BACKGROUND The high prevalence and persistence of Helicobacter pylori(H. pylori) infection, as well as the diversity of pathologies related to it, suggest that the virulence factors used by this microorganism are varied. Moreover, as its proteome contains 340hypothetical proteins, it is important to investigate them to completely understand the mechanisms of its virulence and survival. We have previously reported that the hypothetical protein HP0953 is overexpressed during the first hours of adhesion to inert surfaces, under stress conditions, suggesting its role in the environmental survival of this bacterium and perhaps as a virulence factor.AIM To investigate the expression and localization of HP0953 during adhesion to an inert surface and against gastric(AGS) cells.METHODS Expression analysis was performed for HP0953 during H. pylori adhesion. HP0953 expression at 0,3, 12, 24, and 48 h was evaluated and compared using the Kruskal-Wallis equality-of-populations rank test. Recombinant protein was produced and used to obtain polyclonal antibodies for immunolocalization. Immunogold technique was performed on bacterial sections during adherence to inert surfaces and AGS cells, which was analyzed by transmission electron microscopy. HP0953 protein sequence was analyzed to predict the presence of a signal peptide and transmembrane helices, both provided by the ExPASy platform, and using the GLYCOPP platform for glycosylation sites. Different programs, via, I-TASSER, RaptorX, and HHalign-Kbest, were used to perform three-dimensional modeling.RESULTS HP0953 exhibited its maximum expression at 12 h of infection in gastric epithelium cells.Immunogold technique revealed HP0953 localization in the cytoplasm and accumulation in some peripheral areas of the bacterial body, with greater expression when it is close to AGS cells.Bioinformatics analysis revealed the presence of a signal peptide that interacts with the transmembrane region and then allows the release of the protein to the external environment. Experimental approaches to select tree species for forest restoration:effects of light,water availability and interspecific competition in degraded areas

Knowing what native trees can recr uit on degraded areas allows selecting the best species to restore these sites.However,as this information is not often available,experimentation is required before large-scale planting.This study used ex situ experiments to make these decisions on recruitment.Competition with r-strategist plants,excessive solar radiation and water shortage commonly impair tree recruitment in open habitats.The experiments focused on the interactions among these factors and were conducted with three pioneer species from seasonally dry forests of northwest Argentina,Anadenanthera colubrina,Ceiba chodatii and Jacaranda mimosifolia.Seeds of each species were sown at two light levels(sunlight/shade),two rainfall levels(full/reduced)and two levels of interspecific competition(with/without competitor)in a tree nursery.Seedling emergence and survival were monitored over a year and the results indicate that species differentially respond to varying levels of light and water.Seedlings of A.colubrina tolerated water shortages under elevated solar radiation,which are desirable features for forest restoration.Seedlings of C.chodatii tolerated shade and drought,suggesting that they require shading for establishing in open areas.However,J.mimosifolia seedlings neither tolerated full sunlight nor water shortages,suggesting that this species requires shading and regular watering if used in reforestation.Regardless of the effects of light and water,the survival of all species was reduced by interspecific competition.These results highlight the importance of experimentation for selecting the best species for forest restoration and can enhance the cost/benefit ratios of these actions.

despite the widely accepted view that planted forests are valuable habitat for wildlife species,there is a lack of empirical evidence showing the suitability of this novel habitat,especially in relation to plantations of exotic tree species.Furthermore,little is known about the effects of forest management on the ecology and dynamics of wild populations in the Neotropics.During the breeding season,the migrant flycatcher White-crested Elaenia is the most abundant bird species in forested landscapes in Chile.For several years we have studied different aspects of these artificial forests in the coastal range of south-central Chile as habitat for the species,particularly in contrast to the native forest.In general,our results indicate that plantation forests offer a poorer quality habitat,where the density of the species tends to be lower than in the native forest,although a significant edge effect suggests that its quality can be mitigated by planting as a matrix mosaic with native forest.Furthermore,compared to native forests,populations in plantations contained a higher proportion of younger adult individuals maintaining larger territories,and showing poorer body condition.We suggest that these differences could also have an impact on the birds'longevity and survival.

