A systematic characterization of the similarities and differences among different methods for detecting structural brain abnormalities in schizophrenia,such as voxel-based morphometry(VBM),tensor-based morphometry(TBM),and projection-based thickness(PBT),is important for understanding the brain pathology in schizophrenia and for developing effective biomarkers for a diagnosis of ***,such studies are still ***,we performed VBM,TBM,and PBT analyses on T1-weighted brain MR images acquired from 116 patients with schizophrenia and 116 healthy *** found that,although all methods detected wide-spread structural changes,different methods captured different information-only 10.35%of the grey matter changes in cortex were detected by all three methods,and VBM only detected 11.36%of the white matter changes detected by ***,pattern classification between patients and controls revealed that combining different measures improved the classification accuracy(81.9%),indicating that fusion of different structural measures serves as a better neuroimaging marker for the objective diagnosis of schizophrenia.

Identifying genetic risk factors for Alzheimer's disease(AD)is an important research *** date,different endophenotypes,such as imaging-derived endophenotypes and proteomic expression-derived endophenotypes,have shown the great value in uncovering risk genes compared to case-control ***,a co-varying pattern of different omics-derived endophenotypes could result from the shared genetic ***,existing methods mainly focus on the effect of endophenotypes alone;the effect of cross-endophenotype(CEP)associations remains largely *** this study,we used both endophenotypes and their CEP associations of multi-omic data to identify genetic risk factors,and proposed two integrated multi-task sparse canonical correlation analysis(inMTSCCA)methods,i.e.,pairwise endophenotype correlationguided MTSCCA(pcMTSCCA)and high-order endophenotype correlation-guided MTSCCA(hocMTSCCA).pcMTSCCA employed pairwise correlations between magnetic resonance imaging(MRI)-derived,plasma-derived,and cerebrospinal fluid(CSF)-derived endophenotypes as an additional *** used high-order correlations among these multi-omic data for *** figure out genetic risk factors at individual and group levels,as well as altered endophenotypic markers,we introduced sparsity-inducing penalties for both *** compared pcMTSCCA and hocMTSCCA with three related methods on both simulation and real(consisting of neuroimaging data,proteomic analytes,and genetic data)*** results showed that our methods obtained better or comparable canonical correlation coefficients(CCCs)and better feature subsets than *** importantly,the identified genetic loci and heterogeneous endophenotypic markers showed high ***,jointly using multi-omic endophenotypes and their CEP associations is promising to reveal genetic risk factors.

Central insulin resistance, the diminished cellular sensitivity to insulin in the brain, has been implicated in diabetes mellitus, Alzheimer’s disease and other neurological disorders. However, whether and how central insulin resistance plays a role in the eye remains unclear. Here, we performed intracerebroventricular injection of S961, a potent and specific blocker of insulin receptor in adult Wistar rats to test if central insulin resistance leads to pathological changes in ocular structures. 80 mg of S961 was stereotaxically injected into the lateral ventricle of the experimental group twice at 7 days apart, whereas buffer solution was injected to the sham control group. Blood samples, intraocular pressure, trabecular meshwork morphology, ciliary body markers, retinal and optic nerve integrity, and whole genome expression patterns were then evaluated. While neither blood glucose nor serum insulin level was significantly altered in the experimental or control group, we found that injection of S961 but not buffer solution significantly increased intraocular pressure at 14 and 24 days after first injection, along with reduced porosity and aquaporin 4 expression in the trabecular meshwork, and increased tumor necrosis factor α and aquaporin 4 expression in the ciliary body. In the retina, cell density and insulin receptor expression decreased in the retinal ganglion cell layer upon S961 injection. Fundus photography revealed peripapillary atrophy with vascular dysregulation in the experimental group. These retinal changes were accompanied by upregulation of pro-inflammatory and pro-apoptotic genes, downregulation of anti-inflammatory, anti-apoptotic, and neurotrophic genes, as well as dysregulation of genes involved in insulin signaling. Optic nerve histology indicated microglial activation and changes in the expression of glial fibrillary acidic protein, tumor necrosis factor α, and aquaporin 4. Analysis of fMRI Single Subject Data in the Fourier Domain Acquired Using a Multiple Input Stimulus Experimental Design

Analysis of functional MRI (fMRI) blood oxygenation level dependent (BOLD) data is typically carried out in the time domain where the data has a high temporal correlation. These analyses usually employ parametric models of the hemodynamic response function (HRF) where either pre-whitening of the data is attempted or autoregressive (AR) models are employed to model the noise. Statistical analysis then proceeds via regression of the convolution of the HRF with the input stimuli. This approach has limitations when considering that the time series collected are embedded in a brain image in which the AR model order may vary and pre-whitening techniques may be insufficient for handling faster sampling times. However fMRI data can be analyzed in the Fourier domain where the assumptions made as to the structure of the noise can be less restrictive and hypothesis tests are straightforward for single subject analysis, especially useful in a clinical setting. This allows for experiments that can have both fast temporal sampling and event-related designs where stimuli can be closely spaced in time. Equally important, statistical analysis in the Fourier domain focuses on hypothesis tests based on nonparametric estimates of the hemodynamic transfer function (HRF in the frequency domain). This is especially important for experimental designs involving multiple states (drug or stimulus induced) that may alter the form of the response function. In this context a univariate general linear model in the Fourier domain has been applied to analyze BOLD data sampled at a rate of 400 ms from an experiment that used a two-way ANOVA design for the deterministic stimulus inputs with inter-stimulus time intervals chosen from Poisson distributions of equal intensity.

Background and purpose While extracranial carotid artery stenosis is more common among Caucasians and intracranial artery stenosis is more common among Asians,the differences in atherosclerotic plaque characteristics have not yet been extensively *** sought to investigate plaque location and characteristics within extracranial carotid and intracranial arteries in symptomatic Caucasians and Chinese using vessel wall *** Subjects with recent anterior circulation ischaemic stroke were recruited and imaged at two sites in the USA and China using similar *** extracranial carotid and intracranial arteries were reviewed to determine plaque location and *** The prevalence of extracranial carotid plaque in Caucasians and Chinese was 73.1%and 49.1%,respectively(p=0.055).Prevalence of intracranial plaque was 38.5%and 69.1%in Caucasians and Chinese,respectively(p=0.02).Furthermore,42% of Caucasians and 16%of Chinese had high-risk plaque(HRP)features(intraplaque haemorrhage,luminal surface disruption)in the extracranial carotid artery(p=0.03).The prevalence of HRP features in intracranial arteries was not significantly different between the two cohorts(4%vs 11%;p=0.42).Conclusions Differences in the location and characteristics of cerebrovascular atherosclerosis were identified by vessel wall MRI in US Caucasian and Chinese subjects with recent anterior circulation ischaemic *** carotid plaques with HRP features were more common in *** plaques were more common in Chinese subjects,but no significant difference between the two cohorts in intracranial HRP prevalence was *** studies using vessel wall imaging to investigate racial differences in cerebrovascular disease may inform underlying mechanisms of HRP development and may ultimately help guide appropriate therapy.
