Establishment and application of a surrogate model for human Ebola virus disease in BSL-2 laboratory

The Ebola virus(EBOV)is a member of the Orthoebolavirus genus,Filoviridae family,which causes severe hemorrhagic diseases in humans and non-human primates(NHPs),with a case fatality rate of up to 90%.The development of countermeasures against EBOV has been hindered by the lack of ideal animal models,as EBOV requires handling in biosafety level(BSL)-4 ***,accessible and convenient animal models are urgently needed to promote prophylactic and therapeutic approaches against *** this study,a recombinant vesicular stomatitis virus expressing Ebola virus glycoprotein(VSV-EBOV/GP)was constructed and applied as a surrogate virus,establishing a lethal infection in *** infection with VSV-EBOV/GP,3-week-old female Syrian hamsters exhibited disease signs such as weight loss,multi-organ failure,severe uveitis,high viral loads,and developed severe systemic diseases similar to those observed in human EBOV *** animals succumbed at 2–3 days post-infection(dpi).Histopathological changes indicated that VSV-EBOV/GP targeted liver cells,suggesting that the tissue tropism of VSV-EBOV/GP was comparable to wild-type EBOV(WT EBOV).Notably,the pathogenicity of the VSV-EBOV/GP was found to be species-specific,age-related,gender-associated,and challenge ***,equine anti-EBOV immunoglobulins and a subunit vaccine were validated using this ***,this surrogate model represents a safe,effective,and economical tool for rapid preclinical evaluation of medical countermeasures against EBOV under BSL-2 conditions,which would accelerate technological advances and breakthroughs in confronting Ebola virus disease.

In recent years,the issue of aircraft icing has gained widespread *** breaking and detachment of dynamic ice can pose a threat to flight ***,the shedding and fracture mechanisms of dynamic ice are unclear and cannot meet the engineering needs of ice-shedding hazard ***,studying the fracture toughness of ice bodies has extremely important practical *** address this issue,this article uses a centrally cracked Brazilian disk(CCBD)specimen to measure the pure modeⅠtoughness and pure modeⅡfracture toughness of freshwater ice at different loading *** mixed-mode(Ⅰ–Ⅱ)fracture characteristics of ice are discussed,and the experimental results are compared and analyzed with the theoretical values of the generalized maximum tangential stress(GMTS)criterion considering the influence of *** results indicated that as the loading rate increases,the pure modeⅠtoughness and pure modeⅡfracture toughness of freshwater ice decrease,and the fracture toughness of freshwater ice is more sensitive to the loading rate.Ⅰn terms of fracture criteria,the theoretical value of the ratio of pure modeⅡfracture toughness to pure modeⅠfracture toughness based on the GMTS criterion is in good agreement with the experimental value,while the theoretical value based on the maximum tangential stress(MTS)criterion deviates significantly from the experimental value,indicating that the GMTS criterion considering the influence of T-stress can better predict the experimental results.

The biosecurity hazards caused by pathogenic fungus have been widely *** the long-term coexistence of eukaryotic pathogens and quorum sensing bacteria in different habitats in environments,we hypothesized that they have social interactions via signal *** this work,we firstly discovered the well-known bacterial signal molecules play an adverse role in the cell morphology and metabolism in a model pathogen Trichosporon asahii.N-Tetradecanoyl-L-homoserine lactone(C14-HSL)was discovered to increase pathogen hazards of ***,which limited mycelium by 52%,but enhanced cell aggregation by 93%.Higher fluorescence intensity of tryptophan(59%)and aromatic protein(2-fold)contents after the treatment of C14-HSL,indicating that aromatic proteins helped aggregate Trichosporon and showed *** analysis revealed that C14-HSL upregulated the shikimate pathway(above1-fold)located in downstream of tricarboxylic acid cycle,which contributed to the synthesis of more aromatic proteins and the formation of larger *** limited mycelial growth of *** attributed to the up-regulated expressions of cell cycle *** fungal transboundary response to bacterial C14-HSL was controlled by signal transduction *** study provides new insights into the co-evolution of bacterial and pathogenic fungi in microecosystems.

Harmful components in cigarette smoke,such as nicotine,tar,and organic particulate matter,are the primary culprits behind lung *** conventional filter materials based on cellulose,carbon,and molecular sieves exhibit commendable filtration capabilities,their high cost restricts their widespread *** on this,the authors aim to prepare PET-based filter materials with good adsorption properties through a simple surface functionalisation *** adsorption performance of the PET-based non-woven fabric was enhanced by the introduction of sodium alginate(SA).The gas adsorption experiments results reveal that SA-modified PET(SA-PET)exhibits significantly improved filtration efficiency for nicotine,tar,and total particulate matter—increasing by 27.1%,26.2%,and 21.3%,***,SA-PET exhibits more odour control ability than traditional activated carbon-based filter *** results prove that surface-functionalised SA-PET has better filtration performance for harmful substances in smoke and provides a new strategy for the design of high-performance filtration materials.