Magnitude of the contralateral efferent olivocochlear effect as a function of the frequency

Background:The activation of the medial olivocochlear reflex reduces the cochlear gain,which is manifested perceptually as decreased auditory sensitivity.However,it has remained unclear whether the extent of this suppression varies according to the cochlear region involved.Here we aims to assess the magnitude of contralateral efferent suppression across human cochlea,at low levels,and its impact on hearing sensitivity.Methods:Assuming that acoustic stimulation activates the contralateral medial olivocochlear reflex,we evaluated the magnitude of the suppressive effect as a function of frequency in 17 subjects with normal hearing.Absolute thresholds were measured for bursts tones of various durations(10,100,and 500 ms)and frequencies(250,500,1000,4000,and 8000 Hz)in the presence or absence of contralateral white noise at 60 d B SPL.Results:We found that contralateral noise raised the absolute threshold for the burst tones evaluated.The effect was greater at lower than higher frequencies(3.85 d B at 250 Hz vs.2.22 d B at 8000 Hz).Conclusions:Our findings suggest that in humans,the magnitude of this suppression varies according to the cochlear region stimulated,with a greater effect towards the apex(lower frequencies)than the base(higher frequencies)of the cochlea.

Estrogen rapid effects:a window of opportunity for the aging brain?

Estrogen produces several beneficial effects in healthy neurological tissues and exhibits cardioprotective effects.Hormone therapy has been widely used to treat menopausal estrogen deficiency for more than 80 years.despite high initial expectations of cardioprotective effects,there has been substantial distrust following important randomized clinical trials,such as the Women’s Health Initiative.Subsequently,the timing of treatment in relation to the onset of menopause came under consideration and led to the proposal of the timing hypothesis,that early initial treatment is important,and benefits are lost as the timing since menopause becomes prolonged.Subsequent analyses of the Women’s Health Initiative data,together with more recent data from randomized and observational trials,consistently show reductions in coronary heart disease and mortality in younger menopausal women.Regarding cognitive function,the timing hypothesis is consistent with observations from basic and animal studies.There is some clinical evidence to support the benefits of hormonal therapy in this context,though skepticism remains due to the paucity of clinical trials of substantial length in younger menopausal women.It is likely that the effects of estrogens on cognitive performance are due to rapid mechanisms,including mechanisms that influence Ca2+homeostasis dynamics,provide protection in a hostile environment and reduce inflammatory signals from neural tissues.In the future,inflammatory profiles accounting for early signs of pathological inflammation might help identify the‘window of opportunity’to use estrogen therapy for successful cognitive protection.

The blood-brain barrier is the interface through which the brain interacts with the milieu and consists mainly of a sophisticated network of brain endothelial cells that forms blood vessels and selectively moves molecules inside and outside the brain through multiple mechanisms of transport.Although brain endothelial cell function is crucial for brain homeostasis,their role in neurodegenerative diseases has historically not been considered with the same importance as other brain cells such as microglia,astroglia,neurons,or even molecules such as amyloid beta,Tau,or alpha-synuclein.Alzheimer's disease is the most common neurodegenerative disease,and brain endothelial cell dysfunction has been reported by several groups.However,its impairment has barely been considered as a potential therapeutic target.Here we review the most recent advances in the relationship between Alzheimer's disease and brain endothelial cells commitment and analyze the possible mechanisms through which their alterations contribute to this neurodegenerative disease,highlighting their inflammatory phenotype and the possibility of an impaired secretory pattern of brain endothelial cells that could contribute to the progression of this ailment.Finally,we discuss why shall brain endothelial cells be appreciated as a therapeutic target instead of solely an obstacle for delivering treatments to the injured brain in Alzheimer's disease.

Promiscuous mating systems provide the opportunity for females to bias fertilization toward particular males.However,distinguishing between male sperm com-petition and active female sperm choice is difficult for species with internal fertilization.Nevertheless,species that store and use sperm of different males in different storing struc-tures and species where females are able to expel all or part of the ejaculates after cop-ulation may be able to bias fertilization.We report a series of experiments aimed at pro-viding evidence of female sperm choice in Euxesta eluta(Hendel),a species of ulidiid fly that expels and consumes ejaculates after copulation.We found no evidence of greater reproductive success for females mated singly,multiply with the same male,or mated multiply with different males.Female E.eluta possesses two spherical spermathecae and a bursa copulatrix for sperm storage,with a ventral receptacle.There was no significant difference in storing more sperm in spermathecae 24 h after copulation than immediately after copulation.Females mated with protein-fed males had greater reproductive success than similar females mated to protein-deprived males.Protein-fed females prevented to consume the ejaculate,retained more sperm when mated to protein-fed males than when mated to protein-deprived males.Our results suggest that female E.eluta can exert control of sperm retention of higher quality males through ejaculate ejection.

