Neutralizing antibodies in hepatitis C virus infection

Hepatitis C virus (HCV) is a major cause of hepatitis world-wide. The majority of infected individuals develop chronic hepatitis which can then progress to liver cirrhosis and hepatocellular carcinoma. Spontaneous viral clearance occurs in about 20%-30% of acutely infected individuals and results in resolution of infection without sequaelae. Both viral and host factors appear to play an important role for resolution of acute infection. A large body of evidence suggests that a strong, multispecific and long-lasting cellular immune response appears to be important for control of viral infection in acute hepatitis C. Due too the lack of convenient neutralization assays, the impact of neutralizing responses for control of viral infection had been less defined. In recent years, the development of robust tissue culture model systems for HCV entry and infection has finally allowed study of antibody-mediated neutralization and to gain further insights into viral targets of host neutralizing responses. In addition, detailed analysis of antibody-mediated neutralization in individual patients as well as cohorts with well defined viral isolates has enabled the study of neutralizing responses in the course of HCV infection and characterization of the impact of neutralizing antibodiesfor control of viral infection. This review will summarize recent progress in the understanding of the molecular mechanisms of antibody-mediated neutralization and its impact for HCV pathogenesis.

This paper deals with the synthesis of a new lateral macrobicyclic cryptand ***-tion of 2,6-di（bromomethyl）pyridine with 3-（n-tosylamino）methyl propionate in the presence ofanhydrous KCOat room temperature gave 5 which was hydrolyzed to *** of the latter withexcess sulfonyl chloride afforded 2,6-di（4’-chloroformyl-2’-tosyl-2’-azabutyl）pyridine（7）.Pentaaza-1,5,13,17,28-dithia-20,25-tricyclo[15,5,5,1]-octacosa-7,9,11（28）-triene（10）has been synthesized bythe following sequence of reactions:Condensation of 7 with 1,7-diaza-4,10-dithia-dodecane（3）by highdilution technique in toluene-THF at room femperature afforded 8 which was reduced by diboranein THF solution to give *** of 9 with sodium anthracene in monoglyme produced 10.

We consider first order quasilinear hyperbolic systems with vertical characteristics. It was shown in [4] that such systems can be exactly controllable with the help of internal controls applied to the equations corresponding to zero eigenvalues. However, it is possible that, for physical or engineering reasons, we can not put any control on the equations corresponding to zero eigenvalues. In this paper, we will establish the exact controllability only by means of physically meaningfnl internal controls applied to the equations corresponding to non-zero eigenvalues. We also show the exact controllability for a very simplified model by means of switching controls.

Anti-hepatitis B virus(HBV)therapy leads to the emer- gence of mutant viral strains during the treatment of chronic hepatitis B with nucleos(t)ides analogues. The existence of HBV variants with primary antiviral resistance may be important for treatment choice. We studied two patients with chronic HBV infection by sequencing the HBV polymerase *** had adefovir-and tenofovir-related mutations in the viral polymerase,although they had never been treated. These mutations were rtV214A/rtN238T in one patient and rtA194T in the ***,mutations in untreated patients deserve cautious *** data indicate that mutations that can theoretically confer adefovir or tenofovir resistance may emerge in treatmentnaive patients.

Cuprous chloride hydrochloric acid solutions were electrolysed in a two compartments cell without agitation for copper extraction. It is found that the current density affects the colour and the size of copper deposits. During electrodeposition of copper from cuprous solution in the presence of various concentrations of lead, zinc or iron ions at different current densities, it is observed that lead is codeposited with copper by increasing current *** all experiments, the current efficiency for the copper deposition reaction fluctuates between 88.50% and 95.50%.

SDG714 Regulates Specific Gene Expression and Consequently Affects Plant Growth via H3K9 Dimethylation

Histone lysine methylation is known to be involved in the epigenetic regulation of gene expression in all eukaryotes including plants. Here we show that the rice SDG714 is primarily responsible for dimethylation but not trimethylaUon on histone H3K9 in vivo. Overexpression of YFP-SDG714 in Arabidopsis significantly inhibits plant growth and this inhibition is associated with an enhanced level of H3K9 dimethylation. Our microarray results show that many genes essential for the plant growth and development were downregulated in transgenic Arabidopsis plants overexpressing YFP-SDG714. By chromatin immunoprecipitation analysis, we show that YFP-SDG714 is targeted to specific chromatin regions and dimethylate the H3Kg, which is linked with heterochromatinization and the downregulation of genes. Most interestingly, when YFP-SDG714 production is stopped, the inhibited plants can partially restore their growth, suggesting that the perturbation of gene expression caused by YFP-SDG714 is revertible. Taken together, our results point to an important role of SDG714 in H3K9 dimethylation, suppression of gene expression and plant growth, and provide a potential method to regulate gene expression and plant development by an on-off switch of SDG714 expression.