Birnessite is ubiquitous in the natural environment where heavy metals are retained and easily *** surface properties and structure of birnessite change with the changes in external environmental conditions,which also affects the fate of heavy *** the effect and mechanism of the birnessite phase transition process on heavy metals is the key to taking effective measures to prevent and control heavy metal ***,the four transformation pathways of birnessite are summarized first in this ***,the relationship between transformation pathways and environmental conditions is *** relevant environmental conditions include abiotic(e.g.,co-existing ions,pH,oxygen pressure,temperature,electric field,light,aging,pressure)and biotic factors(e.g.,microorganisms,biomolecules).The phase transformation is achieved by the key intermediate of Mn(Ⅲ)through interlayer-condensation,folding,neutralizationdisproportionation,and dissolution-recrystallization *** AOS(average oxidation state)of Mn and interlayer spacing are closely correlated with the phase transformation of *** but not least,the mechanisms of heavy metals immobilization in the transformation process of birnessite are summed *** involve isomorphous substitution,redox,complexation,hydration/dehydration,*** transformation of birnessite and its implication on heavy metals will be helpful for understanding and predicting the behavior of heavy metals and the crucial phase of manganese oxides/hydroxides in natural and engineered environments.

Seismic fragility analysis of three-tower cable-stayed bridges with three different structural systems,including rigid system(RS),floating system(FS),and passive energy dissipation system(PEDS),is conducted to study the effects of connection configurations on seismic responses and *** element models of bridges are established using OpenSees.A new ground motion screening method based on the statistical characteristic of the predominant period is proposed to avoid irregular behavior in the selection process of ground motions,and incremental dynamic analysis(IDA)is performed to develop components and systems fragility *** effects of damper failure on calculated results for PEDS are examined in terms of seismic response and fragility *** results show that the bridge tower is the most affected component by different structural *** RS,the fragility of the middle tower is significantly higher than other components,and the bridge failure starts from the middle tower,exhibiting a characteristic of local *** FS and PEDS,the fragility of the edge tower is higher than the middle *** system fragility of RS is higher than FS and *** the failure of dampers into account is necessary to obtain reliable seismic capacity of cable-stayed bridges.

Automation advancements prompts the extensive integration of collaborative robot(cobot)across a range of *** to the commonly used design approach of increasing the payload-to-weight ratio of cobot to enhance load capacity,equal attention should be paid to the dynamic response characteristics of cobot during the design process to make the cobot more *** this paper,a new method for designing the drive train parameters of cobot is ***,based on the analysis of factors influencing the load capacity and dynamic response characteristics,design criteria for both aspects are established for cobot with all optimization design criteria normalized within the design ***,with the cobot in the horizontal pose,the motor design scheme is discretized and it takes the joint motor diameter and gearbox speed ratio as optimization design ***,all the discrete values of the optimization objectives are obtained through the enumeration method and the Pareto front is used to select the optimal solution through multi-objective *** on the cobot design method proposed in this paper,a six-axis cobot is designed and compared with the commercial *** result shows that the load capacity of the designed cobot in this paper reaches 8.4 kg,surpassing the 5 kg load capacity commercial cobot which is used as a *** minimum resonance frequency of the joints is 42.70 Hz.

水体温度变化对河流可溶性无机氮(DIN)输送有着强烈控制作用.然而,在全球尺度上河流DIN输送量对水温度变化的响应尚不清楚.因此,本文基于陆面过程模式,耦合河流水温估算和DIN传输方案,设定有,无动态水温情景,对比研究陆面模拟中水温变化对河流DIN通量变化的影响.结果表明:在考虑水温动态变化后,在30°N和30°S之间, DIN通量年振幅减小5%–25%.在中国东部地区,水温动态变化使河流DIN通量在夏季减少1%–3%,在冬季增加1%–5%,对DIN通量具有明显的季节性约束作用,表明动态水温的表达在河流DIN输送模拟中的重要性.

In this paper,a seismic and vibration reduction measure of subway station is developed by setting a segmented isolation layer between the sidewall of structure and the diaphragm *** segmented isolation layer consists of a rigid layer and a flexible *** rigid layer is installed at the joint section between the structural sidewall and slab,and the flexible layer is installed at the remaining sections.A diaphragm wall-segmented isolation layer-subway station structure system is *** and vibration control performance of the diaphragm wall-segmented isolation layer-subway station structure system is evaluated by the detailed numerical ***,a three-dimensional nonlinear time-history analysis is carried out to study the seismic response of the station structure by considering the effect of different earthquake motions and stiffness of segmented isolation ***,the vibration response of site under training loading is also studied by considering the influence of different train velocities and stiffness of the segmented isolation *** results demonstrate that the diaphragm wall-segmented isolation layer-subway station structure system can not only effectively reduce the lateral deformation of station structure,but also reduce the tensile damage of the roof *** the other hand,the developed reduction measure can also significantly reduce the vertical peak displacements of site under training loading.
