Autophagy-targeting modulation to promote peripheral nerve regeneration

Nerve regeneration following traumatic peripheral nerve injuries and neuropathies is a complex process modulated by diverse factors and intricate molecular *** studies have focused on factors that stimulate axonal outgrowth and myelin ***,recent studies have highlighted the pivotal role of autophagy in peripheral nerve regeneration,particularly in the context of traumatic ***,autophagy-targeting modulation has emerged as a promising therapeutic approach to enhancing peripheral nerve *** current understanding suggests that activating autophagy facilitates the rapid clearance of damaged axons and myelin sheaths,thereby enhancing neuronal survival and mitigating injury-induced oxidative stress and *** actions collectively contribute to creating a favorable microenvironment for structural and functional nerve regeneration.A range of autophagyinducing drugs and interventions have demonstrated beneficial effects in alleviating peripheral neuropathy and promoting nerve regeneration in preclinical models of traumatic peripheral nerve *** review delves into the regulation of autophagy in cell types involved in peripheral nerve regeneration,summarizing the potential drugs and interventions that can be harnessed to promote this *** hope that our review will offer novel insights and perspectives on the exploitation of autophagy pathways in the treatment of peripheral nerve injuries and neuropathies.

Comparison of semen quality and outcome of assisted reproductive techniques in Chinese men with and without hepatitis B

In this study, we aimed to determine the effects of hepatitis B virus （HBV） infection on sperm quality and the outcome of assisted reproductive technology （ART）. A total of 916 men （457 HBV-positive and 459 HBV-negative） seeking fertility assistance from January 2008 to December 2009 at the women＇s Hospital in the school of Medicine at Zhejiang University were analysed for semen parameters. Couples in which the men were hepatitis B surface antigen （HBsAg）-seropositive were categorized as HBV-positive and included 587 in vitrofertilisation （IVF） and 325 intracytoplasmic sperm injection （ICsI） cycles from January 2004 to December 2009; negative controls were matched for female age, date of ova retrieval, ART approach used （IVF or ICsI） and randomized in a ratio of 1：1 according to the ART treatment cycles （587 for IVF and 325 for ICsI）. HBV-infected men exhibited lower semen volume, lower total sperm count as well as poor sperm motility and morphology （P〈O.05） when compared to control individuals. Rates of two-pronuclear （2PN） fertilisation, high-grade embryo acquisition, implantation and clinical pregnancy were also lower among HBV-positive patients compared to those of HBV-negative patients after ICsI and embryo transfer （P〈0.05）; IVF outcomes were similar between the two groups （P〉0.05）. Logistic regression analysis showed that HBV infection independently contributed to increased rates of asthenozoospermia and oligozoospermia/azoospermia （P〈0.05） as well as decreased rates of implantation and clinical pregnancy in ICsI cycles （P〈0.05）. Our results suggest that HBV infection in men is associated with poor sperm quality and worse ICsI and embryo transfer outcomes but does not affect the outcome of IVF and embryo transfer.

An overview of studies on psychological well-being in children born following assisted reproductive technologies

Over the course of the past 35 years,assisted reproductive technologies(ARTs)have been increasingly used worldwide,while debates on their safety have been *** defects and imprinting disorders were reported in previous ***,the psychological development of children born following ARTs has become a major concern *** review gives a systematic view of psychological well-being of children conceived by different types of ART,including in vitro fertilization,intracytoplasmic sperm injection(ICsI),preimplantation genetic diagnosis/screening,and in vitro *** previous studies are analyzed in three sections:(1)cognitive,motor,and language developments,(2)behavior problems and socio-emotional development,and(3)parent-child *** conclude that although the majority of the studies on cognitive,motor,and language developments reported comparable achievements in the ART group *** naturally conceived group,lower intelligence quotient(IQ)scores,worse visual-motor ability or locomotor development,and delayed receptive language competence were found in the ART *** results on the socio-emotional development were *** for the behavior problems,a higher prevalence of behavior problems existed in ART children;moreover,ICsI children were found to be at a higher risk of autism than the general ***,ART parents tended to have positive parental attitudes and be more protective of their *** suggestions for further research are also given in this review.

