Rapid monitoring of indoor airborne influenza and coronavirus with high air flowrate electrostatic sampling and PCR analysis

The World Health Organization has raised concerns about the possibility of airborne transmission in enclosed and poorly ventilated ***,rapid monitoring of airborne viruses is necessary in multi-use facilities with dense ***,an electrostatic air sampler(250 L/min)was developed in this work to obtain indoor viral aerosol samples for analysis via the Polymerase Chain Reaction(PCR).Aerosol tests with H1N1 and HCoV-229E were performed to evaluate the sample collection *** analysis,along with another aerosol test,was conducted to evaluate the recovery of the virus particles collected by the *** laboratory tests,our electrostatic sampler obtained viral samples that were detectable by PCR under the simulated viral pandemic scenario(3000 RNA copies per cubic meter of air)within 40 *** resulting cycle threshold(C)values were 35.07 and 37.1 for H1N1 and HCoV-229E,*** the performance evaluation in the laboratory,field tests were conducted in a university classroom from October 28 to December 2,*** A and HCoV-229E were detected in two air samples,and the corresponding C,values were 35.3 and *** PCR results are similar to those obtained from laboratory tests,considering the simulatedviralpandemic scenario.

Objective:To conduct a comparative analysis of the VP4 gene sequences of Indian wild type (06361,0613158,061060 and 0715880) and cell culture adapted(06361-CA,0613158-CA.061060- CA and 0715880-CA) G1P[8]rotavirus ***:Full-length VP4 genes of each of the four wild type G1P[8]rotavirus strains and their cell culture adapted counterparts displaying consistent cytopathic effect were subjected to RT-PCR amplification and nucleotide sequencing. Results:All four cell culture adapted G1P[8]rotavirus strains showed nucleotide and amino acid substitutions in the VP4 gene as compared to their wild type *** number of substitutions however,varied from 1-64 and 1-13 *** substitutions were distributed in both VP5* and VP8* subunits of VP4 gene respectively of permeabilizalion and hemagglutinaling activity. The presence of unique amino acid substitutions was identified in two of the four wild type(V377G. S387N in 061060 and 1644L in 0715880) and all four cell culture adapted(A46V in 0613158-CA. T60R in 06361-CA,L237V.G389V and Q480H in 061060-CA and S615G and T625P in 0715880-CA) strains for the first time in the VP4 gene of P[8]*** acid substitutions generated increase in the hydrophilicity in the cell culture adapted rotavirus strains as compared to their corresponding wild type ***:Amino acid substitutions detected in the VP4 genes of G1P[8]rotavirus strains from this study together with those from other studies highlight occurrence of only strain and/or host specific substitutions during cell culture adaptation. Further evaluation of such substitutions for their role in attenuation,immunogenicity and conformation is needed for the development of newer rolavirus vaccines.

Molecular characterization of VP4,VP6,VP7 and NSP4 genes of group B rotavirus strains from outbreaks of gastroenteritis

Objective:To characterize VP4,VP6,VP7 and NSP4 genes of representative GBR strains(NIV- ***-04622 and NIV-094456) delected as the major eliolngic agenl in the outbreaks of gastroenteritis in western ***:Fecal specimens collected during the outbreaks of gastroenteritis were processed for RNA ***-PCR using GBR ***7 and NSP4 gene specific primers,nucleotide sequencing of the amplicons and phylogenetic analysis of the ***:Phylogenetic analysis of all of the ***7 and NSP4 gene sequences revealed clustering of GBR strains in Indian-Bangladeshi lineage of genotype G2 with 95.8%- 99.4%nucleotide and 97.3%-100.0%amino acid ***,all three strains showed the presence of unique amino acid substitutions in the VP4 protein suggesting alteration in the antigenicity of outbreak strains of *** VP8* and VP5* regions of VP4 proteins showed respectively 0.5%-6.3%and 0.2%-1.1%amino acid divergence from human GBR strains of Indian-Bangladeshi ***:These data confirm the reported variability of VP8* region and suggest the possible role of this region in the perpetuation of GBR infections in the *** is the first study to document the phylogenetic relationship of VP4,***7 and NSP4 genes of GBR strains detected in the outbreaks of gastroenteritis from India with the CBR strains from other parts of world.

