Horizontal gene transfer(HGT)has been well documented as a driving force in the evolution of *** has been shown that a horizontally acquired gene,xoc_2868,involved in the global response against oxidative stress and pathogenicity of Xanthomonas oryzae *** strain ***,as a transcriptional factor(TF),the regulatory mechanism of XOC_2868 has not yet been ***,evolutionary analysis suggested XOC_2868 might be co-transferred with its physically proximate downstream genes from a Burkholderiaceae ***,RNA-seq data of wild-type(BLS256)andΔxoc_2868 strains under oxidative stress showed that XOC_2868 did not regulate the expression of its adjacent genes,but remarkably influenced the expression of several genes involved in the extracellular polysaccharide(EPS)production and xanthan *** immunoprecipitation-sequence(ChIP-seq)combined with transcriptome analysis revealed that XOC_2868 directly regulates a cydAB operon,encoding two subunits of cytochrome bd oxidase and involved in redox *** withΔxoc_2868 strain,cydA-and cydAB-knockout mutants also showed a higher sensitivity to H_(2)O_(2)along with a reduced bacterial virulence compared with the wild-type ***,our findings raise the possibility of regulatory circuit evolution shaped by HGT and driven by selection and reveal a novel regulatory pathway that regulates the expression of cytochrome bd oxidase and thus contributes to the virulence of BLS256.

Photonics integration of an optoelectronic oscillator(OEO)on a chip is attractive for fabricating low cost,compact,low power consumption,and highly reliable microwave sources,which has been demonstrated recently in silicon on insulator(SOI)and indium phosphide(InP)platforms at X-band around 8 *** we demonstrate the first integration of OEOs on the thin film lithium niobate(TFLN)platform,which has the advantages of lower Vπ,no chirp,wider frequency range,and less sensitivity to *** have successfully realized two different OEOs operating at Ka-band,with phase noises even lower than those of the X-band OEOs on SOI and InP *** is a fixed frequency OEO at 30 GHz realized by integrating a Mach–Zehnder modulator(MZM)with an add-drop microring resonator(MRR),and the other is a tunable frequency OEO at 20–35 GHz realized by integrating a phase modulator(PM)with a notch *** work marks the first step of using TFLN to fabricate integrated OEOs with high frequency,small size,low cost,wide range tunability,and potentially low phase noise.

Hyperparameter optimization of YOLOv8 for smoke and wildfire detection:Implications for agricultural and environmental safety

In this study,we extensively evaluated the viability of the state-of-the-art YOLOv8 architecture for object detec-tion tasks,specifically tailored for smoke and wildfire identification with a focus on agricultural and environmen-tal *** available versions of YOLOv8 were initially fine-tuned on a domain-specific dataset that included a variety of scenarios,crucial for comprehensive agricultural ***‘large’version(YOLOv8l)was se-lected for further hyperparameter tuning based on its performance *** model underwent a detailed hyperparameter optimization using the One Factor At a Time(OFAT)methodology,concentrating on key param-eters such as learning rate,batch size,weight decay,epochs,and *** from the OFAT study were used to define search spaces for a subsequent Random Search(RS).The final model derived from RS demon-strated significant improvements over the initial fine-tuned model,increasing overall precision by 1.39%,recall by 1.48%,F1-score by 1.44%,mAP@0.50 by 0.70%,and mAP@0.50:0.95 by 5.09%.We validated the enhanced model's efficacy on a diverse set of real-world images,reflecting various agricultural settings,to confirm its ro-bustness in detecting smoke and *** results underscore the model's reliability and effectiveness in scenar-ios critical to agricultural safety and environmental *** work,representing a significant advancement in the field of fire and smoke detection through machine learning,lays a strong foundation for fu-ture research and solutions aimed at safeguarding agricultural areas and natural environments.

Mitochondrial dysfunction is a hallmark feature of cellular senescence and organ ***,we asked whether the mitochondrial antiviral signaling protein(MAVS),which is essential for driving antiviral response,also regulates human stem cell *** answer this question,we used CRISPR/Cas9-mediated gene editing and directed differentiation techniques to generate various MAVS-knockout human stem cell *** found that human mesenchymal stem cells(hMSCs)were sensitive to MAVS deficiency,as manifested by accelerated senescence *** uncovered that the role of MAVS in maintaining mitochondrial structural integrity and functional homeostasis depends on its interaction with the guanosine triphosphatase optic atrophy type 1(OPA1).Depletion of MAVS or OPA1 led to the dysfunction of mitochondria and cellular senescence,whereas replenishment of MAVS or OPA1 in MAVS-knockout hMSCs alleviated mitochondrial defects and premature senescence *** together,our data underscore an uncanonical role of MAVS in safeguarding mitochondrial homeostasis and antagonizing human stem cell senescence.
