Isolation and characterisation of human gingival margin-derived STRO-1/MACS^+ and MACS^-cell populations

Recently,gingival margin-derived stem/progenitor cells isolated via STRO-1/magnetic activated cell sorting（MACS） showed remarkable periodontal regenerative potential in *** a second-stage investigation,the present study＇s aim was to perform in vitro characterisation and comparison of the stem/progenitor cell characteristics of sorted STRO-1-positive（MACS~＋） and STRO-1-negative（MACS^-） cell populations from the human free gingival *** were isolated from the free gingiva using a minimally invasive technique and were magnetically sorted using anti-STRO-1 ***,the MACS~＋ and MACS^- cell fractions were characterized by flow cytometry for expression of CD14,CD34,CD45,CD73,CD90,CD105,CD146/MUC18 and ***-forming unit（CFU） and multilineage differentiation potential were assayed for both cell *** marker expression was examined using real-time polymerase chain reaction（PCR）.MACS~＋ and MACS- cell fractions showed plastic ***~＋ cells,in contrast to MACS- cells,showed all of the predefined mesenchymal stem/progenitor cell characteristics and a significantly higher number of CFUs（P〈0.01）.More than 95%of MACS~＋ cells expressed CD105,CD90 and CD73;lacked the haematopoietic markers CD45,CD34 and CD14,and expressed STRO-1 and CD146/***- cells showed a different surface marker expression profile,with almost no expression of CD14 or STRO-1,and more than 95%of these cells expressed CD73,CD90 and CD146/MUC18,as well as the haematopoietic markers CD34 and CD45 and ***~＋ cells could be differentiated along osteoblastic,adipocytic and chondroblastic *** contrast,MACS- cells demonstrated slight osteogenic *** MACS~＋ cells showed significantly higher expression of collagen I（P〈0.05） and collagen III（P〈0.01）,whereas MACS^- cells demonstrated higher expression of osteonectin（P〈0.05;MannWhitney）.The present study is the first to compare gingival MACS~＋ and MACS- cell populations demo

Investigation of Color Change of Different Restoration Thickness, Background Color and Resin Cement Shade on CAD/CAM Glass Ceramic Materials

The aim of this study is to investigate the color change of different restoration thicknesses, backgrounds and resin cement colors on lithium disilicate and zirconium reinforced lithium silicate materials in vitro. In this study, IPS emax CAD (LT C14) and Celtra Duo (LT C14) are used as full ceramic materials, and Variolink Esthetic LC (warm, neutral) used as resin cement and Tokuyama Estelite Sigma Quick (A3, A2) is used as composite materials. A total of 160 samples in the form of 40 pieces of 5 × 5 0.4 mm thick 40 pieces of 5 × 5 0.6 mm thick square discs from each of the all-ceramic materials in block form were obtained using a water jet device (DWJ1525-FA;Dardi International Corporation, Nanjing, China). Glass ceramic samples produced in 2 different thicknesses were cemented on 2 different backgrounds with 2 different resin types of cement. Color measurements of the samples before and after cementation were performed on a grey background with spectrophotometer Vita EasyShade V (Vita Zahnfabrik, Bad Sackingen, Germany) and color parameters (L*, a*, b*, ΔE) were calculated according to the CIE Lab (Commission Internationale de L’Eclairage) system. Average values for each group (ΔE) were not affected by ceramic type, material thickness, background color, resin cement color, and the interaction of these four variables (p > 0.05). When the triple interactions between the groups were examined, there were no statistically significant differences (p > 0.05). In the evaluation of pairwise interactions between two groups (material type-material thickness, material type-background color, and thickness of material-background interactions) statistically significant differences (p Implications: The material type, thickness, background and cement color used did not cause any statistically significant color change in lithium disilicate and zirconium-reinforced lithium silicate glass ceramic materials (p > 0.05).

Applied Anatomic Site Study of Palatal Anchorage Implants Using Cone Beam Computed Tomography

Aim The purpose of this study was to conduct quantitative research on bone height and bone mineral density of palatal implant sites for implantation, and to provide reference sites for safe and stable palatal implants. Methodology Three-dimensional reformatting images were reconstructed by cone beam computed tomography （CBCT） in 34 patients, aged 18 to 35 years, using EZ Implant software. Bone height was measured at 20 sites of interest on the palate. Bone mineral density was measured at the 10 sites with the highest implantation rate, classified using K-mean cluster analysis based on bone height and bone mineral density. Results According to the cluster analysis, 10 sites were classified into three clusters. Significant differences in bone height and bone mineral density were detected between these three clusters （P〈0.05）. The greatest bone height was obtained in cluster 2, followed by cluster 1 and cluster 3. The highest bone mineral density was found in cluster 3, followed by cluster 1 and cluster 2. Conclusion CBCT plays an important role in pre-surgical treatment planning. CBCT is helpful in identifying safe and stable implantation sites for palatal anchorage.

Three-dimensional FEM analysis of stress distribution in dynamic maxillary canine movement

An alteration in stress/strain distribution within periodontal ligament (PDL) occurs when the tooth is mechanical stimulated in orthodontic treatment. The majority of previous finite element studies were confined to a static evaluation on initial loading status, and a dynamic long-term analysis was rarely performed in orthodontics. The present study sought to analyze the stress distribution patterns within tooth root, PDL, and alveolar bone during dynamic canine tipping and bodily movement with three-dimensional finite element method in four consecutive therapeutic weeks. We observed that the highest stress was on root surface, secondly alveolar bone, and finally, PDL for either tipping or bodily movement. Throughout the PDL and root surface, the highest stress was always around the cervical area in both patterns of movement through four phases. Moreover, a lower and more uniform stress distribution was produced during bodily movement compared with tipping movement. These results indicated that canine translation might be healthier for periodontal tissue than tipping movement; the cervical region bore a greater load and might also be a susceptible area for tissue damage (except for the apex) regardless of movement pattern.

