Machine learning models reveal how biochar amendment affects soil microbial communities

The biochar amendment plays a vital role in maintaining soil health largely due to its effects on soil microbial ***,individual cases and the variability in biochar properties are not sufficient to draw universal *** present study aimed to reveal how the biochar application affects soil microbial *** of 525 ITS and 128816S rRNA sequencing samples from previous studies were reanalyzed and machine learning models were applied to explore the dynamics of soil microbial communities under biochar *** results showed that biochar considerably changed the soil bacterial and fungal community composition and enhanced the relative abundances of Acidobacteriota,Firmicutes,Basidiomycota,and *** enhanced the robustness of the soil microbial community but decreased the interactions between fungi and *** random forest model combined with tenfold cross-validation were used to predict biomarkers of biochar response,indicating that potentially beneficial microbes,such as Gemmatimonadetes,Microtrichales,Candidatus_Kaiserbacteria,and Pyrinomonadales,were enriched in the soil with biochar amendment,which promoted plant growth and soil nutrient *** addition,the biochar amendment enhanced the ability of bacteria to biosynthesize and led to an increase in fungal nutrient patterns,resulting in an increase in the abundance and diversity of saprophytic fungi that enhance soil nutrient *** machine learning model more accurately revealed how biochar affected soil microbial community than previous independent *** study provides a basis for guiding the reasonable use of biochar in agricultural soil and minimizing its negative effects on soil microecosystem.

Colloidal gold immunochromatographic assay(CGIA)is commonly used for the on-site detection ofβ-agonists that are sometimes used illegally as feed additives in swine ***,few studies have evaluated the causes of false-positive results that sometimes occur when applying CGIA in agricultural *** this study,we investigated if this false-positive phenomenon is related to the addition of certain traditional Chinese medicines(TCMs)to swine *** established and verified an extraction method for TCMs,and then applied CGIA to detectβ-agonists in the extracts of 105 TCMs and in the urine of swine dosed with TCMs,*** chromatography-tandem mass spectrometry was used to validate the results of the urine samples tested positive forβ-agonists using *** results were also verified using TCMs and colloidal gold test strips produced by different *** extracts of Citri Reticulatae Pericarpium Viride,Citri Reticulatae Pericarpium,Magnoliae Officinalis Cortex,Chaenomelis Fructus,and Rhodiolae Crenulatae Radix Et Rhizoma were tested positive forβ-***,the addition of Citri Reticulatae Pericarpium Viride and Citri Reticulatae Pericarpium to swine feed resulted in false-positive results forβ-agonists in swine *** results provide a new way to explain false-positive CGIA results and provide valuable information for livestock feeding programs.

Evaluation of genetically modified rice detection methods 2011/884/EU and 2008/289/EC proposed by the European Union

Increases in the number of cases of identified genetically modified （GM） rice contamination can be traced back to the first Rapid Alert System for Food and Feed （RASFF） in 2006. In response to the lack of reliable detection methods, Decision 2011/884/EU proposed that new screening methods replace Decision 2008/289/EC, to identify all possible GM rice products originating in China. However, the synergy brands （SYBR） Green real-time PCR assay proposed by Decision 2011/884/EU has been shown to lack conformity with other TaqMan methods currently in use. To evaluate the specificity and repeatability of the methods recommended in Decision 2011/884/EU and Decision 2008/289/EC, we collected 74 rice products originating from six countries or districts. The 74 rice samples were tested using the Decision 2011/884/EU and Decision 2008/289/ EC methods. The parallel use of different instruments and reagents were used for testing in parallel, and the results were analyzed statistically. To avoid the limitations of specific laboratories, eight GM organism detection laboratories in China participated in a collaborative trial. In our tests, 24.3% （18/74） of the samples tested were positive with the SYBR Green real-time PCR assay using the Decision 2011/884/EU method, but were negative with the TaqMan real-time PCR assay using the Decision 2011/884/EU and Decision 2008/289/EC methods. Sequencing the PCR-amplified CrylA（b/c） genes in three samples （6, 30 and 43） showed that the products consisted of primer dimers rather than the targeted sequence. The combined experimental results showed that testing for the nopaline synthase gene （NOS） of agrobacterium tumefasciens terminator and CrylA（b/c） produced false-positive results when the Decision 2011/884/EU method was used. Because of the high rate of false-positive results, the Decision 2011/884/EU SYBR Green method to detect GM rice requires improvement.

