Characterization of DNA polymerase δ from deep-sea hydrothermal vent shrimp Rimicaris exoculata

DNA polymerase δ(Polδ)plays a crucial and versatile role in DNA replication and DNA repair *** shrimp Rimicaris exoculata is the primary megafaunal community living in hydrothermal *** this study,the Polδfrom shrimp Rimicaris exoculata was cloned,expressed and *** results showed that the Polδcatalytic subunit(POLD1),852 amino acids in length,shared high homology with crayfish Procambarus clarkii and shrimp Oratosquilla *** recombinant POLD1 expressed in Escherichia coli showed that the enzyme was active in a range of 20℃ to 40℃ with an optimum temperature at 25℃ and in a wide range of p H with an optimum at pH *** activities of POLD1 were significantly enhanced in the presence of Triton-X 100,Tween 20 and Mn^(2+).The K_(m)(dNTP)value of POLD1 was 4.7μmol/*** present study would be helpful to reveal the characterization of Polδof deep-sea vent animals.

Carbon dots(CDs)with intriguing fluorescent property,good biocompatibility,high stability,easy interaction with substrates,are burgeoning carbon nanoparticles with large potential in various *** CDs into the polymer matrix is becoming a popular strategy to endow the complex with new ***,the green-synthesized CDs was integrated into the mixture of gelatin(derived from waste fish scale)and chitosan,and a multifunctional bio-nanocomposite(defined as Gelatin/Chitosan/CDs)film was developed,which showed the excellent antibacterial,antioxidant,pH-sensitivity,UV shielding,and blue-emission *** effects of different concentrations of CDs on the physical,mechanical,structural,and functional activity of bio-nanocomposite film were *** with the Gelatin/Chitosan film,the Gelatin/Chitosan/CDs film with an optimum addition of 20%CDs showed the enhanced antibacterial,antioxidant as well as UV shielding *** importantly,it was used as an effective packaging material for fish meat preservation,reducing the loss of nutritional quality consumption,extending the shelf life of ***,the bio-nanocomposite films also possessed the anti-counterfeiting and pH-responsive properties due to the strong fluorescent emission of CDs,and had the great potential in developing the intelligent packaging *** work shed new light on the new application of CDs and the synthesis of bio-nanocomposite film in food industry.

The optimization of fermentation medium is important for synthetic biological secondary metabolite *** effect of rotation speed,inoculum amount,and medium supplements on the cell growth and Lycogen^TM secretion of photobacterium Rhodobacter sphaeroides WL-APD911 was *** results reveal that a higher rotational speed exhibit a higher cell density,and the increasing in the amount of inoculum amount show a slight augment on the growth of *** *** the case of nitrogen sources adding,Lycogen^TM production was achieved with a 0.5 mM l-lysine ***,the attention of Tween 80 presented a tremendous increase in the secondary *** surface methodology(RSM)exhibited the optimization of medium supplements for Lycogen^TM invention is accomplished at molasses concentration of 10 g/L,yeast extract concentration of 40 g/L,0.3%Tween 80 and NaCl concentration of 5 g/L,***,the batch fermentation is carried out in both 5 L and 20 L fermentors to study the scale-up process factors to be *** a 20 L fermentor,Lycogen^TM yields under the optimal culture condition are over 2 times than in the shake *** present results provide the Lycogen^TM optimal culture mediums,scale-up procedures and efficient extractions from *** WL-APD911.