H2 Stimulated Raman Scattering in a Multi-pass Cell with a Herriott Configuration

Stimulated raman scattering （SRS） is an effective method for expanding the spectral range of high power lasers, especially in the regime of near IR and middle IR. We report the SRS of high pressure H2 with a multiple-pass cell configuration. The SRS with the multiple-pass cell configuration is found to be very efficient for reduction of threshold of the first Stokes （S1）. Due to the coherent SRS （CSRS） process, the multiple-pass cell configuration is more effective for reduction of the threshold for the second Stokes （S2） SRS and for increasing the conversion efficiency of S2. This contributes to the relatively low conversion efficiency of S1 for the multiple-pass cell configuration. Multiple-pass cell SRS is also found to be very effective for improving the beam quality and the stability of S1.

A robust methodology for the quantitative assessmentof the rat jawbone microstructure

Micro-computed tomography can be applied for the assessment of the micro-architectural characteristics of the cortical and trabecular bones in either physiological or disease conditions. However, reports often lack a detailed description of the methodological steps used to analyse these images, such as the volumes of interest, the algorithms used for image filtration, the approach used for image segmentation, and the bone parameters quantified, thereby making it difficult to compare or reproduce the studies. This study addresses this critical need and aims to provide standardized assessment and consistent parameter reporting related to quantitative jawbone image analysis. Various regions of the rat jawbones were screened for their potential for standardized micro-computed tomography analysis. Furthermore, the volumes of interest that were anticipated to be most susceptible to bone structural changes in response to experimental interventions were defined. In the mandible, two volumes of interest were selected, namely, the condyle and the trabecular bone surrounding the three molars. In the maxilla, the maxillary tuberosity region and the inter-radicular septum of the second molar were considered as volumes of interest. The presented protocol provides a standardized and reproducible methodology for the analysis of relevant jawbone volumes of interest and is intended to ensure global, accurate, and consistent reporting of its morphometry. Furthermore, the proposed methodology has potential, as a variety of rodent animal models would benefit from its implementation.

Changes in sleep parameters following biomimetic oral appliance therapy: A case report

BACKGROUND Mandibular advancement devices(MADs)are used to treat mild to moderate obstructive sleep apnea(OSA),but there is a risk that the underlying condition can worsen in the ***,this case report is based on biomimetic oral appliance therapy as an alternative to MADs,which was found to be beneficial in the treatment of a case with severe *** SUMMARY An overnight sleep study was undertaken in a 50-year-old male with excessive daytime sleepiness that lead to a diagnosis of severe OSA as the apnea-hypopnea index(AHI)was found to be 32.8/*** the patient was unable to comply with continuous positive airway pressure therapy and declined surgical intervention,treatment with a MAD was *** 10 years later,another sleep study was performed with no MAD in the mouth,which revealed an AHI of 67.9/*** view of the deterioration in sleep quality,the patient sought alternative treatment and elected on biomimetic oral appliance therapy,using a mandibular repositioning nighttime appliance(mRNA appliance®,Vivos Therapeutics,Inc.,United States).After 10 mo,another sleep study was performed with no device in the patient’s mouth,which revealed an AHI of 11.8/h,a mean oxygen saturation of 94%and a mean oxygen desaturation index of 5.3%while *** analysis of the pre-and post-treatment study models of the upper jaw showed localized size increases of 15%-17%in the premolar regions and 15%-23%in the molar *** In adults with severe OSA that are unable to accept continuous positive airway pressure or surgical treatment,biomimetic oral appliance therapy may be preferable over MADs since biomimetic oral appliance therapy may be able to prevent worsening of sleep parameters by remodeling the nasomaxillary ***-term follow up studies are required to verify these novel findings.

Extracellular matrix remodelling in dental pulp tissue of carious human teeth through the prism of single-cell RNA sequencing

Introduction: Oral health management is often difficult in patients after surgical tumor resection and/or radiotherapy due to defects, bulky flaps, limitation of tongue movement, and trismus. In addition, patients who have undergone radiotherapy have serious disabilities such as oral mucositis, mucosal weakening, soft tissue fibrosis, salivary gland disorder, and osteoradionecrosis. dental hygienists must understand the characteristics of patients after surgical tumor resection and/or radiotherapy. In this report, the oral health management of three patients after maxillectomy, mandibulectomy, and radiotherapy was shown. Case Description: Case 1: A 53-year-old male patient visited our clinic after chemotherapy, radiotherapy, and partial maxillectomy for cancer of the right maxillary gingiva. Case 2: A 65-year-old male patient visited our clinic after radiotherapy for a right-sided tongue cancer, partial mandibulectomy of the right tongue to the oropharynx, a right-sided segmental mandibulectomy and a speech aid and a mandibular prosthesis were provided. Case 3: A 36-year-old female visited our clinic for radiotherapy for left-sided tongue cancer. dental hygienists provided oral health management to the patients. In oral health management, it was suggested that intervention before cancer treatment, oral hygiene instructions tailored to patients’ symptoms, and frequent professional care are important. Conclusion: Oral health management by dental hygienists is extremely important for patients undergoing maxillofacial cancer treatment to maintain their quality of life in the long term.
