Objective Lindqvist-type polyoxometalates(POMs)exhibit potential antitumor *** study aimed to examine the effects of Lindqvist-type POMs against breast cancer and the underlying *** Using different cancer cell lines,the present study evaluated the antitumor activities of POM analogues that were modified at the body skeleton based on molybdenum-vanadium-centered negative oxygen ion polycondensations with different side *** colony formation assay,autophagy detection,mitochondrial observation,qrT-PCr,Western blotting,and animal model were used to evaluate the antitumor activities of POMs against breast cancer cells and the related *** MO-4,a Lindqvist-type POM linking a proline at its side strain,was selected for subsequent experiments due to its low half maximal inhibitory concentration in the inhibition of proliferation of breast cancer *** was found that MO-4 induced the apoptosis of multiple types of breast cancer ***,MO-4 activated intracellular mitophagy by elevating mitochondrial reactive oxygen species(rOS)levels and resulting in *** vivo,breast tumor growth and distant metastasis were significantly reduced following MO-4 *** Collectively,the results of the present study demonstrated that the novel Lindqvist-type POM MO-4 may exhibit potential in the treatment of breast cancer.

Mitogen-activated protein kinase(MAPK)cascades play important roles in disease resistance in model plant ***,the functions of MAPK signaling pathways in crop disease resistance are largely *** we report the function of HvMKK1-HvMPK4-HvWrKY1 module in barley immune ***4 is identified to play a negative role in barley immune response against Bgh,as virus-induced gene silencing of HvMPK4 results in enhanced disease resistance whilst stably overexpressing HvMPK4 leads to super-susceptibility to Bgh ***,the barley MAPK kinase HvMKK1 is found to specifically interact with HvMPK4,and the activated HvMKK1^(dd) variant specifically phosphorylates HvMPK4 in ***,the transcription factor HvWrKY1 is identified to be a downstream target of HvMPK4 and phosphorylated by HvMPK4 in vitro in the presence of HvMKK1^(dd).Phosphorylation assay coupled with mutagenesis analyses identifies S122,T284,and S347 in HvWrKY1 as the major residues phosphorylated by ***1 is phosphorylated in barley at the early stages of Bgh infection,which enhances its suppression on barley immunity likely due to enhanced dNA-binding and transcriptional repression *** data suggest that the HvMKK1-HvMPK4 kinase pair acts upstream of HvWrKY1 to negatively regulate barley immunity against powdery mildew.

Ginger polysaccharide UGP1 suppressed human colon cancer growth via p53,Bax/Bcl-2,caspase-3 pathways and immunomodulation

In previous study,we got a purified ginger polysaccharide UGP1 and verified its significant antitumor activities on colon cancer HCT116 *** this article,we aimed to illustrate the underlying mechanism of UGP1 exerted antitumor activities on colon cancer by using in vitro cell models and in vivo animal *** results demonstrated that UGP1 could induce S-phase cell cycle arrest,up-regulate the expression of Bax and p53,down-regulate the expression of Bcl-2,and activate the downstream protein caspase-9 and caspase-3,which was related to intrinsic apoptosis pathway on HCT116 ***,UGP1 significantly stimulated rAW264.7 cell proliferation and secretion ***,UGP1 inhibited tumor proliferation on tumor-bearing mice,increased the expression of p53 and the ratio of Bax/Bcl-2,enhanced the secretion of pro-inflammatory cytokines TNF-α,IL-2,IL-6 and decreased the secretion of pro-tumor cytokines TGF-βand b FGF in *** conclusion,it indicated that the UGP 1 could sup press human colon cancer growth by inducing apoptosis via the regulation of p53,caspase-3,and Bax/Bcl-2 ratio-dependent pathway and regulating immune system *** s investigation provided basic theoretical mechanism of ginger polysaccharideexerted antitumor activities,and contributed to develop a possible functional food or adjuvant agent for prevention or treatment of colon cancer.

Strain H_2-419-4 of Haematococcus pluvialis induced by ethyl methanesulphonate and ultraviolet radiation

