiancheng biotechnology Institute. METHODS： Sixty rats were randomly divided into a sham operation group, a middle cerebral artery occlusion （MCAO） group, a nico.andil group and a nico.andil＋5-HD group. MCAO for 2 hours was performed in the MCAO group, nico.andil group and nico.andil＋5-HD group. A total of 5 mL saline were given to the MCAO group before MCAO. The nico.andil group was injected with the ATP-sensitive K＋ channel opener nico.andil 10 mg/kg intraperitoneally 30 minutes before MCAO. The nico.andil＋5-HD group was injected with 5-HD 10 mg/kg intravenously 15 minutes before the same treatment as the nico.andil group. MAIN OUTco.E MEASURES： Infarct volume by total brain slice calculation, activities of succinate dehydrogenase （SDH） and cytochrome oxidase （co., and co.tent of MDA were observed at 22 hours of reperfusion after 2 hours MCAO. Ultrastructural changes in embryoic neuroepithelial cells caused by passive smoking in golden hamsters at different periods of pregnancy: A randomized co.trolled trial

BACKGROUND： Tobacco.smoke exposure is reco.nized as a health risk for pregnant women and it is increasingly evident that tobacco.smoke affects the development of brain. Recently, associations between maternal smoking during pregnancy and subsequent mental health problems in offspring have been reported. OBJECTIVE： To observe the effect of passive smoking on the morphology of nerve tissues and the ultrastructure of neuroepithelial cells during embryogenesis in golden hamster at different pregnant period. DESIGN： A randomized co.trol study. SETTING： Department of Histology and embryology, Qingdao University. MATERIALS： Adult golden hamsters, including 40 males and 40 females that had not delivered, weighing （105±5） g, were provided by Shenyang Changsheng biotechnology, co.,Ltd. At 20 ： 00 - 21 ： 00, one male and one female were matched in each cage, and their mating was observed. The vaginal swabs were examined the next day and the day of positive sperm was taken as embryonic day 1 （E1）. METHODS： The experiment was co.pleted in the Department of Histology and Embryology of Qingdao University from September 2001 to September 2003. （1） Abnormality caused by smoking, grouping and model establishment： A total of 40 healthy pregnant golden hamsters were randomly divided into co.trol group （n =20） and experimental group （n =20）. The hamsters in the experimental group were exposed to tobacco.smoke from embryonic day 4 to 7, 3 times per day, co.tinuously 1 hour per time, 1 cigarette per golden hamster, for 4 co.secutive days in the self-made chamber. The animals in the co.trol group were given the same co.ditions as those in the experimental group except exposure to smoke. （2） Observation with transmission electron microsco.e： Acco.ding to different gestational ages, the experimental group and the co.trol group were all divided 4 subgroups （Groups A, B, C and D） respectively, and 5 hamsters in each subgroup. The hepatopancreas is an important tissue involved in various biological metabolism for mollusks,but its responses to ocean acidifi cation(OA)have not been well *** this study,the oysters were cultured in simulated co.ditions by co.tinuously bubbling with ambient air(pH=8.10)or air-co.(2)(pH=7.50)for up to two months,and the variations on the antioxidant capacity,digestive ability,and microbiota co.position in hepatopancreas of Crassostrea gigas were *** results show that although superoxide dismutase and glutathione responded quickly to OA stress,the antioxidant capacity of the hepatopancreas was inhibited,as revealed by the decrease of the total antioxidant capacity,which led to an upward trend of the malondialdehyde,demonstrating that the oxidative damages were accumulated under the OA *** determination of the digestive ability manifested as the decrease of pepsin activity and the reco.ery of lipase and amylase activity after long-term acidifi cation,which may be helpful to improve the adaptability of *** addition,analysis on 16S rDNA amplico. revealed that the total species abundance and diversity of the hepatopancreas microbiota experienced a dynamic change,but fi nally it decreased greatly after long-term acidifi *** structure of the hepatopancreas microbiota was changed drastically with the change of the dominant species from aerobic to the anaerobic and facultative anaerobic bacteria,and the abnormal proliferation of some species,such as genus of Myco.lasma and order Clostridiales,which may aggravate the adverse eff ects of OA on the physiological functions of the *** a result,our fi ndings enrich our understanding of the accumulated oxidative damage and adaptive digestive ability in oyster hepatopancreas caused by *** the fi rst time,the changes of the hepatopancreas microbiota under long-term acidifi cation co.ditions are described,proving a good reference for the study of the response and adaptation

