Expression of airway mucus-associated proteins in rats with chronic obstructive pulmonary disease with a cold-dryness symptom pattern

OBJECTIVE: To reveal the effects on expression of airway mucus-associated proteins in rats with chronic obstructive pulmonary disease(COPD) and a cold-dryness symptom pattern induced by elastase and ***: The COPD model was established with an elastase dose into the trachea combined with exposure to smoking; the COPD model cold-dryness symptom pattern was further developed by exposure to a cold, dry environment. After 90 days,pathologic lung sections, inflammatory cytokine levels(measured by enzyme linked immunosorbent assay), m RNA and protein expression of mucus-associated proteins and aquaporins(measured by real-time polymerase chain reaction and western blots) were ***: Cytokines interleukin-6(IL-6), interleukin-8(IL-8), and tumor necrosis factor-α(TNF-α) in the COPD and the cold-dryness symptom pattern COPD groups were all significantly higher than in controls(each P < 0.01). Inflammatory responses of the rat lungs in cold-dryness syndrome in the northwest of China

OBJECTIVE:To examine changes in body weight and the lung inflammation factors interleukin-1β(IL-1β),interleukin-8(IL-8),IL-10 and tumor necrosis factor-α(TNF-α) in a rat model of cold-dryness syndrome in Northwest(xinjiang) China to provide a reference for treating chronic obstructive pulmonary disease(COPD) with local ***:The rat COPD model was established by intratracheal instillation of porcine pancreatic elastase(PPE) in combination with cigarette smoking(CS).The rat model of cold-dryness syndrome of COPD in the northwest of China was set up by intratracheal instillation of PPE in combination with CS and environmental cold-dryness *** level of IL-1β,IL-8,IL-10 and TNF-α in Bronchoalveolar lavage fluid(BALF) were measured by enzyme-linked immunosorbent assay(ELISA).The data were analyzed using the software SPSS ***:1) Body weight was less in the two model groups than that of control group(P<0.01),PPE plus CS cold-dryness group was less than that of PPE plus CS group(P<0.01).2) IL-1β in BALF significantly increased in PPE plus CS and cold-dryness group than that of control group(P<0.01).3) IL-8 and TNF-α in BALF significantly increased in PPE plus CS and cold-dryness group and PPE plus CS group than that of control group(P<0.01).CONCLUSION:Body weight in COPD model rats was reduced compared with ***-dryness may aggravate such a condition Lung inflammation in the model was mainly manifested by an increase in IL-1β,IL-8 and TNF-α levels,with no change in IL-10 ***-dryness may aggravate lung inflammation of COPD.

BACKGROUND: Dexmedetomidine has already been used in septic patients as a new sedative agent, few studies have examined its effects on immunomodulation. Therefore, the authors have designed a controlled experimental study to characterize the immunomodulation effects of dexmedetomidine in the cecal ligation and puncture(CLP) model in rats. METHODS: After CLP, 48 Wistar rats were randomly allocated into four groups:(1) CLP group;(2) small-dose treatment group(2.5 g·kg^(-1)·h^(-1));(3) medium-dose treatment group(5.0 g·kg^(-1)·h^(-1)); and(4) large-dose treatment group(10.0 g·kg^(-1)·h^(-1)). HLA-DR and plasma cytokine(IL-4, IL-6, IL-10 and TNF-α) levels were measured, and the mean arterial blood pressure(MAP), heart rate(HR), arterial blood gases, lactate concentrations and mortality were also documented. RESULTS: The HLA-DR level, inflammatory mediator levels, MAP and HR had no obvious changes among Dexmedetomidine treatment groups(DEX groups). Compared with the CLP group, the DEX groups exhibited decreased HLA-DR levels(P_(group)=0.0202) and increased IL-6 production, which was increased at 3 h(P= 0.0113) and was then attenuated at 5 h; additionally, the DEX groups exhibited decreased HR(P<0.001) while maintaining MAP(P_(group)=0.1238), and remarkably improving lactate(P<0.0001). All of these factors led to a significant decrease in the mortality, with observed rates of 91.7%, 66.7%, 25% and 18% for the CLP, DEX2.5, DEX5.0, DEX10.0 groups, ***: Dexmedetomidine treatment in the setting of a CLP sepsis rat model has partially induced immunomodulation that was initiated within 5 h, causing a decreased HR while maintaining MAP, remarkably improving metabolic acidosis and improving mortality dosedependently.

Results:In all 65 patients,41 cases (41/65,63.1%;Group 1)had normal immunity and 24 cases (24/65,36.9%;Group 2)had at least one identifiable underlying ***,higher percentage of neutrophil (NEUT)in white blood cell (WBC),and increased cell number of cerebral spinal fluid (CSF)were much common in patients with underlying disease (Group 1 *** 2:Fever:21/41 vs.21/24,x^2 =8.715,P =0.003;NEUT in WBC:73.15%vs.79.60%,Z=-2.370,P =0.018;cell number of CSF:19 vs.200,Z=-4.298,P <0.001;respectively).Compared to the patients with normal immunity,the lesions are more common in the basal ganglia among patients with identifiable underlying disease (Group 1 *** 2:20/41 vs.20/24,x^2 =7.636,P =0.006).The number of the involved brain areas in patients with identifiable underlying disease were well correlated with the number of cells and pressure of CSF (r =-0.472,P =0.031,r =0.779,P =0.039;respectively). Collagen,commonly used in tissue engineering,is widespread in various *** bone tissue regeneration,collagen can stimulate the cellular response and determine the fate of *** this work,we integrated collagen type II with procyanidin(PC)onto an implant coating by applying a layer-by-layer technique to demonstrate that collagen and PC can participate in the construction of new biomaterials and serve as multifunctional *** effects of PC/collagen multilayers on the viability of cocultured bone marrow mesenchymal stem cells(BMSCs)were analyzed by cell counting kit-8 analysis and phalloidin *** reactive oxygen species level of BMSCs was revealed through immunofluorescent staining and flow ***-related genes were detected,and in vivo experiment was performed to reveal the effect of newly designed material on the osteogenic differentiation of *** data demonstrated that in BMSCs PC/collagen multilayers accelerated the proliferation and osteogenic differentiation through Wnt/β-catenin signaling pathway and enhanced bone generation around the implant in the bone defect model of rabbit *** summary,combination of collagen and PC provided a new sight for the research and development of implant materials or coatings in the future.