Comprehensive Characterization Model of Integrated Cotton Fiber quality Index

An integrated cotton fiber quality index （ICFQI） model with cotton fiber qualities which can directly express cotton fiber integrated quality and spinning yarn quality was studied. The fiber length, strength, Micronalre （fiber fineness and fiber maturity）, uniformity of fiber length, and short fiber content are the pivotal indexes expressing ICFQI. All of the results above are the basic knowledge to build up the models of ICFQI. According to spinning consistency index （SCI）, spinning strength and spinning yarn integrated quality, ICFQI was the best choice. As the methods of ICFQI had quite a lot of advantages like explicit mechanism, few independent variables. The integrated fiber quality index had a significant positive correlation with yarn strength and spinning consistency, significant negative correlation with yarn evenness and yarn thin places. In additional, the model of the relationship between ICFQI and SCI was established as： SCI=0. 235 6·ICFQI ＋56.153. It was concluded that ICFQI value was the shared reference index for the testing of fiber inspection agency and the selection and distribution of raw cotton bales by textile mills.

Genetic diversity of Listeria monocytogenes isolates from farm products in Shanghai,China

Farm products are considered important vehicles for the transmission of Listeria monocytogenes(***).The typing of *** from farm products contributes to the surveillance and source tracing of the *** this study,77 *** strains from seven farm product categories in Shanghai were analyzed by serological typing,multilocus sequence typing(MLST),pulsed-field gel electrophoresis(PFGE),and whole-genome nucleotide polymorphism(wgSNP).The results showed that the 77 isolates were classified into four serovars(1/2a,1/2c,1/2b,and 4b),and of these,1/2a(n=47)was the most *** sequence types(STs)were generated by MLST with two novel STs(ST1402 and ST1403),and 20 of 77 *** isolates had high genetic identity with previously documented outbreak isolates according to the minimum spanning tree from the MLST ***,34 PFGE patterns(PF1-PF34)were differentiated,and based on a similarity value higher than 80%by the unweighted pair group method dendrogram,the discriminatory index was relatively low(equal to 0.775).Furthermore,14 isolates were chosen and further analyzed by wgsNP based on the previous typing results,which demonstrated that wgSNP and MLST yielded mostly consistent typing results but higher resolution than *** conclusion,77 *** isolates from farm products collected in nine districts in Shanghai were highly genetically diverse,and 20 of these isolates had high relatedness with previously documented outbreak strains *** results indicate a possible cross-contamination risk of *** and a potential public health concern resulting from farm products during the supply chain in Shanghai,China.

Generation and Discharge of Pollutants in Closed Aquaculture

[Objectives]To gain an in-depth understanding of pollutant generation and discharge from closed aquaculture.[Methods]In 2018,the pollutant generation and discharge coefficients of 7 farming modes in 16 farming bases were calculated and analyzed.[Results]A total of 28 pollutant generation coefficients and 28 pollutant discharge coefficients were obtained,and the generation and discharge of pollutants in closed aquaculture were grasped *** with the data of the first census of pollutions sources published by the state in 2008,7 kinds of pollution generation coefficients increased,while 14 kinds decreased;and 4 kinds of pollutant discharge coefficients increased,while 17 kinds decreased.[Conclusions]In recent years,the pollutant generation and discharge coefficients of aquaculture show a decreasing trend.

Spatiotemporal characteristics and Monte Carlo simulationbased human health risk of heavy metals in soils from a typical coal-mining city in eastern China

