AIM:To characterise expression of interleukin 6(IL-6),a potent proinflammatory cytokine,in the occurrence and development of inflammatory bowel disease(IBD) and investigate its effect on neuroimmunomodulation and immune homeostasis ***:In this study,rats with colitis induced by trinitrobenzene sulfonic acid(TNBS) were sacrificed on days 3,7,14,21 and 28 after *** the controls,the TNBS was just replaced by equivalent amount of phosphate buffered solution(PBS,0.01 mol/L).IL-6 mRNA expression in brain and colon tissues in each phase was evaluated by real-time reverse transcriptionpolymerase chain reaction,and cellular localisation and protein level of IL-6 was determined by ***:At day 7,mRNA expression of IL-6 was significantly higher in the colon and brain of IBD rats than that of the *** protein level was also significantly higher in colon,hypothalamus and cerebral cortex of IBD rats compared with the *** there are similar temporal trends in IL-6 mRNA expression and protein levels in all positions with a persistent increase to a peak at day 7,followed by a decline and gradual return to normal ***:These results revealed that changes in IL-6 expression in brain and colon tissues occur in different phases of ***,we propose that the nerve centre regulates and controls the occurrence and development of IBD via IL-6.

Our trial was conducted to study the effects of formic acid(FA) and potassium di-formate(KDF) in broiler ration on performance, carcass traits, blood biochemical, intestinal microbial load, histological picture of intestine and immune parameters of broilers. In this study 360 one-day-old broiler chicks were divided to 3 groups with 3 replicates of 40 chicks each. The trial continued for 35 days. The control group was fed only basal diet(G1). Group 2(G2) were fed basal diet supplemented with FA(5 g/kg diet), and group 3(G3) received basal diet supplemented with KDF(5 g/kg diet). The results showed that both FA and KDF significantly increased body weight gain(BWG), dressing percentage of broilers and significantly decreased feed conversion ratio(FCR)(P < 0.05). The highest percent of breast and thigh was observed in G3. The improvement in villus height was observed in G2 and G3 compared with the control one, and the highest was in G3. The results evidence that the using of FA or KDF in broiler feeds have significant effects on performance, immune parameters, and gut health without having any significant effects on blood biochemical. However, KDF is more effective than FA as little amount of FA reaches the small intestine due to metabolism and absorption, whereas KDF permits a proportion of FA to pass through the fore-gut intact and enter the small intestinal tract. In addition, FA has a strong odor and corrosiveness to gastrointestinal tract which limits its use.

Feline calicivirus (FCV) is a common cause of upper respiratory and oral disease in cats. Highly virulent systemic strains of FCV (vs FCV) have been described. These vs FCV isolates cause characteristic edema, cutaneous ulcers and other clinical signs typically associated with FCV infection. Vs FCV isolates also cause high mortality even in previously vaccinated cats. We reported previously that the FCV serum cross-neutralization profile of cat serum generated using the oralnasal route of administration is broader than with subcutaneous administration (SC), as measured with a 26-FCV viral panel (Rong et al., Virus Research 122:95-108, 2006). In this report, we tested the in vivo ef- ficacy of the FCV vaccine, in a 4-way (FCV-FHV-FPV-FCp) format, by using a highly virulent vs FCV- 33585 as the challenge virus. Vaccines were administered as 2-dose subcutaneouly (SC/SC), or subcutaneously followed by orally (SC/Oral). The mortality induced by vs FCV-33585 in unvaccinated control cats was 78% (7 out of 9 cats). The mortality decreased to 44% (4 out of 9 cats) with cats vaccinated with the 4-way vaccine given SC/SC. However, when this vaccine was given SC/Oral, the mortality decreased to 10% (1 out of 10 cats). The clinical scores, calculated based on frequency and severity of various clinical signs, correlated with mortality data. These results demonstrated that oral administration of FCV vaccines, as the second dose following the first dose of subcutaneious administration, ehances FCV efficacy against challenge of a highly virulent vs FCV. We propose that not only oral vaccination offers convenience and needle-free inoculation, it also enhances FCV vaccine efficacy.

To investigate the effects of etomidate,emulsified isoflurane,and their combination with dexmedetomidine on physiological parameters,electrocardiogram(ECG) results,and the quality of induction and recovery during isoflurane maintenance anaesthesia.5 mixed-breed dogs received each of four treatments:etomidate(E group);emulsified isoflurane(EI group);both dexmedetomidine and etomidate(DE group);or both dexmedetomidine and emulsified isoflurane.(DEI group).All drugs were Ⅳ injection administered for induction,followed by 1.5 MAC(minimal alveolar concentration) of isoflurane to maintain *** temperature(RT),respiratory rate(RR),heart rate(HR),mean arterial pressure(MAP),and ECG were measured at baseline,0,5,10,20,40,and 60 minutes after *** quality of induction and recovery was evaluated for all *** the anaesthetic procedures provided good conditions for induction of *** quality of induction and recovery in the E group was worse than other *** decrease of RR in the E and DE groups was stronger than that in the El and DEI *** dogs in the E group had the most significant prolongation of the Q-T interval and changes in the S-T *** and extension of the S-T segment were noted in the El *** dogs in the DE and DEI groups had fewer changes in the ECG results than those in the E and EI *** addition of dexmedetomidine caused less effect on cardiopulmonary parameters and the ECG results than either etomidate or emulsified isoflurane ***,etomidate or emulsified isoflurane in combination with dexmedetomidine may be useful clinically for the induction of anaesthesia.

