Low crude protein formulation with supplemental amino acids for its impacts on intestinal health and growth performance of growing-finishing pigs

Ursolic acid inhibiting excessive reticulophagic flux contributes to alleviate ochratoxin A-induced endoplasmic reticulum stress- and mitochondrial-mediated apoptosis

Foods and animal feeds frequently become contaminated with the nephrotoxic ochratoxin A (OTA). Our prior research has indicated that ursolic acid (UA)， which is widely present in fruits and medicinal plants， has the potential to alleviate nephrotoxicity triggered by OTA. Additionally， excessive induction of endoplasmic reticulum (ER)-phagy exacerbates OTA-induced apoptosis. Therefore， further investigation is essential to comprehend whether UA can mitigate OTA-induced apoptosis by influencing ER-phagy. This objective is accomplished through a series of experiments involving assessments of cell viability， apoptosis， fluorescence microscopy， and western blot analysis. The outcomes of these experiments reveal that pre-treatment with 4 μmol/L UA for 2 h can markedly reverse the elevated apoptotic rate， the co-localization of ER and lysosomes， and the protein expressions of GRP78， p-eIF2α， Chop， Bax， and Bak， as well as the reduced cell viability and the protein expressions of Lonp1， Trap1， p62， Tex264， FAM134B， Bcl-2， and Bcl-xl， all caused by exposure to 1 μmol/L OTA for 24 h in human proximal tubule epithelial-originated kidney-2 (HK-2) cells (P increased expression of LC3B-II induced by OTA is further amplified by UA pre-treatment (P < 0.05). In conclusion， OTA triggers a harmful feedback loop between ER stress (ERS) and excessive ER-phagy， thereby further promoting ERS- and mitochondrial-mediated apoptosis in vitro. However， this effect is significantly mitigated by UA through the inhibition of autophagosome-lysosome fusion， consequently blocking the excessive ER-phagic flux.

Effects of essential oils on economically important characteristics of ruminant species: A comprehensive review

Essential oils derived from plants can provide biological impacts to livestock species. Scientific studies researching essential oils in livestock have investigated various essential oils for prevention and treatment of microbial infection and parasites as well as to enhance milk production, animal performance and rumen function. Despite the availability of several commercial products containing essential oils to promote animal health and production, the vast amount of essential oils, modes of application, and effective concentrations of the essential oils suggest there are more opportunities for essential oils to be utilized in commercial livestock production and veterinary medicine. The objective of this review is to contribute to the understanding of the value that essential oils can provide to the ruminant diet and to examine the biological impact of various essential oils on economically important production traits of ruminant species.

Interactions between maternal parity and feed additives drive the composition of pig gut microbiomes in the post‑weaning period

Background Nursery pigs undergo stressors in the post-weaning period that result in production and welfare *** challenges disproportionately impact the offspring of primiparous sows compared to those of mul-tiparous *** is known regarding potential interactions between parity and feed additives in the post-weaning period and their effects on nursery pig ***,the objective of this study was to investigate the effects of maternal parity on sow and offspring microbiomes and the influence of sow parity on pig fecal microbi-ome and performance in response to a prebiotic *** weaning,piglets were allotted into three treat-ment groups:a standard nursery diet including pharmacological doses of Zn and Cu(Con),a group fed a commercial prebiotic only(Preb)based on an Aspergillus oryzae fermentation extract,and a group fed the same prebiotic plus Zn and Cu(Preb+ZnCu).Results Although there were no differences in vaginal microbiome composition between primiparous and mul-tiparous sows,fecal microbiome composition was different(R^(2)=0.02,P=0.03).The fecal microbiomes of primiparous offspring displayed significantly higher bacterial diversity compared to multiparous offspring at d 0 and d 21 post-weaning(Pinc-tions between primiparous and multiparous offspring(d 21:R^(2)=0.13,P=0.01;d 42:R^(2)=0.19,P=0.001).Composi-tional differences in pig fecal microbiomes between treatments were observed only at d 21(R^(2)=0.12,P=0.001).Pigs in the Con group gained significantly more weight throughout the nursery period when compared to those in the Preb+ZnCu *** Nursery pig gut microbiome composition was influenced by supplementation with an Aspergillus oryzae fermentation extract,with varying effects on performance

