摘      要:Oocytes with better quality and development competence are highly demanded in both basic biological research and assisted reproduction *** brilliant cresyl blue(BCB) staining test is able to discern oocytes with better maturation and developmental competence in different mammalian ***,the underlying molecular mechanism remains *** the present study,we first confirmed that compared to BCB-negative oocytes,BCB-positive oocytes had higher rates of meiotic maturation,cleavage and blastocyst rates of parthenotes,but lower death rate,indicating the BCB staining could be used to select porcine immature oocytes with better potential for meiotic maturation and subsequent ***,using single oocyte RNA-seq technique,we profiled transcriptome of immature porcine oocytes positively and negatively stained by the BCB test,and identified significant abundance differences of 155 genes,including CDC5 L,LDHA,SPATA22,RGS2,PAIP1,WEE1 B and *** enrichment analysis on these differential expressed genes found functionally important signaling pathways,such as cell cycle,oocyte meiosis,spliceosome,and nucleotide excision repair *** of γH2 A.X validated more frequent occurrence of DNA double stand breaks in BCB-negative porcine GV *** blots validated CDC5 L protein level was significantly higher in BCB-positive GV *** of CDC5 L protein using antibody microinjection into oocytes indicated that CDC5 L is vital for porcine oocyte meiosis and early embryo *** investigations are still needed to study the molecular roles of identified genes and pathways in the regulation of oocyte meiotic maturation and developmental competence.