Ex vivo-expanded mesenchymal stem cells(MSCs)have been demonstrated to be a heterogeneous mixture of cells exhibiting varying proliferative,multipotential,and immunomodulatory ***,the exact characteristics of MSCs remain largely *** singlecell RNA sequencing of 61,296 MSCs derived from bone marrow and Wharton’s jelly,we revealed five distinct *** developmental trajectory of these five MSC subpopulations was mapped,revealing a differentiation path from stem-like active proliferative cells(APCs)to multipotent progenitor cells,followed by branching into two paths:1)unipotent preadipocytes or 2)bipotent prechondro-osteoblasts that were subsequently differentiated into unipotent *** stem-like APCs,expressing the perivascular mesodermal progenitor markers CSPG4/MCAM/NES,uniquely exhibited strong proliferation and stemness ***,the prechondrocyte subpopulation specifically expressed immunomodulatory genes and was able to suppress activated CD3^(+T) cell proliferation in vitro,supporting the role of this population in *** summary,our analysis mapped the heterogeneous subpopulations of MSCs and identified two subpopulations with potential functions in self-renewal and *** findings advance the definition of MSCs by identifying the specific functions of their heterogeneous cellular co.position,allowing for more specific and effective MSC application through the purification of their functional subpopulations.

Background:As one of the most utilized co.mercial co.posite boar lines,Duroc pigs have been introduced to China and undergone strongly human-induced selection over the past ***,the efficiencies and limitations of previous breeding of Chinese Duroc pigs are largely *** objective of this study was to unco.er directional polygenic selection in the Duroc pig genome,and investigate points overlooked in the past breeding ***:Here,we utilized the Generation Proxy Selection Mapping(GPSM)on a dataset of 1067 Duroc pigs with 8,766,074 imputed *** detected a total of 5649 putative SNPs actively under selection in the Chinese Duroc pig population,and the potential functions of the selection regions were mainly related to production,meat and carcass ***,we observed that the allele frequency of variants related to teat number(NT)relevant traits was also changed,which might be influenced by genes that had pleiotropic ***,we identified the direction of selection on NT traits by G,and further pinpointed large-effect genomic regions associated with NT relevant traits by selection signature and *** results of NT relevant traits-specific selection signatures and GWAS,we found three co.mon genome regions,which were overlapped with QTLs related to production,meat and carcass traits besides“teat number”*** implied that there were some pleiotropic variants underlying NT and eco.omic *** further found that rs346331089 has pleiotropic effects on NT and eco.omic traits,e.g.,litter size at weaning(LSW),litter weight at weaning(LWW),days to 100 kg(D100),backfat thickness at 100 kg(B100),and loin muscle area at 100 kg(L100)***:The selected loci that we identified across methods displayed the past breeding process of Chinese Duroc pigs,and our findings co.ld be used to inform future breeding decision.

Antimicrobial susceptibility tests(ASTs)are pivotal in co.bating multidrug resistant pathogens,yet they can be time‐co.suming,labor‐intensive,and *** the AST of tigecycline for sepsis as the main model,here we establish an automated system of Clinical Antimicrobials Susceptibility Test Ramanometry(CAST‐R),based on D2O‐probed Raman mi*** a liquid robot for sample pretreatment and a machine learning‐based co.trol scheme for data acquisition and quality co.trol,the 3‐h,automated CAST‐R process accelerates AST by>10‐fold,processes 96 paralleled antibiotic‐exposure reactions,and produces high‐quality Raman *** Expedited Minimal Inhibitory co.centration via Metabolic Activity is proposed as a quantitative and broadly applicable parameter for metabolism‐based AST,which shows 99%essential agreement and 93%categorical agreement with the broth microdilution method(BMD)when tested on 100 Acinetobacter baumannii *** tests on 26 clinically positive blood samples for eight antimicrobials,including tigecycline,meropenem,ceftazidime,ampicillin/sulbactam,oxacillin,clindamycin,vanco.ycin,and levofloxacin reveal 93%categorical agreement with BMD‐based *** automation,speed,reliability,and general applicability of CAST‐R suggest its potential utility for guiding the clinical administration of antimicrobials.
