Some Haematological Parameters in Primary Hypertensive Subjects Attending a Tertiary Hospital in Nigeria

Background: Hypertension is a persistent elevation of blood pressure in the arteries which if not properly managed can lead to stroke, heart failure, atrial fibrillation, peripheral vascular disease and other life threatening outcomes. This study investigated some haematological parameters of Primary hypertensive subjects. Objectives: To compare some haematological parameters (haematocrit, haemoglobin concentration, Platelets count, White blood Cells count, red blood cell count and Red Cells Indices between hypertensive and normotensive subjects. Materials and methods: blood samples were collected from 76 known hypertensive subjects between 30 - 70 years attending the Cardiology clinic of the University of Abuja Teaching Hospital Gwagwalada. Another 37 normotensive subjects between 30 - 65 years served as the control. All subjects gave their consents. Platelets count, Haemoglobin estimation, Packed Cell Volume, Red blood Cell Count, Mean Cell Volume, Mean Cell Haemoglobin, Mean Cell Haemoglobin Concentration and White blood Cell count were determined using Mythic 22 five parts haematology analyzer. Results: Results for Hypertensive and Control subjects were, White blood Cell, 5.76 ± 1.45 × 109/l and 4.76 ± 1.03 × 109/l, Platelet count, 248.7552.45 × 109/l and 284.95 ± 27.66 × 109/l, Mean Cell Volume, 91.81 ± 3.05 fl and 85.68 ± 6.48 fl, Mean Cell Haemoglobin, 30.59 ± 1.04 pg and 27.922 ± 2.74 pg, Mean Cell Haemoglobin Concentration, 33.34 ± 0.61 g/dl and 32.32 ± 0.93 g/dl, Red blood Cell, 4.33 ± 0.39 × 109/l and 4.50 ± 0.52 × 109/l, Packed Cell Volume, 39% ± 3.15% and 40% ± 4.41% and Haemoglobin, 13.21 ± 1.10 g/dl and 13.50 ± 1.63 g/dl respectively. Mean Cell Volume, Mean Cell Haemoglobin, Mean Cell Haemoglobin Concentration (Red cells indices) were significantly higher in hypertensive compared to normotensive subjects (P 0.05), total White blood Cell count was also higher in hypertensive than normote

Aberrant tumor blood vessels are prone to propel the malignant progression of tumors,and targeting abnormal metabolism of tumor endothelial cells emerges as a promising option to achieve vascular normalization and antagonize tumor ***,we demonstrated that salvianic acid A(SAA)played a pivotal role in contributing to vascular normalization in the tumor-bearing mice,thereby improving delivery and effectiveness of the chemotherapeutic *** was capable of inhibiting glycolysis and strengthening endothelial junctions in the human umbilical vein endothelial cells(HUVECs)exposed to ***,SAA was inclined to directly bind to the glycolytic enzyme PKM2,leading to a dramatic decrease in endothelial *** importantly,SAA improved the endothelial integrity via activating theβ-Catenin/Claudin-5 signaling axis in a PKM2-dependent *** findings suggest that SAA may serve as a potent agent for inducing tumor vascular normalization.

Head and neck squamous cell carcinoma(HNSCC)represents the sixth most common malignancy ***,very few established diagnostic biomarkers for HNSCC have been universally applied in clinical ***,much attention has been paid to extracellular vesicles(EVs)regarding their roles as cancer biomarkers because EVs carry plentiful cargoes,including lipids,proteins,nucleic acids,and *** HNSCC,several molecules carried by EVs,which are derived from peripheral blood and saliva,have been implicated to be effective in cancer detection,staging,treatment planning,response monitoring,and prognosis *** several EV molecules have been identified to be significantly correlated with a set of clinicalpathological parameters of HNSCC,several key limitations need to be resolved before the clinical application of EVs as carriers of biomarkers in *** this review,we discuss current knowledge in the literature regarding EV-based biomarkers in HNSCC,emphasizing current limitations of their clinical applications.