Two strains H2-410 and H2-419 were obtained from the chemically mutated survivors of wild Haematococcus pluvialis 2 by using ethyl methanesulphonate (EMS). Strains H2-410 and H2-419 showed a fast cell growth with 13% and 20% increase in biomass compared to wild type, respectively. Then H2-419-4, a fast cell growth and high astaxanthin accumulation strain, was obtained by exposing the strain H2-419 to ultraviolet radiation (UV) further. The total biomass, the astaxanthin content per cell, astaxanthin production of H2-419-4 showed 68%, 28%, and 120% increase compared to wild H. pluvialis 2, respectively. HPLC (High Performance Liquid Chromatography) data showed also an obvious proportional variation of different carotenoid compositions in the extracts of H2-419-4 and the wild type, although no peak of carotenoids appeared or disappeared. Therefore, the main compositions in strain H2-419-4, like its wild one, were free of astaxanthin, monoester, and diester of astaxanthin. The asexual reproduction in survivors after exposed to UV was not synchronous, and different from the normal synchronous asexual reproduction as the mother cells were motile instead of non-motile. Interestingly, some survivors from UV irradiation produced many mini-spores (or gamete?), the spores moved away from the mother cell gradually 4 or 5 days later. This is quite similar to sexual reproduction described by Elliot in 1934. However, whether this was sexual reproduction remains questionable, as no mating process has been observed.

Objective: The human cluster of differentiation(Cd)300A, a type-I transmembrane protein with immunoreceptor tyrosine-based inhibitory motifs, was investigated as a potential immune checkpoint for human natural killer(NK) cells targeting hematologic malignancies(HMs).Methods: We implemented a stimulation system involving the Cd300A ligand, phosphatidylserine(PS), exposed to the outer surface of malignant cells. Additionally, we utilized Cd300A overexpression, a Cd300A blocking system, and a xenotransplantation model to evaluate the impact of Cd300A on NK cell efficacy against HMs in in vitro and in vivo settings. Furthermore, we explored the association between Cd300A and HM progression in ***: Our findings indicated that PS hampers the function of NK cells. Increased Cd300A expression inhibited HM lysis by NK cells. Cd300A overexpression shortened the survival of HM-xenografted mice by impairing transplanted NK cells. Blocking PS–Cd300A signals with antibodies significantly amplified the expression of lysis function-related proteins and effector cytokines in NK cells, thereby augmenting the ability to lyse HMs. Clinically, heightened Cd300A expression correlated with shorter survival and an “exhausted” phenotype of intratumoral NK cells in patients with HMs or solid ***: These results propose Cd300A as a potential target for invigorating NK cell-based treatments against HMs.

Supporting IrO_(x) nanosheets on hollow TiO_(2) for highly efficient acidic water splitting

The efficiency of proton exchange membrane water electrolysis(PEM-WE)for hydrogen production is heavily dependent on the noble metal iridium-based ***,the scarcity of iridium limits the large-scale application of *** address this issue,it is promising to select an appropriate support because it not only enhances the utilization efficiency of noble metals but also improves mass transport under high ***,we supported amorphous IrO_(x) nanosheets onto the hollow TiO_(2) sphere(denoted as IrO_(x)),which demonstrated excellent performance in acidic electrolytic water ***,the annealed IrO_(x)catalyst at 150℃in air exhibited a mass activity of 1347.5 A·gIr^(−1),which is much higher than that of commercial IrO_(2) of 12.33 A·gIr^(−1) at the overpotential of 300 mV for oxygen evolution reaction(OEr).Meanwhile,the annealed IrO_(x) exhibited good stability for 600 h operating at 10 mA·cm^(−2).Moreover,when using IrO_(x) and annealed IrO_(x) catalysts for water splitting,a cell voltage as low as 1.485 V can be achieved at 10 mA·cm^(−2).The cell can continuously operate for 200 h with negligible degradation of performance.

Existing research on fault diagnosis for polymer electrolyte membrane fuel cells(PEMFC)has advanced significantly,yet performance is hindered by variations in data distributions and the requirement for extensive fault *** this study,a cross-domain adaptive health diagnosis method for PEMFC is proposed,integrating the digital twin model and transfer convolutional diagnosis model.A physical-based high-fidelity digital twin model is developed to obtain diverse and high-quality datasets for training diagnosis *** extract long-term time series features from the data,a temporal convolutional network(TCN)is proposed as a pre-trained diagnosis model for the source domain,with feature extraction layers that can be reused to the transfer learning *** is demonstrated that the proposed pre-trained model can hold the ability to accurately diagnose the various fuel cell faults,including pressure,drying,flow,and flooding faults,with 99.92%accuracy,through the effective capture of the long-term dependencies in time series ***,a domain adaptive transfer convolutional network(dATCN)is established to improve the diagnosis accuracy across diverse fuel cells by learning domain-invariant *** results show that the dATCN model,tested on three different target domain devices with adversarial training using only 10%normal data,can achieve an average accuracy of 98.5%(30%improved over traditional diagnosis models).This proposed method provides an effective solution for accurate cross-domain diagnosis of PEMFC devices,significantly reducing the reliance on extensive fault data.
