Selection of Trichoderma mutants with enhanced cellulase production and resistant to catabolite repression

Due to high cost and relatively low efficiency of cellulase enzymes used for the saccharification of pretreated lignocelluloses, the improvement of cellulase secreting microorganisms is of vital importance. Trichoderma reesei QM 6a, an excellent source of cellulase was selected in the late 1960’s. at Natick Laboratories by its performance on pure cellulose (Solka Floc, Avicel) . QM 6a is the wild parent strain of best existing hypercellulolytic mutants such as Rut C30, VTT-D-80133, L27, CL-847 and others. Utilization of cheaper carbon sources (e.g., pretreated wood or straw) both in enzyme production and in hydrolysis necessitates to investigate fungal species other than ***. A screening program was initiated to test 150 wild-type Trichoderma strains in shake flask for cellulase production on SO 2-impregnated and steam pretreated spruce and willow, candidate substrates for bioalcohol program in Sweden. Filter paper activity (FPA) method was used to determine the overall cellulase activity. Strain TUB F-1505 was selected as promising candidate for mutagenesis. This wild strain was isolated from a tropical rain forest area near Manaus, Brazil. Isolate F-1505 was subjected to NTG-mutation to select catabolite (glucose, glycerol) resistant mutants. A Petri plate clearing assay using Walseth cellulose, glycerol or glucose and Triton X100 (colony size inhibitor) was applied for pre-screening of the colonies. Over 6000 colonies were evaluated. Best colonies were tested in shake flask fermentation on pretreated spruce and willow as carbon sources. Mutants producing higher levels of cellulase (FPA) were further mutated by either NTG or UV-light. At least 4 mutants were obtained and freeze-dried exhibiting equivalent or higher cellulase production as compared to Trichoderma reesei Rut C30.

采用EDTA-柠檬酸联合络合法合成了钙钛矿型金属氧化物Sr Co0.8Fe0.2O3-δ(SCF182)粉体。通过XRD和ESEM研究了前驱体溶液不同的p H值对合成的SCF182粉体的晶体结构和微观形貌的影响。并采用固定床实验研究前驱体溶液p H值对SCF182的氧吸附/脱附性能的影响。结果表明,p H值对SCF182晶胞结构和参数影响不大,但影响SCF182的晶粒粒径和微观形貌,当p H值为8的弱碱性条件下时,能够合成蓬松多孔网状纳米结构的钙钛矿粉体。固定床实验显示,前驱体溶液的p H值影响合成的粉体SCF182的氧吸附速率;且SCF182的氧脱附性能随p H值的增大先升高后降低,p H值为8时氧脱附量达到峰值为42.2 mg/g。

PURPOSE: Preoperative, high- dose radiotherapy for rectal cancer reduces local recurrence rates and improves overall survival. However, adverse effects in varying degrees include impaired wound healing and local infection. This study investigates the influence of preoperative, high- dose radiotherapy on subcutaneous accumulation of collagen in a primary rectal cancer group operated on with or without adjuvant radiotherapy. METHODS: Forty- two eligible patients who underwent total mesorectal excision surgery with or without radiotherapy were included in the study. Polytetrafluoroethylene tubings were implanted in the arm ten days before surgery (three days before the start of radiotherapy). Implants were extracted the day before surgery. New implants were inserted before surgery and were extracted ten days after surgery. The hydroxyproline and proline contents of the implants were measured and the hydroxyproline/proline ratio was calculated as a measure for deposited collagen relative to protein. Blood loss, postoperative complications, and blood levels of hemoglob- in, leukocytes, and albumin were recorded. RESULTS: The two groups were similar in relation to Dukes stage, age, and body mass index. Infectious complications developed in 39 percent of patients after radiotherapy compared with 16 percent in the nonirradiated group. In the irradiated patients with infective complications we found a significant decrease in the hydroxyproline/proline ratio compared with that of irradiated patients without infections (P = 0.037). There was a significant decrease in the leukocyte count preoperatively and postoperatively in the irradiated group compared with surgery alone. CONCLUSIONS: Highdose, short- term radiotherapy does not have a systemic effect on collagen accumulation, but a significant reduction is manifested in infected patients. Radiotherapy also impairs leukocyte production and increases the postoperative infective complication rate.