Ovarian endometrioma（OE）, also known as “chocolate cysts,” is a cystic mass that develops in the ovaries due to endometriosis and is a common gynecological condition characterized by the growth of endometrial tissue outside the uterus, leading to symptoms such as dysmenorrhea, pelvic pain, and infertility. However, the precise molecular and cellular mechanisms driving this pathophysiology remain largely unknown, posing challenges for diagnosis and treatment. Here, we employed integrated single-cell transcriptomic profiling of over 52,000 individual cells from endometrial tissues of OE patients and healthy donors and identified twelve major cell populations. We identified notable alterations in cell type-specific proportions and molecular signatures associated with OE. Notably, the activation of IGFBP5+macrophages with pro-inflammatory properties, NK cell exhaustion, and aberrant proliferation of IQCG+and KLF2+epithelium are key features and may be the potential mechanisms underlying the pathogenesis of OE. Collectively, our data contribute to a better understanding of OE at the single cell level and may pave the way for the development of novel therapeutic strategies.

Accuracy of a combined score of zygote and embryo morphology for selecting the best embryos for IVF

Objective:To evaluate the accuracy of a scoring system combining zygote and embryo morphology in predicting the outcome of in vitro fertilization (IVF) treatment. Methods: In a study group, 117 consecutive IVF or intracytoplasmic sperm injection (ICsI) cycles with embryo transfer were carried out and 312 embryos were scored using a combined scoring system (Css) of zygote and embryo morphology before transplantation. In a control group, a total of 420 IVF or ICsI cycles were carried out and 1176 embryos were scored using a cumulative embryo score (CEs). The effects of the combined scoring system on the embryo implantation rate and pregnancy rate per cycle were analyzed. Results: Using the combined scoring system, the embryo implan-tation rate (27.6%) and the clinical pregnancy rate (48.7%) were significantly higher than those in the control group (20.8% and 38.6%, respectively). Also, the implantation rate of embryos scoring ≥70 (38.5%: 82 sacs/213 embryos) was significantly higher (Ps scoring sacs/99 embryos). The pregnancy rate of patients with embryos scoring ≥70 using the combined scoring system (66.7%) was significantly higher (Ps with embryos scoring ≥20 using the cumulative embryo score (59.0%). Conclusion: The results suggest that selecting embryos with a high score (≥70) using the combined scoring system could increase the implantation rate and pregnancy rate, and that using a scoring system combining assessments of human zygotes and pre-implantation embryos might predict IVF outcomes more accurately than using a cumula-tive embryo score.

Antigen unmasking does not improve the visualization of phospholipase C zeta in human spermatozoa

Phospholipase C zeta(PLCζ)is a sperm-specific protein that triggers oocyte *** analysis of PLCζexpression in human spermatozoa can be used as a diagnostic marker for oocyte activation *** laboratory has previously optimized a standard"in-house"assay to determine PLCζexpression in human ***,one study has suggested that an antigen unmasking method(AUM)would be more efficient in visualizing PLCζin human *** study aimed to compare our established assay and AUM(involving HCl,acidic Tyrode's solution[AT],and heat).The mean relative fluorescence(RF)intensity of PLCζin frozen-thawed spermatozoa from fourteen fertile donors stained with the in-house method was significantly higher than three other AUM groups(in-house[mean±standard error of mean]:18.87±2.39 arbitrary units[a.u.]vs non-AUM:11.44±1.61 a.u.,AT-AUM:12.38±1.89 a.u.,and HCl-AUM:12.51±2.16 a.u.,Psis of variance).The mean RF intensity of PLCζin AT-and HCl-treated spermatozoa from 12 infertile males was not significantly different from that of the non-AUM ***,the in-house method resulted in the highest RF intensity(12.11±1.36 a.u.,Pspecificity testing of antibody-antigen binding indicated that the in-house method showed more specific binding than spermatozoa treated by the *** conclusion,our in-house method showed superior visualization and reliability than the AUM,thus supporting the continued use of our in-house assay for clinical research screening.
