Over the past years, infectious disease has caused enormous economic loss in pig industry. Among the pathogens, gram negative bacteria not only cause inflammation, but also cause different diseases and make the pigs more susceptible to virus infection. Vaccination, medication and elimination of sick pigs are major strategies of controlling disease. Genetic methods, such as selection of disease resistance in the pig, have not been widely used. Recently, the completion of the porcine whole genome sequencing has provided powerful tools to identify the genome regions that harboring genes controlling disease or immunity. Immunogenornics, which combines DNA variations, transcriptorne, immune response, and QTL mapping data to illustrate the interactions between pathogen and host immune system, will be an effective genomics tool for identification of disease resistance genes in pigs. These genes will be potential targets for disease resistance in breeding programs. This paper reviewed the progress of disease resistance study in the pig focusing on Gram-negative bacilli. Major porcine Gram-negative bacilli and diseases, suggested candidate genes/pathways against porcine Gram-negative bacilli, and distributions of QTLs for immune capacity on pig chromosomes were summarized. Some tools for immunogenomics research were described. We conclude that integration of sequencing, whole genome associations, functional genomics studies, and immune response information is necessary to illustrate molecular mechanisms and key genes in disease resistance.

Background： Early pregnancy failure has a profound impact on both human reproductive health and animal production. 2/3 pregnancy failures occur during the peri-implantation period; however, the underlying mechanism（s） remains unclear. Well-organized modification of the endometrium to a receptive state is critical to establish pregnancy Aberrant endometrial modification during implantation is thought to be largely responsible for early pregnancy loss. Result： In this study, using well-managed recipient ewes that received embryo transfer as model, we compared the endometrial proteome between pregnant and non-pregnant ewes during implantation period. After embryo transfer, recipients were assigned as pregnant or non-pregnant ewes according to the presence or absence of an elongated conceptus at Day 17 of pregnancy. By comparing the endometrial proteomic profiles between pregnant and non-pregnant ewes, we identified 94 and 257 differentially expressed proteins （DEPs） in the endometrial caruncular and intercaruncular areas, respectively. Functional analysis showed that the DEPs were mainly associated with immune response, nutrient transport and utilization, as well as proteasome-mediated proteolysis. Conclusion： These analysis imply that dysfunction of these biological processes or pathways of DEP in the endometrium is highly associated with early pregnancy loss. In addition, many proteins that are essential for the establishment of pregnancy showed dysregulation in the endometrium of non-pregnant ewes. These proteins, as potential candidates, may contribute to early pregnancy loss.

Background： The p21-activated kinase 1 （PAK1）is essential of microtubule assembly during oocyte meiotic maturation porcine oocytes. for mitosis and plays an important role in the regulatio in mice; however, little is known about its role in Result： Total p21-activated kinase 1 （PAK1） and phosphorylated PAK1 at Thr423 （PAK1^Thr423） were consistently expressed in porcine oocytes from the germinal vesicle （GV） to the second metaphase （MII） stages, but phosphorylation of histone H3 at Serr10 （H3^ser10） was only expressed after the GV stage. Immunofiuorescence analysis revealed that PAK1Thr423 and H3^ser10 colocalized on chromosomes after the GV stage. Blocking of endogenous PAK1^Thr423 by injecting a specific antibody decreased the phosphorylation level of H3^ser10; however, it had no impact on chromatin condensation, meiotic progression, cleavage rate of blastomeres or the rate of blastocyst formation. Conclusion： Phosphorylation of PAK1^Thr423 is a spontaneous activation process and the activated PAK1^Thr423 can promote the phosphorylation of H3^ser10; however, this pathway is not required for meiotic maturation of porcine oocytes or early embryonic development.

Background: Different production systems and climates could lead to genotype-by-environment(G × E) interactions between populations, and the inclusion of G × E interactions is becoming essential in breeding decisions. The objective of this study was to investigate the performance of multi-trait models in genomic prediction in a limited number of environments with G × E ***: In total, 2,688 and 1,384 individuals with growth and reproduction phenotypes, respectively, from two Yorkshire pig populations with similar genetic backgrounds were genotyped with the PorcineSNP80 ***-and multi-trait models with genomic best linear unbiased prediction(GBLUP) and BayesC π were implemented to investigate their genomic prediction abilities with 20 replicates of five-fold *** results regarding between-environment genetic correlations of growth and reproductive traits(ranging from 0.618 to 0.723) indicated the existence of G × E interactions between these two Yorkshire pig populations. For single-trait models, genomic prediction with GBLUP was only 1.1% more accurate on average in the combined population than in single populations, and no significant improvements were obtained by BayesC π for most traits. In addition, single-trait models with either GBLUP or BayesC π produced greater bias for the combined population than for single populations. However, multi-trait models with GBLUP and BayesC π better accommodated G × E interactions,yielding 2.2% – 3.8% and 1.0% – 2.5% higher prediction accuracies for growth and reproductive traits, respectively,compared to those for single-trait models of single populations and the combined population. The multi-trait models also yielded lower bias and larger gains in the case of a small reference population. Thalassemia is a group of genetically heterogeneous diseases characterized by hemolytic *** investigate molecular characteristics ofα-andβ-thalassemia among young individuals of marriageable age in Guangdong Province,24,788 subjects with suspected thalassemia were genetically tested forα-andβ-thalassemia by GapPCR and reverse dot blot during 2018–*** suspected rare thalassemia cases,DNA sequencing was performed to identify rare and unknown thalassemia gene mutations.A total of 14,346 thalassemia carriers were detected,including 7,556 cases ofα-thalassemia with 25 genotypes and 8α-gene mutations identified,5,860 cases ofβ-thalassemia with 18 genotypes and 18β-gene mutations identified,and 930 cases of compoundα/β-*** them,the frequency of--SEA deletion was the highest inα-thalassemia(66.01%),followed by-α3.7(17.98%)and-α.24(8.22%),and the frequency of CD41-42(-TCTT)mutation was the highest inβ-thalassemia(38.38%),followed by IVS-II-654(C>T)(25.67%),-28(A>G)(15.76%),and CD17(10.01%).In addition,5 rare mutations(--THAI and HKαα,CD113,-90,and CD56)were found in the study *** results revealed molecular epidemiological background ofα-andβ-thalassemia in Guangdong Province,which can support optimization of thalassemia prevention and control *** demonstrated that thalassemia is heterogeneous with significant geographical differences and population specificity.
