Ultrasound imaging of abdominal sarcoidosis: State of the art

Since it has been recognized that sarcoidosis(SA) is not an exclusive disorder of the lungs but can also affect other organs such as the liver and spleen, efforts have been made to define specific imaging criteria for the diagnosis of the single organ involvement, and the concept has been reinforced that the exclusion of alternative causes is important to achieve the correct diagnosis. Ultrasound(US)is a useful tool to evaluate patients with suspected abdominal SA, such as of the liver, spleen, kidney, pancreas and other organs, showing findings such as organomegaly, focal lesions and lymphadenopathy. While the diagnosis of abdominal SA is more predictable in the case of involvement of other organs(e.g.,lungs), the problem is more complex in the case of isolated abdominal SA. The recent use of contrast-enhanced ultrasound and endoscopic ultrasound elastography has provided additional information about the enhancement patterns and tissue rigidity in abdominal SA. Here we critically review the role of US in abdominal SA, reporting typical findings and limitations of current evidence and by discussing future perspectives of study.

Assessment of health benefit of PM_(2.5) reduction during COVID-19 lockdown in China and separating contributions from anthropogenic emissions and meteorology

The national lockdown policies have drastically disrupted socioeconomic activities during the COVID-19 pandemic in China,which provides a unique opportunity to investigate the air quality response to such anthropogenic *** it is meaningful to evaluate the potential health impacts of air quality changes during the lockdown,especially for PM_(2.5) with adverse health *** this study,by using PM_(2.5) observations from 1388 monitor-ing stations nationwide in China,we examine the PM_(2.5) variations between the COVID-19 lockdown(february and March in 2020)and the same period in 2015-2019,and find that the national average of PM_(2.5) decreases by 18 μg/m^(3),and mean PM_(2.5) for most sites(about 75%)decrease by 30%-60%.The anthropogenic and meteorological contributions to these PM_(2.5) variations are also determined by using a stepwise multiple linear regression(MLR)model combined with the Kolmogorov-Zurbenko *** results show that the change of anthropogenic emissions is a leading contributor to those widespread PM_(2.5) reductions,and meteorological conditions have the negative influence on PM_(2.5) reductions for some re-gions,such as Beijing-Tianjin-Hebei(BTH).Additionally,the avoided premature death due to PM_(2.5) reduction is estimated as a predicted number based on a log-linear concentration-response *** total avoided premature death is 9952 in China,with dominant con-tribution(94%)from anthropogenic emission *** BTH,Yangtze River Delta,Pearl River Delta and Hubei regions,the reductions of PM_(2.5) are 24.1,24.3,13.5 and 29.5 μg/m^(3),with the avoided premature deaths of 1066,1963,454 and 583,respectively.

Prevalence and risk factors of lymphatic dysfunction in cirrhosis patients with refractory ascites:An often unconsidered mechanism

N-acetylcysteine protect lymphocytes and cytokines against heavy ion irradiation via counteracting the glutamate

We evaluated the protective effect of N-acetylcysteine (NAC) on immunity system irradiated by 12C6+ ion beam. Kun-Ming mice were whole-body irradiated by 12C6+ ion at doses of 0, 0.5, 1, 1.5, 2, 2.5 and 3 gy. The results showed that in saline group, the lymphocytes DNA double-strand breaks (DSBs), maleic dialdehyde, thymocytes number in g 0 /g 1 and apoptosis percentage increased with dose increment, and the levels of interferon-γ, glutathione, superoxide radical (SOD) and natural killer cells activity decreased with dose increment. However, there were no significant changes in NAC-treated group. The data indicated that pre-treatment with NAC could significantly remove the ROS by counteracting the glutamate, decrease excessive lipid peroxidation reaction and SOD damages, and protect DNA, lymphocytes and cytokines against irradiation.

POST-TRANSCRIPTIONAL REgULATION Of P21^(WAf1/CIP1) BY P53

Objective: To investigate the post-transcriptional regulation of p21WAf1/CIP1 by p53. Methods: The MDA-MB-468 cells have endogenous mutant p53 and the MCf7 cells lines have wtp53. Recombinant p53 expression and p21WAf1/CIP1 induction were detected by Western blot analysis. Northern blot analysis was carried out to examine whether changes in p21WAf1/CIP1 protein levels in MCf7 cells treated with AdCMVp53 are reflected at the mRNA level. flow cytometric analysis of MCf7 cells following overexpression of recombination. Results: The ratio of p53: p21WAf1/CIP1 was below 1 at the early stages of AdCMVp53 infection, but increased to 1.6 by day 3 and to 9.7 by day 5 post-infection. As expected, p21WAf1/CIP1 expression was not detectable in MDA-MB-468 cells despite the presence of high levels of mutant p53 protein. The g1/S ratios in untreated controls and AdCMVβgal infected MCf7 cells were 1.10 and 1.35, respectively. By Northern blot analyzing the p21WAf1/CIP1: gAPDH ratios at different time points against the ratio at time point 0, a maximum 3-fold induction of p21WAf1/CIP1 mRNA expression relative to untreated control was observed on day 1 post-infection. The flow cytometric analysis indicated that MCf7 cells infected with AdCMVp53 undergo g1 arrest at both time points studied, with g1/S ratios ranging from 5.54 at day 1 to 5.65 at day 7. The g1/S ratios in untreated controls and AdCMVβgal infected MCf7 cells were 1.10 and 1.35, respectively. Conclusion: This study demonstrated that p53 could regulate p21WAf1/CIP1 gene expression at both the transcriptional and post-transcriptional levels in MCf7 cells. The latter mechanism may be involved in or be responsible for, the induction of cell cycle arrest by transcription-defective mutants of p53.

Pancreatic stellate cell: Pandora's box for pancreatic disease biology

Pancreatic stellate cells(PSCs) were identified in the early 1980 s, but received much attention after 1998 when the methods to isolate and culture them from murine and human sources were developed. PSCs contribute to a small proportion of all pancreatic cells under physiological condition, but are essential for maintaining the normal pancreatic architecture. Quiescent PSCs are characterized by the presence of vitamin A laden lipid droplets. Upon PSC activation, these perinuclear lipid droplets disappear from the cytosol, attain a myofibroblast like phenotype and expresses the activation marker, alpha smooth muscle actin. PSCs maintain their activated phenotype via an autocrine loop involving different cytokines and contribute to progressive fibrosis in chronic pancreatitis(CP) and pancreatic ductal adenocarcinoma(PDAC). Several pathways(e.g., JAK-STAT, Smad, Wnt signaling, Hedgehog etc.), transcription factors and mi RNAs have been implicated in the inflammatory and profibrogenic function of PSCs. The role of PSCs goes much beyond fibrosis/desmoplasia in PDAC. It is now shown that PSCs are involved in significant crosstalk between the pancreatic cancer cells and the cancer stroma. These interactions result in tumour progression, metastasis, tumour hypoxia, immune evasion and drug resistance. This is the rationale for therapeutic preclinical and clinical trials that have targeted PSCs and the cancer stroma.
