Hepatic steatosis is associated with dysregulated cholesterol metabolism and altered protein acetylation dynamics in chickens

Background Hepatic steatosis is a prevalent manifestation of fatty liver, that has detrimental effect on the health and productivity of laying hens, resulting in economic losses to the poultry industry. Here, we aimed to systematically investigate the genetic regulatory mechanisms of hepatic steatosis in laying *** Ninety individuals with the most prominent characteristics were selected from 686 laying hens according to the accumulation of lipid droplets in the liver, and were graded into three groups, including the control, mild hepatic steatosis and severe hepatic steatosis groups. A combination of transcriptome, proteome, acetylome and lipidome analyses, along with bioinformatics analysis were used to screen the key biological processes, modifications and lipids associated with hepatic *** The rationality of the hepatic steatosis grouping was verified through liver biochemical assays and RNA-seq. Hepatic steatosis was characterized by increased lipid deposition and multiple metabolic abnormalities. Integration of proteome and acetylome revealed that differentially expressed proteins(DEPs) interacted with differentially acetylated proteins(DAPs) and were involved in maintaining the metabolic balance in the liver. Acetylation alterations mainly occurred in the progression from mild to severe hepatic steatosis, i.e., the enzymes in the fatty acid oxidation and bile acid synthesis pathways were significantly less acetylated in severe hepatic steatosis group than that in mild group(P < 0.05). Lipidomics detected a variety of sphingolipids(SPs) and glycerophospholipids(GPs) were negatively correlated with hepatic steatosis(r ≤-0.5, P < 0.05). Dear Editor,This letter proposes a fuzzy indirect iterative learning(FIIL)active disturbance rejection control(ADRC)scheme to address the impact of uncertain factors of plant-protection unmanned ground vehicle(UGV),in which ADRC is a data-driven model-free control algorithm that only relies on the input and output data of the *** on the established nonlinear time-varying dynamic model including dynamic load(medicine box),the FIIL technology is adopted to turn the bandwidth and control channel gain online,in which the fuzzy logic system is used to update the gain parameters of iterative learning in real *** and experiment show the FIIL-ADRC scheme has better control performance.

Removal of pathogenic indicator microorganisms during partial nitrification:the role of free nitrous acid

Digested wastewater contains pathogenic microorganisms and high ammonia concentrations,which can pose a potential risk to public *** removal of pathogens and nitrogen is crucial for the post-treatment of digested *** nitrification-anammox is an energy-saving nitrogen removal *** nitrous acid(FNA),an intermediate product of partial nitrification,has the potential to inactivate ***,the efficiency and mechanisms of FNA-related inactivation in pathogens during partial nitrification remains *** this study,Enterococcus and Escherichia coli(***)were selected to investigate the efficiency and mechanisms of FNA-related inactivation in partial nitrification *** results revealed that 83%±13%and 59%±27%of *** and Enterococcus were removed,respectively,in partial nitrification process at FNA concentrations of 0.023−0.028 mg/*** the concentration of FNA increased from 0 to 0.5 mg/L,the inactivation efficiencies of *** and Enterococcus increased from 0 to 99.9%and 89.9%,*** exhibited a higher resistance to FNA attack compared to ***.3D-laser scanning microscopy(3D-LSM)and scanning electron microscopy(SEM)revealed that FNA exposure caused the surface collapse of *** and Enterococcus,as well as visible pore formation on the surface of *** cells.4',6-Diamidino-2-phenylindole dihydrochloride n-hydrate(DAPI)/propidium iodide(PI)and biomolecule leakage confirmed that inactivation of *** and Enterococcus occurred due to breakdown of cell walls and cell *** findings indicate that partial nitrification process can be used for the removal of residual pathogenic microorganisms.

Efficiency-first spraying mission arrangement optimization with multiple UAVs in heterogeneous farmland with varying pesticide requirements

Combining multiple crop protection Unmanned Aerial Vehicles(UAVs)as a team for a scheduled spraying mission over farmland now is a common way to significantly increase ***,given some issues such as different configurations,irregular borders,and especially varying pesticide requirements,it is more important and more complex than other multi-Agent Systems(MASs)in common *** this work,we focus on the mission arrangement of UAVs,which is the foundation of other high-level cooperations,systematically propose Efficiency-first Spraying Mission Arrangement Problem(ESMAP),and try to construct a united problem framework for the mission arrangement of crop protection ***,to characterise the differences in sub-areas,the varying pesticide requirement per unit is well considered based on Normalized Difference Vegetation Index(NDVI).Firstly,the mathematical model of multiple crop-protection UAVs is established and ESMAP is ***,an acquisition method of a farmland’s NDVI map is proposed,and the calculation method of pesticide volume based on NDVI is ***,an improved Genetic Algorithm(GA)is proposed to solve ESMAP,and a comparable combination algorithm is *** simulations for algorithm analysis are carried out within MATLAB,and it is determined that the proposed GA is more efficient and accurate than the ***,a mission arrangement tested with three UAVs was carried out to validate the effectiveness of the proposed GA in spraying *** results illustrated that it performed well,which took only 90.6%of the operation time taken by the combination algorithm.