Human RBCs blood group conversion from A to O using a novel α-N-acetylgalactosaminidase of high specific activity

α-N-acetylgalactosaminidase (αNAGA) can convert group A human red blood cells (RBCs) to group O. One novel αNAGA gene was cloned by PCR from Elizabethkingia meningosepticum isolated from a domestic clinical sample. Pure recombinant αNAGA was obtained by genetic engineering and protein purification with a calculated molecule of 49.6 kD. αNAGA was selective for terminal α-N-acetylgalacto-samine residue with a high specific activity. αNAGA could completely remove A antigens of 1 U (about 100 mL) group A1 or A2 RBCs in 1 h at pH 6.8 and 25℃ with a consumption of 1.5 or 0.4 mg recombinant enzyme. Enzyme-converted group A RBCs did not agglutinate after being mixed with monoclonal anti-A or sera of groups A,B,AB and O. Other blood group antigens except ABO had no change. FCM analy-sis showed that A antigens and A1 antigens disappeared while H antigens increased. It indicated that αNAGA successfully converted human blood group A RBCs to universally transfusable group O RBCs without the risk of ABO-incompatible transfusion reactions. This αNAGA was suitable for producing universal RBCs to increase clinical transfusion safety,improve the RBCs supply,and to decrease transfusion cost and support transfusion service in case of emergency.

Effects of Perinatal Complications on Coagulation Function in Pregnant Women

Objectives:To investigate the effects of various complications on the coagulation function of perinatal ***:A total of 210 pregnant women between November 2016 and June 2017 were *** coagulation function indexes,including prothrombin time(PT),activated partial thromboplastin time(APTT),fibrinogen(FIB),and thrombin time(TT),were evaluated in pregnant women with group B Streptococcus(GBS),positive nonstress test(NST),scarred uterus,prenatal fever,macrosomia,gestational hypertension,gestational diabetes,premature rupture of membranes(PROMs),threatened abortion,and ***:No significant differences were found in PT,APTT,FIB,and TT in pregnant women with GBS,positive NST,scarred uterus,macrosomia,gestational hypertension,gestational diabetes,PROMs,and ***,significantly decreased PT and APTT and significantly increased TT were found in pregnant women with prenatal fever compared to normal controls,whereas significantly increased TT was also observed in those with threatened ***:Among the complications that occur in pregnant women,prenatal fever and threatened abortion have important effects on coagulation function.

DEL RBC transfusion should be avoided in particular blood recipient in East Asia due to allosensitization and ineffectiveness

Previously, both primary and secondary anti-D alloimmunizations induced by "Asian type" DEL (RHD1227A allele) were observed in two incidents. We investigated how often these alloimmunization events occur. The transfusions of any D-negative patients were investigated in the First Affiliated Hospital of Xi'an Jiaotong University Medical College, China, during the entire 2009. The antigens of D, C, c, E, and e were routinely serotyped. The "Asian type" DEL variant was genotyped and the RHD heterozygote was determined through two published methods. The changes in anti-D levels were monitored by the indirect antiglobulin test (IAT) and flow cytometry. Thirty D-negative transfused patients were included in the study. We focused on 11 recipients who were transfused with packed red blood cells (RBCs) from DEL donors at least one time. Of those 11 recipients, seven were anti-D negative before transfusion and four were anti-D positive (one patient with an autoantibody). One of the seven pre-transfusion anti-D negative patients produced a primary-response anti-D after being transfused with 400 ml of DEL blood twice. All four pre-transfusion antibody positive patients were not observed hemoglobin (Hb) levels increased, as expected after transfusions. Two patients had an increase in anti-D from 1:8 to 1:64 by IAT, which was also shown by flow cytometry. None of the patients experienced an acute hemolytic episode. Our data indicated that the primary anti-D induced by DEL transfusion or the secondary anti-D elevated by DEL in a truly D-negative patient might not be unusual. We suggest that a truly D-negative childbearing-aged woman should avoid DEL transfusion to protect her from primary anti-D allosensitization. In addition, anti-D positive recipients should also avoid DEL red cell transfusion due to the delayed hemolytic transfusion reaction (DHTR).

