Background： Interleukin-1β（IL-1β） is important mediator of inflammatory-induced suppression of reproductive axis at the hypothalamic level. At the beginning of inflammation, the main source of cytokines in the cerebrospinal fluid（CSF） is peripheral circulation, while over time, cytokines produced in the brain are more important. Melatonin has been shown to decrease pro-inflammatory cytokines concentration in the brain. In ewes, melatonin is used to advance the onset of a breading season. Little is known about CSF concentration of IL-1β in ewes and its correlation with plasma during inflammation as well as melatonin action on the concentration of IL-1β in blood plasma and the CSF, and brain barriers permeability in early stage of lipopolysaccharide（LPS）-induced ***： Systemic inflammation was induced through LPS administration in melatonin-and sham-implanted ewes. Blood and CSF samples were collected before and after LPS administration and IL-1β and albumin concentration were measured. To assess the functions of brain barriers albumin quotient（QAlb） was *** of IL-1β（Il1B） and its receptor type Ⅰ（Il1r1） and type Ⅱ（Il1r2） and matrix metalloproteinase（Mmp） 3 and 9 was evaluated in the choroid plexus（CP）.Results： Before LPS administration, IL-1β was on the level of 62.0 ± 29.7 pg/mL and 66.4 ± 32.1 pg/mL in plasma and 26.2 ± 5.4 pg/mL and 21.3 ± 8.7 pg/mL in the CSF in sham-and melatonin-implanted group, *** LPS it increased to 159.3 ± 53.1 pg/mL and 197.8 ± 42.8 pg/mL in plasma and 129.8 ± 54.2 pg/mL and139.6 ± 51.5 pg/mL in the CSF. Objective:To understand the level of progesterone (P4) in different quality of buffalo cumulus oocyte complexes (COCs) and further to evaluate the effect of exogenous P4 supplementation on maturation and subsequent developmental ability of poor quality brilliant cresyl blue (BCB-)***: Progesterone secreted by different quality of buffalo oocytes was estimated by enzyme linked immunosorbent assay and the concentration differences were translated into P4 doses to be incorporated in the maturation medium of BCB-ve COCs followed by expression analysis of genes involved in the cumulus expansion, extracellular matrix disintegration and progesterone receptor signalling. In addition, the study also evaluated the effect of exogenous P4 on sperm-cumulus interaction. Results: More than 10-fold up-regulated expression of progesterone receptor in P4 supplemented oocytes signified that P4 might be acting predominantly through this receptor. Also, exogenous P4 supplementation had significant effect on transcatheter arterial chemoembolization protease regulated by P4-progesterone receptor pathway which in turn had an important role in extracellular matrix disintegration. On the contrary, cumulus expansion genesHAS2,TNFAIP6,AREG were not altered upon P4 supplementation. Also, it was observed that P4 addition did facilitate passage of significantly more number of spermatozoa through P4 treated cumulus cells. Further, incorporation of different doses of P4 did not improve significantly the cleavage and blastocyst rates of BCB-ve ***: Different qualities of buffalo COCs secrete substantially diverse levels of P4, and its supplementation has a role in oocyte maturation via modulation of cumulus characteristics but perhaps not fertilization.

DNA Methylation Pattern in Pronuclear-Stage Mouse Embryos:Effect of Oocyte Vitrification

This study was conducted to investi- gate the pattern of DNA methylation in pronuclearstage mouse embryos derived from vitrified-warmed oocytes. Mouse oocytes at metaphase II （MII） stage of meiosis were allocated randomly into three groups： 1） untreated （control）; 2 ） exposed to vitrification solution without being plunged into liquid nitrogen （toxicity）; and 3 ） vitrified by open-pulled straw （OPS） method （vitrification）. Oocytes were fertilized in vitro （IVF）. The level of DNA methylation was examined at 8 hpf （hours post-fertilization） by immunofluorescence using an anti-5-methylcytosine （5-MeC） monoclonal antibody and fluorescein isothiocyanate （FITC）-conjugated goat anti-mouse IgG. After IVF, rates of 2-cell embryos （ 51.39% ） and blastocysts （35.82%） for vitrified-warmed oocytes were lower （P〈0.01） than that for control （70.83%, 47.82% ） or vitrification solution treated （64.80%, 46.29% ） oocytes. At 8 hpf, there were more （P 〈 0.05） pronuclear-stage oocytes in which syngamy of pronuclei had not occurred in the vitrifica- tion group. In addition, 5-MeC fluorescent intensities for the female pronucleus and zygote were lower in the vitrification group （P 〈0.01 ） compared to pronuclear-stage embryos in the control and toxicity groups. In conclusion, oocyte vitrification causes a reduction in global genomic methylation in the female pronucleus and zygote, resulting in delayed fusion of pronuclei and compromising the developmental potential of mouse zygotes and embryos.

Understanding the evolutionary history and adaptive process depends on the knowledge that we can acquire from both ancient and modern genomic *** the availability of a deluge of whole-genome sequencing data from ancient and modern goat samples,a user-friendly database making efficient reuse of these important resources is ***,we use the genomes of 208 modern domestic goats,24 bezoars,46 wild ibexes,and 82 ancient goats to present a comprehensive goat genome variation database(GGVD).GGVD hosts a total of~41.44 million SNPs,~5.14 million indels,6,193 selected loci,and 112 introgression *** can freely visualize the frequency of genomic variations in geographical maps,selective sweeps in interactive tables,Manhattan plots,or line charts,as well as the heatmap patterns of the SNP *** data can be shown in haplotypes to track the state of genetic variants of selection and introgression events in the early,middle,and late *** facilitating access to sequence features,the UCSC Genome Browser,BLAT,BLAST,Lift Over,and pcadapt are also integrated into *** will be a convenient tool for population genetic studies and molecular marker designing in goat breeding programs,and it is publicly available at http://***/goat Var.

In this study, we determined species-specific variations by analyzing the mitochondrial 12S rRNA gene sequence variation （-440 bp） in 17 newly obtained sequences and 90 published cattle, yak, buffalo, goat, and pig sequences, which represent 62 breeds and 17 geo- graphic regions. Based on the defined species-specific variations, two endonucleases, Alu I and Bfa I, were selected for species authentication using raw meat/tissue samples and the PCR-RFLP method. goat and pig were identified using the Alu I enzyme, while cattle, yak, and buffalo were identified by digestion with Bfa I. Our approach had relatively high detection sensitivity of cattle DNA in mixed cattle and yak products, with the lowest detectable threshold equaling 20% of cattle DNA in a mixed cattle/yak sample. This method was successfully used to type commercial beef jerky products, which were produced by different companies utilizing various processing technologies. Our results show that several yak jerky products might be implicated in commercial fraud by using cattle meat instead of yak meat.