Estimating wheat spike-leaf composite indicator(SLI)dynamics by coupling spectral indices and machine learning

The contribution of spike photosynthesis to grain yield(GY)has been overlooked in the accurate spectral prediction of ***,it’s essential to construct and estimate a yield-related phenotypic trait considering spike *** on field and spectral reflectance data from 19 wheat cultivars under two nitrogen fertilization conditions in two years,our objectives were to(i)construct a yield-related phenotypic trait(spike–leaf composite indicator,SLI)accounting for the contribution of the spike to photosynthesis,(ii)develop a novel spectral index(enhanced triangle vegetation index,ETVI3)sensitive to SLI,and(iii)establish and evaluate SLI estimation models by integrating spectral indices and machine learning *** results showed that SLI was sensitive to nitrogen fertilizer and wheat cultivar variation as well as a better predictor of yield than the leaf area ***3 maintained a strong correlation with SLI throughout the growth stage,whereas the correlations of other spectral indices with SLI were poor after spike *** spectral indices and machine learning algorithms improved the estimation accuracy of SLI,with the most accurate estimates of SLI showing coefficient of determination,root mean square error(RMSE),and relative RMSE values of 0.71,0.047,and 26.93%,*** results provide new insights into the role of fruiting organs for the accurate spectral prediction of *** high-throughput SLI estimation approach can be applied for wheat yield prediction at whole growth stages and may be assisted with agronomical practices and variety selection.

Assessing the impacts of traffic calming at network level:A multimodal agent-based simulation

The reduction of speed limits in urban roads through traffic calming schemes intends to ensure safer traffic conditions among road users by reducing the probability related to the occurrence of severe *** it from a different perspective,traffic calming measures can potentially resolve congestion problems at the same time by lowering the overall accessibility and attractiveness of private cars in urban *** study proposes a new methodological approach to explore and assess the direct impacts of traffic calming in the transport system efficiency of a metropolitan *** multi-agent transport simulation(MATSim)and Open-Berlin scenario are utilized to perform this simulation *** developing a new external tool,the free flow speed and road capacity of each network link is updated based on new speed limits and different compliance rates,which are defined per road hierarchy *** test scenarios that are formulated present radical conditions,where the speed limit in most urban roads of Berlin drops to 30 km/h or even 15 km/*** findings of this study show a considerably high increase in trips,passenger hours,and passenger kilometers using public transport modes,when traffic calming links are introduced,the reserve change is observed in private cars *** the speed limits are decreased in inner urban roads in most of the scenarios,the decrease of average travel speed of private cars is not so high as it was ***,private cars are used for longer distances in all test *** drivers seem to use already existed motorways and private road to *** simulations,driver compliance to the new speed limits seems to be a determinant factor that is strongly influenced by the design interventions applied in a traffic calming area.

Modifications in aroma characteristics of'Merlot'dry red wines aged in American,French and Slovakian oak barrels with different toasting degrees

Aging in oak barrels is widely used in enology which could bring flavor changes and aromatic complexity to *** the present study,the aroma compounds were analyzed from the‘Merlot’dry red wines,which were fermented in two types of fermenters(stainless steel tank and rotated oak barrel)and aged in six types of oak barrels(three geographic origins×two toasting degrees)for different time(0,3,6 and 9 months,respectively).Results showed that 30 volatiles were associated with barrels and increased during oak *** fermenters could influence the intensities of the toast,leathery,smoky,fruity,floral and caramel *** concentration of whisky lactone,eugenol,cis-isoeugenol,and the intensities of the toast and spicy aromas were highest in the wines aged in American oak and were lowest in the wines aged in French oak *** concentrations of guaiacol,syringol,trans-isoeugenol,furfural alcohol,vanilla,cis-whisky lactone enabled the medium toasting barrels to be distinguished from the light toasting *** compounds originating from the barrels could be used to distinguish the types of different barrels,but the other general grape-derived and fermentation-derived volatiles could *** fermenters,oak species and toasting degrees of the barrels all had significant effects on the aroma profiles of the aged‘Merlot’dry red wines,but the influence of the geographic origin was not obvious.