Mining activities typically discharge considerable amounts of heavy metals into the environment,raising concerns about soil metal pollution,environmental security,and human ***,a systematic regional-scale investigation of soil heavy metal pollution in mining areas is necessary for soil *** this study,5817 soil samples from the Huainan coal mining area collected for studies conducted from 2000 to 2021 were compiled to quantify the pollution level and spatiotemporal variation of heavy metals(Cu,Pb,Zn,Cr,Cd,As,Hg,Ni,and Mn).The associated ecological health risk of heavy metals in soil was assessed using the Hakanson ecological hazard index,Monte Carlo simulation in conjunction with the total hazard quotient,and the hazard *** was the top contaminant,followed by *** terms of spatial distribution,heavy metal contamination was more severe in the eastern area of Fengtai and Datong districts,because these districts of Anhui Province are significant industrial *** addition,the results of the Monte Carlo evaluation of human health risks showed that the total noncarcinogenic risk of heavy metals in soil is below the acceptable level,while the carcinogenic risk was 5.97% for adults and 15.53% for *** accounted for 57.4% of noncarcinogenic risk,Cr contributed 36.1%of carcinogenic *** with adults,children are more vulnerable to the carcinogenic and noncarcinogenic risks posed by heavy metals,with oral consumption being the primary exposure *** research can provide useful details for protecting the environment and managing soil in a coal mining area.

Carbendazim,a widely used fungicide in China,has been found to have reproductive toxicity and mutagenic ***,information on the spatial-temporal variations of carbendazim residues in food in China is ***,we investigated the presence of carbendazim in China's plant-based foods from 2011 to 2020,evaluated the spatial-temporal characteristics,and assessed the associated exposure risks in the Chinese *** results revealed a high detection frequency of carbendazim in fruits(26.4%)and high concentrations in vegetables(~110 mg kg^(-1)),indicating widespread misuse of the *** acute risks of consuming certain vegetables and cereals exceeded the recommended limits by up to 12 and 5 times,*** there has been a decline in carbendazim residue levels in food since the implementation of the Chinese government's action plan for zero growth of pesticide use in 2015,some provinces still exhibited high levels of carbendazim in multiple food categories,which were positively correlated with annual pesticide *** highlight that carbendazim contamination reflects the broader issue of pesticide use in *** emphasizes the need for committed and targeted national policies to reduce carbendazim residues in food and suggests that such measures could also regulate the use of other pesticides,given that pesticide abuse in China is not limited to specific *** call for the re-evaluation of maximum residue limits of carbendazim,particularly in highly consumed foods such as cereals.

Using WGM2012 to Compute Gravity Anomaly Corrections of Leveling Observations in China

Gravity Anomaly Correction(GAC)is a very important term in leveling data *** most cases,it is troublesome for field surveyors to measure gravity when *** this paper,based on the complete Bouguer Gravity Anomaly(BGA)map of WGM2012,the feasibility of replacing in-situ gravity surveying in China is *** leveling application,that is to evaluate the accuracy of WGM2012 in *** WGM2012 is organized with a standard rectangle grid,two interpolation methods,bilinear interpolating and Inverse Distance Weighted(IDW)interpolating,are *** sample areas in China,i.e.,Hanzhong,Chengdu,Linzhi and Shantou,are selected to evaluate the systems bias and precision of *** results show the average system bias of WGM2012 BGA in west China is about-100.1 mGal(1 mGal=10^(-5) m/s^(2))and the standard deviation is about 30.7 *** in Shantou indicate the system bias in plain areas is about-130.4 mGal and standard deviation is about 6.8 *** these experiments means the accuracy of WGM2012 is limited in high mountain areas of western China,but in plain areas,such as Shantou,WGM2012 BGA map is quite good for most leveling applications after calibrating the system bias.

Microbes on fresh apples are closely associated with fruit disease,preservation,and quality *** into the microbial commu-nities on apples from different producing regions could reveal the microbial specificity and help disease prevention and quality *** this paper,the apple surface microbes of 44 samples from two main Chinese apple-producing regions,Bohai Bay(BHB)and the Loess Plateau(LP),were investigated by sequencing fungal internal transcribed spacer and bacterial 16s rRNA hypervariable *** and LP apples con-tained significantly different bacterial and fungal *** apples had a higher fungal diversity than LP apples.A total of 102 different fungal and bacterial taxonomies were obtained between apples from the two regions,in which 24 genera were *** apples had higher phytopathogenic fungal genera,such as Tilletiopsis,Acremonium,Candida,and Phoma,indicating the higher phytopathogenic risks of apples from the humid climate of the BHB *** apples contained more bacterial genera identified as gut microbes,indicating the potential risks of contaminating apples with foodborne pathogens in the arid environment of the LP This study highlighted the environment-oriented mi-crobial specificity on apples from two main apple-producing regions,and provided a basis for further investigation.