The Dongsithouane National production Forest (DNPF) is one of the largest natural forest areas in Savannakhet, Lao PDR, which has been a vital support for the local community’s livelihood, Recently, significant changes in land use and land cover (LULC) have been observed in this area, leading to a reduction of natural forests. There were two separate methods of this study: firstly, to identify LULC changes across three different periods, spectral imagery from the Landsat 5 Thematic Mapper (TM) for the years 2001 and 2011, and the Landsat 8 Operational Land Imager (OLI) for 2021 were used as the primary data sources. The satellite images were preprocessed for various forest classes, including pretreatment of the top of atmosphere reflectance by using QGIS software’s semi-automatic classification plug-in (SCP), and ArcGIS was used for post-classification. A supervised classification approach was applied to the satellite images from 2001, 2011, and 2021 to generate diverse maps of LULC. Secondly, a household survey dataset was used to investigate influential factors. Approximately 220 households were interviewed in order to collect socio-economic information (including data on population growth, increased business activities, location of the area, agriculture land expansion, and need for settlement land). Household survey data was analyzed by using SPSS. Descriptive statistics, including frequency distributions and percentages, were applied to observe characteristics. Additionally, a binary logistic regression model was used to analyze the socioeconomic factors related to LULC change in DNPF. Key findings indicated a decline in natural forest areas within the study site. Specifically, both dry dipterocarp forest (−11.35%) and mixed deciduous forest (−0.18%) decreased from 2001 to 2021. Interannual variability of the terrestrial ecosystem carbon sink is substantially regulated by various environmental variables and highly dominates the interannual variation of atmospheric carbon dioxide(CO2) ***,it is necessary to determine dominating factors affecting the interannual variability of the carbon sink to improve our capability of predicting future terrestrial carbon *** global datasets derived from machine-learning methods and process-based ecosystem models,this study reveals that the inter annual variability of the atmospheric vapor pressure deficit(VPD) was significantly negatively correlated with net ecosystem production(NEP) and substantially impacted the inter annual variability of the atmospheric CO2growth rate(CGR).Further analyses found widespread constraints of VPD interannual variability on terrestrial gross primary production(GPP),causing VPD to impact NEP and *** correlation analysis confirms the persistent and widespread impacts of VPD on terrestrial carbon sinks compared to other environmental *** Earth system models underestimate the interannual variability in VPD and its impacts on GPP and *** results highlight the importance of VPD for terrestrial carbon sinks in assessing ecosystems’ responses to future climate conditions.

Ethylbutylthiourea (EBTU) is an accelerator used in the production of chloropr ene (neoprene) rubber. EBTU occurs in a mixture with diethylthiourea (DETU) and dibutylthiourea (DBTU) in the accelerator. An analytical method originally devel oped for analysis of zinc dithiocarbamates in rubber has been used to analyse EB TU, DETU and DBTU in a knee brace responsible for an allergic contact dermatitis in a gardener suffering from arthrosis. EBTU was isolated and gave positive rea ctions when tested as a pure compound. The test reaction was accompanied by posi tive reactions to DETU and DBTU.

Using a postal questionnaire the prevalence of hand eczemawas determined in a general population of 11 798 individuals aged 20- 77 years who were randomly drawn from the population records. The response rate was 78.1% . One- year prevalence of hand eczema among women varied between 1.9% and 10.8% , with the highest figure among those aged 30- 39 years. The corresponding figures for men were 2.3% and 5.6% , with the highest figure among those aged 20- 29 years. Lifetime prevalence varied between 5.7% and 16.7% among women and between 5.2% and 9.5% among men. Using multiple logistic regression analysis female sex (OR=1.91, 95% CI 1.47- 2.47)- and smoking (OR=1.35, 95% CI 1.04- 1.75) were independent risk factors for reporting 1- year prevalence of hand eczema, whereas age (OR=0.99, 95% CI 0.97- 0.99) was inversely related to the 1- year prevalence of hand eczema. Aggregated risk occupation or categorized occupation such as medical and nursing work, production or service were not significantly associated with 1- year prevalence of hand eczema.

Objective:To provide the most up-to-date data on the burden of malignant mesothelioma(MM)and the projections through 2029 in ***:Data on patients diagnosed with MM from China during 1990-2019 were obtained from the Global Burden of Disease(GBD)2019 database,including annual cases and deaths data and age-standardized rates of incidence,mortality,and disability-adjusted life-years(DALYs)associated with MM among different age *** trends during 1990-2019 were analyzed by the Joinpoint regression models using 95%confidence interval(CI),while the projections through 2029 were calculated by the Bayesian age-period-cohort *** on the production and consumption of asbestos in China were obtained from the United States Geological Survey on Mineral Commodity Summaries during ***:We observed a significant elevation in incident new cases and deaths over the last 3 decades,increasing from 1193 in 1990 to 2815 in 2019 for incident cases and from 1134 in 1990 to 2773 in 2019 for death *** found a roughly 6%increase in the proportion of incident cases for those aged>70 years(30%in 2019 versus 24%in 1990),while for the proportion of deaths similar elevation for those aged>70 years was ***,men had significantly higher DALYs due to MM across age groups compared with *** consumption in China dramatically dropped since 2012 and reached the bottom in 2017 with 230 *** 2029,the projected age-standardized rate for incidence and mortality is expected to reach 1.2 per million for ***:We found,for the first time using GBD data on the Chinese population,that the burden of MM has been significantly increasing in China over the last three decades and will continue to increase in the upcoming decade,suggesting an urgent need for a complete ban on chrysotile asbestos in China.

Dear Editor,In the European region,which shares 22.8%of the global cancer burden for 10%of the global population,there were around 4.4 million new cancer cases and 1.9 million deaths from cancer in 2020[1].The reasons for the high cancer incidence rates are complex;however,diet and dietary components are among the main contributors to cancer risk[2].In modern-day living,a growing proportion of people include in their diets ultra-processed *** of food processing and home-prepared foods are so-called dietary advanced glycation endproducts(AGEs),which are reactive metabolites emerging during the breakdown of reducing *** production is preponderant in dry high-heat processes(e.g.,baking,roasting);hence foods such as cakes,crisps,crackers,cereal products,meat and meat-derived products represent a major source of dietary AGEs[3].
