Constructing Artificial Nucleobase Compilation to Enable Precise molecular medicine

Nucleic acids are the hereditary information storage medium of life. Due to its high programmability and good biocompatibility, the applications of nucleic acids are not limited to their natural function. However, the efficiency of nucleic acids is often hampered by inherent limitations in numerous practical applications, including limited access to complex functional patterns due to only four building blocks, facile degradation by nucleases, rapid renal clearance, poor pharmacokinetic properties, and so on. To end this, the researchers developed unnatural base pairs (UBPs) and numerous artificial analogues of nucleosides and oligonucleotides in recent decades. The developed UBPs and nucleoside base analogues together constitute artificial nucleobase compilation, which promotes the development of biomedical sciences. Here, we describe the development of artificial nucleobase compilation and summarized its applications in precise molecular medicine, including PCR-based diagnostics, aptamer-based diagnostics, nucleobase analogue drugs construction, ASO modification, aptamer-based therapeutics and biomaterials construction. This review provides an overview of current opportunities and challenges of artificial nucleobase-related precise molecular medicine.

Advancing cancer cachexia diagnosis with -omics technology and exercise as molecular medicine

Muscle atrophy exacerbates disease outcomes and increases mortality,whereas the preservation of skeletal muscle mass and function play pivotal roles in ensuring long-term health and overall *** atrophy represents a significant clinical challenge,involving the continued loss of muscle mass and strength,which frequently accompany the development of numerous types of *** cachexia is a highly prevalent multifactorial syndrome,and although cachexia is one of the main causes of cancer-related deaths,there are still no approved management strategies for the *** etiology of this condition is based on the upregulation of systemic inflammation factors and catabolic stimuli,resulting in the inhibition of protein synthesis and enhancement of protein *** necessary cellular processes are disrupted by cachectic pathology,which mediate intracellular signalling pathways resulting in the net loss of muscle and ***,the exact underpinning molecular mechanisms of how these changes are orchestrated are incompletely *** work is still required,but structured exercise has the capacity to counteract numerous detrimental effects linked to cancer *** through the stimulation of muscle protein synthesis,enhancement of mitochondrial function,and the release of *** a result,muscle mass and strength increase,leading to improved mobility,and *** review summarises existing knowledge of the complex molecular networks that regulate cancer cachexia and exercise,highlighting the molecular interplay between the two for potential therapeutic ***,the utility of mass spectrometry-based proteomics is considered as a way of establishing early diagnostic biomarkers of cachectic patients.

The development of an effective classification method for human health conditions is essential for precise diagnosis and delivery of tailored therapy to individuals. Contemporary classification of disease systems has properties that limit its information content and usability. Chinese medicine pattern classification has been incorporated with disease classification, and this integrated classification method became more precise because of the increased understanding of the molecular mechanisms. However, we are still facing the complexity of diseases and patterns in the classification of health conditions. With continuing advances in omics methodologies and instrumentation, we are proposing a new classification approach： molecular module classification, which is applying molecular modules to classifying human health status. The initiative would be precisely defining the health status, providing accurate diagnoses, optimizing the therapeutics and improving new drug discovery strategy. Therefore, there would be no current disease diagnosis, no disease pattern classification, and in the future, a new medicine based on this classification, molecular module medicine, could redefine health statuses and reshape the clinical practice.

In this article, we present our previous research, which highlighted adenosine triphosphate(ATP) as the cause of neuropathic pain during the acute phase of neuromyelitis optica spectrum disorder(NMOSD). In NMOSD pathology, damageassociated molecular patterns(DAMPs), including ATP, are released from damaged astrocytes, triggering the activation of innate immune cells. ATP is a central mediator of acute pain in NMOSD.