BACKGROUND Bisphenol A(BPA)is present in many plastic products and food *** the other hand,fertaric acid(FA)is a hydroxycinnamic *** To investigate the effect of FA on BPA-related liver,kidney,and testis toxicity,DNA breakdown,and histopathology in male *** Thirty male albino rats were divided into five equal groups(6 rats/group):Control,paraffin oil,FA-,BPA-,and FA+BPA-treated *** control and paraffin oil groups were administered orally with 1 mL distilled water and 1 mL paraffin oil,*** FA-,BPA-,and FA+BPA-treated groups were administered orally with FA(45 mg/kg,bw)dissolved in 1 mL distilled water,BPA(4 mg/kg,bw)dissolved in 1 mL paraffin oil,and FA(45 mg/kg,bw)followed by BPA(4 mg/kg,bw),*** these treatments were given once a day for 6 *** BPA induced a significant decrease in serum alkaline phosphatase,acid phosphatase,sodium,potassium and chloride,testosterone,dehydroepiandrosterone sulfate,glucose-6-phosphate dehydrogenase,3β-hydroxysteroid dehydrogenase,and testis protein levels but a highly significant increase in serum aspartate aminotransferase,alanine aminotransferase,γ-glutamyl transpeptidase,lactate dehydrogenase,bilirubin,urea,creatinine,uric acid,luteinizing hormone,follicle stimulating hormone,sex hormone binding globulin,blood urea nitrogen,and testis cholesterol ***,FA inhibited the degradation of liver,kidney,and testis DNA *** administration of FA to BPA-treated rats restored all the above parameters to normal *** FA ameliorates BPA-induced liver,kidney,and testis toxicity,DNA breakdown,and histopathological changes.

Insights into the Impact of Bifidobacterium bifidum， B. breve， and B. longum on the Immune Response Against Salmonella Infection in Neonatal Rats

Infants exhibit diminished susceptibility to external infections， a phenomenon closely linked to the pervasive presence of the bifidobacterial community within their intestinal tract. Nonetheless， a significant knowledge gap remains concerning the distinct species of bifidobacteria initiating anti-infection immune responses during the early developmental stages. In this study， the impact of early intervention with Bifidobacterium bifidum (B. bifidum)， B. breve， and B. longum on neonatal rats infected with Salmonella enterica serovar Typhimurium (S. typhimurium) SL1344 were compared. The study encompasses various immunity levels， including gut immunity， central immunity (thymus)， peripheral immunity (spleen)， and brain immunity. Following Salmonella infection， significant alterations in neonatal rats were observed in growth and developmental levels， immune markers， cytokine levels， balance of T lymphocyte subpopulations， intestinal barrier function， and blood-brain barrier integrity. Compared with B. breve and B. longum， B. bifidum demonstrated more pronounced efficacy in regulating these physiological processes. By conducting multi-level analyses of gut microbiota， bifidobacterial community， colonic content metabolomics， and serum metabolomics， the significance of B. bifidum's role is underscored， and the immune-enhancing function of messenger metabolites is unveiled. Among these metabolites， γ-L-Glutamyl-L-glutamic acid and Orotic acid were found to be shared by all three species， while Hippuric acid and 1a，1b-dihomo Prostaglandin F2α were unique to B. bifidum， and DL-Arginine was specific to B. longum. Overall， this study has provided novel insights into the intervention and immunomodulation by Bifidobacterium in early-life infections， emphasizing the significant role of B. bifidum.

Role of exosomal long non-coding RNAs in colorectal cancer

Exosomes are a class of small extracellular vesicles,30-150 nm in diameter,that transfer biological information(e.g.,DNA,RNA,and protein)via cell-to-cell *** play critical roles in the occurrence and development of human cancers,including colorectal cancer(CRC).Recent studies have shown that long non-coding RNAs(lncRNAs)can be encapsulated in exosomes,which transfer lncRNAs from secretory cells into recipient *** process affects the progression of CRC,since exosomal lncRNAs display special and extensive functions in CRC tumorigenesis,including malignant proliferation,metastasis,chemoresistance,and inflammatory ***,due to their specificity and sensitivity,exosomal lncRNAs are released into body fluids(e.g.,urine,sputum,and plasma),which have the potential to be biomarkers of CRC tumorigenesis within screening efforts and medical and epidemiologic *** this review,we aim to clarify the function and mechanism of exosomal lncRNAs in CRC tumorigenesis and provide a strategy for early diagnosis and medical treatment of this malignancy.