Research Background and Purpose: The number of diabetic patients is rapidly increasing, making it crucial to find methods to prevent diabetic retinopathy (DR), a leading cause of blindness. We investigated the effects of prophylactic pattern scanning laser retinal photocoagulation on DR development in Spontaneously Diabetic Torii (SDT) fatty rats as a new prevention approach. Methods: Photocoagulation was applied to the right eyes of 8-week-old Spontaneously Diabetic Torii (SDT) fatty rats, with the left eyes serving as untreated controls. Electroretinography at 9 and 39 weeks of age and pathological examinations, including immunohistochemistry for vascular endothelial growth factor and glial fibrillary acidic protein at 24 and 40 weeks of age, were performed on both eyes. Results: There were no significant differences in amplitude and prolongation of the OP waves between the right and left eyes in SDT fatty rats at 39 weeks of age. Similarly, no significant differences in pathology and immunohistochemistry were observed between the right and left eyes in SDT fatty rats at 24 and 40 weeks of age. Conclusion: Prophylactic pattern scanning retinal laser photocoagulation did not affect the development of diabetic retinopathy in SDT fatty rats.

We investigated the effects of adding up to 11% rice bran(RB) in corn-soybean meal diets fed to broiler chickens without or with a multi-enzyme supplement(MES). The MES supplied xylanase, b-glucanase invertase, protease, cellulase, a-amylase and mannanase with targeted activity of 2,500, 300, 700, 10,0001,200, 24,000, and 20 U/kg of feed, respectively. The study used a two-phase feeding program(starter d 0 to 24; finisher, d 25 to 35) with RB added at 5% and 11%, respectively creating 4 diets in each phase Diets were iso-caloric and iso-nitrogenous and contained phytase(500 FTU/kg) and TiO_2 as a digestibility marker. Three hundred and sixty d-old male Ross 708 broiler chicks were placed in cages based on BW(15 birds/cage) and allocated to 4 diets(n = 6). Birds had free access to feed and water. Body weight and feed intake were recorded. Excreta samples were collected 3 d prior to the end of each phase for apparent retention(AR) of components. Samples of birds were sacrificed on d 24 and 35 for gut weight and ceca digesta for organic acid content. There was no interaction(P > 0.10) between RB and MES on BWG and FCR in starter or finisher phase. In finisher phase, birds fed MES had better BWG(961 versus 858 g) and FCR(1.69 versus 1.86) than birds fed non-MES diets(P increased gizzard weight and reduced final BW whereas MES improved growth and energy utilization.

Development of Novel Antiviral Therapies for Hepatitis C Virus

Over 170 million people worldwide are infected with hepatitis C virus (HCV), a major cause of liver diseases. Current interferon-based therapy is of limited efficacy and has significant side effects and more effective and better tolerated therapies are urgently needed. HCV is a positive, single-stranded RNA virus with a 9.6 kb genome that encodes ten viral proteins. Among them, the NS3 protease and the NS5B polymerase are essential for viral replication and have been the main focus of drug discovery efforts. Aided by structure-based drug design, potent and specific inhibitors of NS3 and NS5B have been identified, some of which are in late stage clinical trials and may significantly improve current HCV treatment. Inhibitors of other viral targets such as NS5A are also being pursued. However, HCV is an RNA virus characterized by high replication and mutation rates and consequently, resistance emerges quickly in patients treated with specific antivirals as monotherapy. A complementary approach is to target host factors such as cyclophilins that are also essential for viral replication and may present a higher genetic barrier to resistance. Combinations of these inhibitors of different mechanism are likely to become the essential components of future HCV therapies in order to maximize antiviral efficacy and prevent the emergence of resistance.
