Bioreductive prodrugs as cancer therapeutics:targeting tumor hypoxia

Hypoxia, a state of low oxygen, is a common feature of solid tumors and is associated with disease progression as well as resistance to radiotherapy and certain chemotherapeutic drugs. Hypoxic regions in tumors, therefore, represent attractive targets for cancer therapy. To date, five distinct classes of bioreactive prodrugs have been developed to target hypoxic cells in solid tumors. These hypoxia-activated prodrugs, including nitro compounds, N-oxides, quinones, and metal complexes, generally share a common mechanism of activation whereby they are reduced by intracellular oxidoreductases in an oxygensensitive manner to form cytotoxins. Several examples including PR-104, TH-302, and EO9 are currently undergoing phase II and phase III clinical evaluation. In this review, we discuss the nature of tumor hypoxia as a therapeutic target, focusing on the development of bioreductive prodrugs. We also describe the current knowledge of how each prodrug class is activated and detail the clinical progress of leading examples.

Metagenomic analysis to determine the characteristics of antibiotic resistance genes in typical antibiotic-contaminated sediments

Comprehensive studies of the effects of various physical and chemical variables(including heavymetals),antibiotics,and microorganisms in the environment on antibiotic resistance genes are *** collected sediment samples from the Shatian Lake aquaculture area and surrounding lakes and rivers located in Shanghai,*** spatial distribution of sediment ARGs was assessed by metagenomic analysis that revealed 26 ARG types(510 subtypes),dominated by Multidrug,β-lactam,Aminoglycoside,Glycopeptides,Fluoroquinolone,and *** discriminant analysis indicated that antibiotics(SAs and MLs)in the aqueous environment and sediment along with water TN and TP were the key variables affecting the abundance distribution of total ***,the main environmental drivers and key influences differed among the different *** total ARGs,the environmental subtypes affecting their structural composition and distribution characteristics were mainly antibiotic *** analysis showed a significant correlation between ARGs and microbial communities in the sediment in the survey *** analysis revealed that most of the target ARGs were significantly and positively correlated with microorganisms,and a small number of ARGs(e.g.,rpoB,mdtC,and efpA)were highly significantly and positively correlated with microorganisms(e.g.,Knoellia,Tetrasphaera,and Gem-matirosa).Potential hosts for the major ARGs included Actinobacteria,Proteobacteria,and *** study provides new insight and a comprehensive assessment of the distribution and abundance of ARGs and the drivers of ARG occurrence and transmission.

Enhanced urease inhibition activity of Ag nanomaterials capped with N-substituted methyl 5-acetamido-β-resorcylate

Medically, bacterial ureases are important virulent factors and are used for treatment of peptic ulcers and urinary tones. Reported urease inhibitors are associated with various side effects including antibiotic resistance as a major one. Still there is an urgent need to synthesize new urease inhibitors. In this context we have synthesized new urease inhibitor i.e. AgL that is composed of Ag nanomaterials capped with N-substituted methyl 5-acetamido-β-resorcylate(L). The conjugation of L to silver was confirmed through FTIR, UV–vis and TEM *** silver nanomaterials(Ag) were also prepared. The stability of AgL nanostructures was determined against various parameters(temperature, high salt concentration, pH) and found to be stable. The in vitro antimicrobial(antibacterial, antifungal), enzyme inhibition(xanthine oxidase, urease, carbonic anhydrase, α-chymotrypsin,cholinesterase) and antioxidant activities of AgL were investigated and compared with Ag, L and standard *** comparison to other bioactivities, AgL shows statistically enhanced selective enzyme inhibition activity against urease enzyme. Urease inhibition activity of AgL was significantly greater than standard drug(thiourea),L and Ag. On a per weight basis, AgL required about 11–18 times less amount of L for inhibition of urease enzyme.

Ocular flora in patients undergoing intravitreal injections: antibiotic resistance patterns and susceptibility to antiseptic picloxydine

AIM: To study antibiotic resistance patterns and susceptibility to eye antiseptic picloxydine of conjunctival flora in patients undergoing intravitreal injections(IVIs).METHODS: Conjunctival swabs were taken in 4 groups of patients, 20 patients in each group(n=80): without IVIs and ophthalmic operations in history(group N1;control group);with the first IVI and antibiotic eye drops Tobrex applied 3 d before IVI and 5 d after it(group N2);with 20 or more IVIs and repeated courses of antibiotic eye drops(group N3);with the first IVI and antiseptic eye drops Vitabact(picloxydine) applied 3 d before IVI and 5 d after it(group N4). In groups N2 and N4 swabs were taken at baseline and after the treatment. Efficacy of picloxydine in inhibition of growth of conjunctival isolates susceptible and resistant to antibiotic was studied in vitro. Minimal inhibition concentrations(MIC) were determined with microdilution ***: Two of the three patients who had to undergo the IVI procedure showed conjunctiva bacterial contamination. Along with few Staphylococcus aureus and Gram-negative isolates susceptible to most antibiotics, the majority(71%-77%) of causative agents were coagulase-negative Staphylococci(Co NS), 40%-50% of which were multidrug resistant(MDR). Eye disinfection in the operating room and peri-injection courses of Tobrex or Vitabact resulted in total elimination of isolates found at baseline. However, in 10% and 20% of patients, respectively, recolonization of the conjunctiva with differing strains occurred. In patients with repeated IVI and Tobrex/Maxitrol treatment, the conjunctival flora showed high resistance rates: 90% of Co NS were MDR. In the in vitro study, picloxydine showed bactericidal effect against Staphylococci isolates both antibiotic resistant and susceptible with MIC≥13.56 μg/m L. Effects of exercise-based cardiac rehabilitation on inflammatory biomarkers in patients with cardiovascular disease:a systematic review with meta-analysis

Background:Evidence supports the anti-inflammatory effect of exercise-based cardiac rehabilitation(CR)in patients with cardiovascular disease(CVD).Nonetheless,the influence of training variables on the effects of exercise training is ***,this systematic review with meta-analysis aims to i)estimate the effects of exercise-based CR on the circulating levels of inflammatory biomarkers in patients with CVD based on the type of exercise,and ii)analyse the influence of other potential moderator ***:Electronic searches were performed in PubMed,Scopus,and Web of Science databases up to October 2022.A random-effects models of standardised mean difference(SMD)were *** analyses and meta-regressions were used to carry out heterogeneity ***:Aerobic exercise diminished the levels of C-reactive protein compared with the control group in patients with CVD(SMD+=-0.33[95%CI=-0.47,-0.20),as well as the levels of tumor necrosis factor-alfa exclusively in patients with chronic heart failure(SMD+=-0.38[95%CI=-0.59,-0.17]).Moreover,heterogeneity analyses showed that the effect of aerobic exercise on vascular cell adhesion molecule-1 is higher in studies that carried out a CR programme based exclusively on exercise,as well as in studies that performed a shorter intervention period or carried out a lower number of exercise sessions(P≤0.05).Conclusion:Our findings support the anti-inflammatory effect of aerobic exercise in patients with *** low number of studies testing interventions using resistance or combined exercise precludes a definitive answer about their anti-inflammatory effects in this patient population.