Senecavirus A(SVA)has a positive-sense,single-stranded RNA *** 5´untranslated region harbors an internal ribosome entry site(IRES),comprising 10 larger or smaller stem-loop structures(including a pseudoknot)that have been demonstrated to be well ***,it is still unclear whether each stem-loop subdomain,such as a single stem or loop,is also highly *** clarify this issue in the present study,a set of 29 SVA cDNA clones were constructed by site-directed mutagenesis(SDM)on the *** SDM-modified scenarios included:(1)stem-formed complementary sequences exchanging with each other;(2)loop transversion;(3)loop transition;and(4)point *** cDNA clones were separately transfected into cells for rescuing viable viruses,whereas only four SVAs of interest could be recovered,and were genetically stable during 20 *** progeny grew significantly slower than the other three *** dual-luciferase reporter assay showed that none of the SDM-modified IRESes significantly inhibited the IRES *** previous study indicated that a single motif from any of the ten stem structures,if completely mutated,would cause the failure of virus ***,our present study revealed three stem structures,whose individual complementary sequences could exchange with each other to rescue sequence-modifying ***,one apical loop was demonstrated to have the ability to tolerate its own full-length transition,also having no impact on the recovery of sequence-modifying *** present study suggested that not every stem-loop structure was strictly conserved in its conformation,while the full-length IRES itself was well *** provides a new research direction on interaction between the IRES and many factors.

Jingmenviruses are a group of flavi-like viruses with segmented genome and have been found in various types of hosts,including humans,cattle,monkeys,bats,rodents,sheep,ticks,mosquitoes and ***,including the Jingmen tick virus(JMTV)and Alongshan virus(ALSV),have been associated with febrile illness and flu-like symptoms in *** polymerase plays critical roles in genome replication and transcription and is an ideal target for antiviral ***,we determined the crystal structures of RNA-dependent RNA polymerase(RdRp)domains of JMTV and ALSV at 2.6Åand 3.2Åresolutions,*** overall structures of JMTV and ALSV RdRp domains are similar to those from the typical unsegmented viruses in Flaviviridae family,especially the Flavivirus *** and ALSV RdRps can be divided into three subdomains and the catalytical Motif A-G are conserved like the typical flaviviruses,whereas the zinc-binding pockets are absent from the JMTV and ALSV *** 50-ends of jingmenvirus genomes are varied in length and sequence,and a highly conserved 8-nucleotide element located on the tip of stem loop A was identified and shown to be required for binding with RdRp and performing de novo replication *** findings provide important structural insights into RdRp of segmented flavivirus and reveal the key region of virus genome responsible for replication initiation,which would promote molecular understanding of segmented flavivirus replication and the structure-based design of antiviral drugs against flaviviruses.

One-hundred years have passed since the original description of the commonly described phenomenon of persistent abdominal symptoms being triggered by an acute enteric infection. This first account was generated out of astute observations by Sir Arthur Hurst in World War I. Additional descriptions followed from military and non-military practitioners adding the evidence which has transitioned this recognized condition from association to causation. While mechanistic understanding is an area of active pursuit, this historical accounting of a centuries progress highlights important advances and contributions of military medicine and scientists to advances benefiting global populations.

Bioaccumulation of selenium and arsenic resulting from geogenic activities and aggravated by anthropogenic actions has now become a serious environmental issue. Different parts of the world are experiencing issues of selenium and arsenic contamination, especially in soil and ground water. The present selenium and arsenic contamination scenario has been well represented in contemporary literature, and there are an array of remediation techniques targeting the decontamination of environmental contamination. The use of microbe-mediated remediation of selenium and arsenic contamination is gaining scientific attention for sustainable environmental bioremediation. The present review elucidates the different aspects of selenium and arsenic contamination, bioaccumulation, and bioremediation with implications for successful decontamination of these heavy metals.

Oral delivery of specific IgY has been reported to be beneficial against rotavirus infection. However, the production of IgYs against globally prevalent human rotavirus (HRV) serotypes and their evaluation detailing the influence on the virological/histopathological consequences have not been reported to date. In the present study, anti-HRVIgY was generated in the eggs of specific pathogen free hens immunized with HRV serotypes G1 - G4 and G9 independently. Purified anti-HRVIgY preparations were tested to determine the ELISA and neutralizing antibody titers respectively in an indirect ELISA and cell culture based neutralization assay. Efficacy of pre and post infection treatment of anti-HRV-3IgY was assessed in an infant BALB/c mouse model of human rotavirus infection by monitoring percent diarrhea, severity and duration of diarrhea, intestinal viral load and histopathology. High (1:64000 - 1:512000) titered anti-HRVIgYs were obtained from the egg yolk of immunized hens with peak titer value (1:256000/1:512000) at 40 - 60 day of immunization. In-vitro, each of the anti-HRVIgY preparations showed the presence of multiserotypic neutralizing activity with high (1:1600 - ≥1:6400) homologous and low (≤1:50 - 1:800) heterologous titers. However, anti-HRV-3IgY neutralized all of the serotypes tested in the study indicating broader in-vitro neutralizing activity. In mice, post exposure treatment with anti-HRV-3IgY significantly reduced the extent of diarrhea and intestinal virus load and inhibited histopathological changes whereas pre exposure anti-HRV-3IgY treatment imparted immediate protection from development of rotavirus gastroenteritis. Thus, the anti-HRVIgY administered orally decreased morbidity and disease incidences in mice suggesting its potential implication in prophylactic and therapeutic usage in human to achieve reduction in rotavirus disease burden.
