Oocytes with better quality and development competence are highly demanded in both basic biological research and assisted reproduction *** brilliant cresyl blue(BCB) staining test is able to discern oocytes with better maturation and developmental competence in different mammalian ***,the underlying molecular mechanism remains *** the present study,we first confirmed that compared to BCB-negative oocytes,BCB-positive oocytes had higher rates of meiotic maturation,cleavage and blastocyst rates of parthenotes,but lower death rate,indicating the BCB staining could be used to select porcine immature oocytes with better potential for meiotic maturation and subsequent ***,using single oocyte RNA-seq technique,we profiled transcriptome of immature porcine oocytes positively and negatively stained by the BCB test,and identified significant abundance differences of 155 genes,including CDC5 L,LDHA,SPATA22,RGS2,PAIP1,WEE1 B and *** enrichment analysis on these differential expressed genes found functionally important signaling pathways,such as cell cycle,oocyte meiosis,spliceosome,and nucleotide excision repair *** of γH2 A.X validated more frequent occurrence of DNA double stand breaks in BCB-negative porcine GV *** blots validated CDC5 L protein level was significantly higher in BCB-positive GV *** of CDC5 L protein using antibody microinjection into oocytes indicated that CDC5 L is vital for porcine oocyte meiosis and early embryo *** investigations are still needed to study the molecular roles of identified genes and pathways in the regulation of oocyte meiotic maturation and developmental competence.

Pluripotent stem cells(PSCs) are characterized by their capacity for high self-renewal and multiple differentiation potential and include embryonic stem cells, embryonic germ cells and induced PSCs. PSCs provide a very suitable model for the studies of human diseases, drugs screening, regenerative medicine and developmental biology research. Pigs are considered as an ideal model for preclinical development of human xenotransplantation, therapeutic approaches and regenerative medicine because of their size and physiological similarity to humans. However, lack of knowledge about the derivation, characterization and pluripotency mechanisms of porcine PSCs hinders progress in these biotechnologies. In this review, we discuss the latest progress on porcine PSCs generation, evaluation criteria for pluripotency, the scienti?c and technical questions arising from these studies. We also introduce our perspectives on porcine PSC research, in the hope of providing new ideas for generating naive porcine PSCs and animal breeding.

With gradual ban on the use of antibiotics, the deficiency and excessive use of trace elements in intestinal health is gaining attention. In mammals, trace elements are essential for the development of the immune system, specifically T-cell proliferation,and differentiation. However, there remain significant gaps in our understanding of the effects of certain trace elements on T-cell immune phenotypes and functions in pigs. In this review, we summarize the specificity, development, subpopulations, and responses to pathogens of porcine T cells and the effects of functional trace elements(e.g., iron, copper, zinc, and selenium) on intestinal T-cell immunity during early-life health in pigs. Furthermore, we discuss the current trends of research on the crosstalk mechanisms between trace elements and T-cell immunity. The present review expands our knowledge of the association between trace elements and T-cell immunity and provides an opportunity to utilize the metabolism of trace elements as a target to treat various diseases.

The placenta plays a crucial role in successful mammalian *** animals possess a semi-invasive placenta characterized by a highly vascularized structure formed by maternal endometrial caruncles and fetal placental cotyledons,essential for full-term fetal *** cow placenta harbors at least two trophoblast cell populations:uninucleate(UNC)and binucleate(BNC)***,the limited capacity to elucidate the transcriptomic dynamics of the placental natural environment has resulted in a poor understanding of both the molecular and cellular interactions between trophoblast cells and niches,and the molecular mechanisms governing trophoblast differentiation and *** fill this knowledge gap,we employed Stereo-seq to map spatial gene expression patterns at near single-cell resolution in the cow placenta at 90 and 130 days of gestation,attaining high-resolution,spatially resolved gene expression *** on clustering and cell marker gene expression analyses,key transcription factors,including YBX1 and NPAS2,were shown to regulate the heterogeneity of trophoblast cell *** communication and trajectory analysis provided a framework for understanding cell-cell interactions and the differentiation of trophoblasts into BNCs in the placental *** analysis of cell trajectories identified a set of genes involved in regulation of trophoblast ***,spatial modules and co-variant genes that help shape specific tissue structures were ***,these findings provide foundational insights into important biological pathways critical to the placental development and function in cows.

Several potential oxidative agents have damaging effects on mammalian reproductive *** study was conducted to investigate the effects of glutamate(Glu)and aspartate(Asp)supplementation on antioxidant enzymes and immune defense systems in the outer scrotum of boars injected with H2O2.A total of 24 healthy boars were randomly divided into 4 treatment groups:control(basal diet,saline-treated),H2O2(basal diet,H2O2-challenged outer scrotum(1 m L kg^–1 BW)),Glu(basal diet+2%Glu,H2O2-challenged),and Asp(basal diet+2%Asp,H2O2-challenged).Our results showed that both Glu and Asp supplementation improved testicular morphology and decreased the genital index in the H2O2-treated *** and Asp administration increased the antioxidant enzyme activities and affected the testicular inflammatory cytokine secretion but had no effect on sex hormone ***,the m RNA expression of CAT,Cu Zn SOD,and GPx4 was altered in the testes and epididymis of boars treated with Asp and *** and Asp supplementation also modulated the expression of TGF-β1,IL-10,TNF-α,IL-6 and IL-1βin the testis and *** results indicate that dietary Glu and Asp supplementation might enhance antioxidant capacity and regulate the secretion and expression of inflammatory cytokines to protect the testes and epididymis of boars against oxidative stress.

Nonalcoholic fatty liver disease(NAFLD) is characterized by hepatic steatosis and insulin resistance and there are currently no approved drugs for its *** of mTOR complex1(mTORCl) and subsequent impairment of the transcription factor EB(TFEB)-mediated autophagy-lysosomal pathway(ALP) are implicated in the development of ***,agents that augment hepatic TFEB transcriptional activity may have therapeutic potential against *** objective of this study was to investigate the effects of nuciferine,a major active component from lotus leaf,on NAFLD and its underlying mechanism of *** we show that nuciferine activated ALP and alleviated steatosis,insulin resistance in the livers of NAFLD mice and palmitic acid-challenged hepatocytes in a TFEB-dependent *** investigation revealed that nuciferine interacts with the Ragulator subunit hepatitis B X-interacting protein and impairs the interaction of the Ragulator complex with Rag GTPases,thereby suppressing lysosomal localization and activity of mTORC1,which activates TFEB-mediated ALP and further ameliorates hepatic steatosis and insulin *** present results indicate that nuciferine may be a potential agent for treating NAFLD and that regulation of the mTORCl-TFEB-ALP axis could represent a novel pharmacological strategy to combat NAFLD.

Recent experiments have pointed out that cellular uptake is strongly dependent on the physical dimensions of endocytosed nanoparticles and tile optimal radius of endocytosed virus-like particle coated by transferrin is around 50 nm. As the same time, the dimensions of receptor-ligand complex have strong effects on the size-dependent exclusion of proteins ill cell environments. Inspired by these experimental results, a continuum elastic model is constructed to resolve the relationship between the dinlensions of receptor-ligand complex and the optimal radius of endocytosed virus-like particle. These results demonstrate that the optimal radius of endocytosed virus-like particle depends on the dimensions of reeeptor-ligand complex and the dimension of receptor-ligand complex reduces the depletion zone.
