BACKGROUND Improving the intraoperative and postoperative performance of laparoscopic hepatectomy was quite a challenge for liver *** To determine the benefits of indocyanine green(ICG)fluorescence imaging in patients with hepatocellular carcinoma(HCC)who underwent laparoscopic hepatectomy during and after *** We retrospectively collected the clinicopathological data of 107 patients who successfully underwent laparoscopic hepatectomy at Zhongshan Hospital(Xiamen),Fudan University from June 2022 to June *** using the ICG fluorescence imaging technique,we divided them into the ICG and non-ICG *** eliminate statistical bias,a 1:1 propensity score matching analysis was *** comparison of perioperative outcomes,including inflammationrelated markers and progression-free survival,was analyzed *** Intraoperatively,the ICG group exhibited lower blood loss,a shorter surgical time,lower hepatic inflow occlusion(HIO)frequency,and a shorter total HIO ***,the participation of ICG resulted in a shorter duration of hospitalization(6.5 vs 7.6 days,P=0.03)and postoperative inflammatory response attenuation(lower neutrophil-lymphocyte ratio on the first day after surgery and platelet-lymphocyte ratio on the third day,P<0.05).Although the differences were not significant,the levels of all inflammation-related markers were lower in the ICG *** rates of postoperative complications and the survival analyses,including progression-free and overall survivals showed no significant difference between the *** The involvement of ICG fluorescence imaging may lead to improved perioperative outcomes,especially postoperative inflammatory response attenuation,and ultimately improve HCC patients’recovery after surgery.

Viral epidemics pose major threats to global health and economies. A hallmark of viral infection is the reshaping of host cell membranes and cytoskeletons to form organelle-like structures, known as viral factories, which support viral genome replication. Viral infection in many cases induces the cytoskeletal network to form cage-like structures around viral factories, including actin rings, microtubule cages, and intermediate filament cages. Viruses hijack various organelles to create these replication factories, such as viroplasms, spherules, double-membrane vesicles, tubes, and nuclear viral factories. This review specifically examines the roles of cytoskeletal elements and the endomembrane system in material transport, structural support, and biochemical regulation during viral factory formation. Furthermore, we discuss the broader implications of these interactions for viral replication and highlight potential future research directions.

Fungi are known to be crucial in the formation and stabilization of soil organic matter through their exudates and dead residues(necromass).Yet,it remains unclear how fungi contribute to the persistence of carbon in soils over millennial scales across ***,this study fills the knowledge gap by linking fungal carbon stocks with minerals-associated carbon stocks across major biomes.A strong correlation between fungal biomass and reactive mineral-associated carbon stocks in soils across six biomes supports the substantial role of fungi in soil carbon persistence at the global *** spatial resolution nanoscale secondary ion mass spectrometry revealed that fungi may stabilize carbon by forming organo-mineral associations through their close physical connection to reactive minerals and weathered *** propose a conceptual model that emphasizes the dualistic role of hypha-mineral interactions:(i)fungi accelerate organic matter decomposition by producing reactive oxygen species on the hypha-mineral interfaces,and(ii)fungi stabilize their residues on mineral surfaces,leading to millennial scale persistence of soil ***,fungi play a fundamental role in ecosystems that extends beyond the decomposition of persistent *** are crucial in stabilizing carbon on mineral surfaces,thereby facilitating the long-term removal of carbon from rapid biotic cycling.

New guidelines for fluorophore application in peroxisome targeting analyses in transient plant expression systems

Peroxisome research has been revolutionized by proteome studies combined with in vivo subcellular targeting analyses. Yellow and cyan fluorescent protein(YFP and CFP) are the classical fluorophores of plant peroxisome research. In the new transient expression system of Arabidopsis seedlings co-cultivated with Agrobacterium we detected the YFP fusion of one candidate protein in peroxisomes, but only upon co-transformation with the peroxisome marker, CFP-PTS1. The data suggested that the YFP fusion was directed to peroxisomes due to its weak heterodimerization ability with CFP-PTS1,allowing piggy-back import into peroxisomes. Indeed, if co-expressed with monomeric Cerulean-PTS1(mCer-PTS1),the YFP fusion was no longer matrix localized. We systematically investigated the occurrence and extent of dimerization-based piggy-back import for different fluorophore combinations in five major transient plant expression systems. In Arabidopsis seedlings and tobacco leaves both untagged YFP and monomeric Venus were imported into peroxisomes if co-expressed with CFP-PTS1 but not with mCer-PTS1. By contrast, piggy-back import of cytosolic proteins was not observed in Arabidopsis and tobacco protoplasts or in onion epidermal cells for any fluorophore combination at any time point. Based on these important results we formulate new guidelines for fluorophore usage and experimental design to guarantee reliable identification of novel plant peroxisomal proteins.

Mixed-charge glycopolypeptides as antibacterial coatings with long-term activity

Planktonic bacteria adhere and subsequently form biofilms on implantable medical devices can cause severe infections that have become the major types of hospital-acquired infections. Traditional coatings for the implants are frequently lack of long-term antifouling and bactericidal activities. It is still a big challenge to simultaneously improve the antifouling and bactericidal activities of the coatings. Herein,we report that mixed-charge glycopolypeptide coatings are of long-term antibacterial activities to efficiently inhibit the biofilm growth. The glycosylation of mixed-charge polypeptides has led to a significant improvement of both antifouling and bactericidal activities. The cooperative effect of the saccharide residues and mixed-charge residues improved the resistance of the polypeptide coatings against protein adsorption. The saccharide and L-glutamic acid(E) residues collectively enhanced the bacterial membrane-disruption of cationic L-lysine(K) residues, leading to potent bactericidal activity. Meanwhile,the glycopolypeptide coatings showed superior biocompatibility, long-term antibiofilm and anti-infection properties in two types of mouse subcutaneous infection models and one type of mouse urinary tract infection model. This work provides a new strategy to achieve antibacterial coatings with long-term activities for preventing implantable medical device associated infections.