Characterizing the diversity and structure of host–parasite communities is crucial to understandingtheir eco-evolutionary dynamics.Malaria and related haemosporidian parasites are responsible forfitness loss and mortality in bird species worldwide.However,despite exhibiting the greatest ornithologicalbiodiversity,avian haemosporidians from Neotropical regions are quite unexplored.Here,we analyze the genetic diversity of bird haemosporidian parasites(Plasmodium andHaemoproteus)in 1,336 individuals belonging to 206 bird species to explore for differences in diversityof parasite lineages and bird species across 5 well-differentiated Peruvian ecoregions.Wedetected 70 different haemosporidian lineages infecting 74 bird species.We showed that 25 out ofthe 70 haplotypes had not been previously recorded.Moreover,we also identified 81 new host–parasite interactions representing new host records for these haemosporidian parasites.Our outcomesrevealed that the effective diversity(as well as the richness,abundance,and Shannon–Weaver index)for both birds and parasite lineages was higher in Amazon basin ecoregions.Furthermore,we also showed that ecoregions with greater diversity of bird species also had highparasite richness,hence suggesting that host community is crucial in explaining parasite richness.Generalist parasites were found in ecoregions with lower bird diversity,implying that the abundanceand richness of hosts may shape the exploitation strategy followed by haemosporidian parasites.These outcomes reveal that Neotropical region is a major reservoir of unidentified haemosporidianlineages.Further studies analyzing host distribution and specificity of these parasites inthe tropics will provide important knowledge about phylogenetic relationships,phylogeography,and patterns of evolution and distribution of haemosporidian parasites.

Effects of Plant Extracts and Beauveria bassiana on the Activity of defense-Related Enzymes in Solanum lycopersicum L.during Interaction with Fusarium oxysporum f.sp.lycopersici

The objective to this work was to evaluate the enzymatic activity in the culture of Solanum lycopersicum L.infected with Fusarium oxysporum after the combined application of Beauveria bassiana and plant extracts.Solanum lycopersicum plantlets were transplanted 15 days after the emergency.Five days after transplanting,Beauveria bassiana spores were applied at a concentration of 1×10^(7)spores mL^(−1)onto soil(along with A.indica(N)and P.auritum(H)leaf extracts)where S.lycopersicum plants were planted.Eight days after transplanting,spores of F.oxysporum strain were applied at a concentration of 1×10^(6)spores mL^(−1)to soil where S.lycopersicum plants were growing.The development of S.lycopersicum plants was monitored for 114 days,whereby a randomized complete block treatment design was used,the roots of the plants were crushed with liquid nitrogen,after which UV/VIS spectrophotometry was used to determine protein concentration,as well as the activity ofβ-1,3-glucanases,peroxidases(POX,EC1.***.1.7),catalase and chitinases.The treatments combining B.bassiana and A.indica and P.auritum extracts,had a significant difference in enzymatic activity levels,thus contributing to better defense mechanisms and a greater protection to S.lycopersicum plants in the presence of F.oxysporum.The application of B.bassiana and plant extracts to the ground mitigated damage caused by F.oxysporum on S.lycopersicum plants.

The onset and mechanisms underlying neurodegenerative diseases remain uncertain. The main features of neurodegenerative diseases have been related with cellular and molecular events like neuronal loss, mitochondrial dysfunction and aberrant accumulation of misfolded proteins or peptides in specific areas of the brain. The most prevalent neurodegenerative diseases belonging to age-related pathologies are Alzheimer's disease, Huntington's disease, Parkinson's disease and amyotrophic lateral sclerosis. Interestingly, mitochondrial dysfunction has been observed to occur during the early onset of several neuropathological events associated to neurodegenerative diseases. The master regulator of mitochondrial quality control and energetic metabolism is the transcriptional coactivator peroxisome proliferator-activated receptor gamma coactivator 1-alpha(PGC-1α). Additionally, it has been observed that PGC-1α appears to be a key factor in maintaining neuronal survival and synaptic transmission. In fact, PGC-1α downregulation in different brain areas(hippocampus, substantia nigra, cortex, striatum and spinal cord) that occurs in function of neurological damage including oxidative stress, neuronal loss, and motor disorders has been seen in several animal and cellular models of neurodegenerative diseases. Current evidence indicates that PGC-1α upregulation may serve as a potent therapeutic approach against development and progression of neuronal damage. Remarkably, increasing evidence shows that PGC-1α deficient mice have neurodegenerative diseases-like features, as well as neurological abnormalities.