Objective:To assess the hepatoprotective effects of flavone on nicotine-induced liver ***:Thirty-six rats were allocated into six groups:the control group,the nicotine group,the flavone alone groups(10 and 25 mg/kg/body weight),and the nicotine groups treated with flavone(10 and 25 mg/kg/body weight).Liver function,oxidative stress,Nrf2 pathway(HO-1,Nrf2,and Keap-1),and inflammatory markers(IL-17,TNF-α,and NF-κB)were ***,a histopathological examination of liver tissues was ***:Nicotine increased liver damage,inflammation,and oxidative ***,flavone suppressed nicotine-induced liver enzymes,oxidative stress,and inflammation,as manifested by increased antioxidants and decreased malondialdehyde level,liver enzymatic activities,and inflammatory ***(10 and 25 mg/kg/body weight)also reduced the level of Keap-1 and increased HO-1 and Nrf2 levels in the liver of nicotine-exposed ***:Flavone has hepatoprotective properties and may slow the progression of liver injury by reducing oxidative stress,liver enzymes,and inflammation possibly via the Nrf2 pathway.

作者用纯化 FE_8和 LA_(15-6)2株抗新城疫病毒(NDV)中和性单抗(MAb_1)分别免疫 BALB/c /小鼠,经杂交瘤技术获得2株抗独特型单克隆抗体(MAb_2),分别命名为AFE_8Id_(16-6)和ALA_(15-6)Id.在血凝抑制阻断试验(BHI)中,2株MAb_2都能阻断FE_8MAb_1抑制NDV血凝活性,BHI效价分别达1:8和1:4.在中和抑制试验(NIT)中,2株MAb_2在 1:10～1:80稀释时均可特异地抑制FE_8MAb_1中和NDV的能力,使部分试验鸡胚死于NDV感染.这些结果不仅表明,2株MAb_2的抗原结合位点是针对FE_8MAb_1独特型决定簇的,而且提示它们的可变区构形和与FE_8MAb_1相结合的NDV抗原决定簇或其邻近结构相似.这2株MAb_2可用于新城疫单克隆抗独特型疫苗的研究。

AIM: To evaluate the effects of positive regulation of recombinant human interleukin 1 receptor antagonist (rhIL-1Ra) on hepatic tissue recovery in acute liver injury in mice induced by carbon tetrachloride (CCl 4 ). METHODS: Acute liver damage was induced by injecting 8-wk-old mice with CCl 4 1 mL/kg (1:3 dilution in corn oil) intraperitoneally (ip). Survival after liver failure was assessed by injecting 8-wk-old mice with a lethal dose of CCl 4 2.6 mL/kg (1:1 dilution in corn oil) ip. Mice were subcutaneously injected with 1 mg/kg recombinant human IL-1Ra twice a day after CCl 4 treatment for 5 d. Serum alanine amino transferase (ALT) and aspartate aminotransferase (AST) levels were determined with a commercial assay kit. Serum IL-1β, IL-1Ra levels were measured by enzyme-linked immunosorbent assay kit. Quantitative real-time polymerase chain reaction was used to determine liver IL-1β, IL-1Ra and IL-6 expression during CCl 4-induced acute liver injury. Liver sections were stained with hematoxylin-eosin. A histology-injury grading system was used to evaluate the degree of necrosis after acute liver injury. Proliferating cell nuclear antigen (PCNA) staining was used to evaluate the role of rhIL-1Ra in promoting hepatocyte proliferation. RESULTS: Quantitative analysis showed a higher level of IL-6 mRNA expression and reduced serum AST and ALT levels in the livers of the rhIL-1Ra-treated group at the early phase of CCl 4-induced acute liver injury. Histological examination indicated a decrease in centrilobular necrotic areas in mice treated with rhIL-1Ra, and a novel role of rhIL-1Ra in promoting hepatocyte proliferation was also supported by an increase of PCNA staining. All these results, accompanied by a strong survival benefit in rhIL-1Ra-treated vs PBS-treated groups, demonstrated that rhIL-1Ra administration ameliorated the histological damage and accelerated the regeneration and recovery process of the liver. 用抗鸡马立克病病毒(MDV)的38Kd磷蛋白的单克隆抗体作萤光抗体试验,直接从用可转染性野生型杆状病毒AcMNPV株DNA与含MDVpp38基因的重组转基因质粒载体DNA共同转染的Sf 9单层细胞培养中筛选到能表达pp38基因产物的重组杆状病毒感染斑,并从保存于琼脂醣凝胶上的复印斑中收复并进一步克隆了该基因重组病毒.该法易于实施,即使未能熟练掌握根据杆状病毒多角体来区别野生型杆状病毒斑与基因重组病毒斑的技术,也可顺利地筛选到能表达目的基因的重组合病毒。
