The Zuo Jin Wan Formula increases chemosensitivity of human primary gastric cancer cells by AKT mediated mitochondrial translocation of cofilin-1

Resistance to cisplatin(DDP)-based chemotherapy is a major cause of treatment failure in human gastric cancer(GC). It is necessary to identify the drugs to re-sensitize GC cells to DDP. In our previous research, Zuo Jin Wan Formula(ZJW) has been proved could increase the mitochondrial apoptosis via cofilin-1 in a immortalized cell line, SGC-7901/DDP. Due to the immortalized cells may still difficult highly recapitulate the important molecular events in vivo, primary GC cells model derived from clinical patient was constructed in the present study to further evaluate the effect of ZJW and the underlying molecular mechanism. Immunofluorescent staining was used to indentify primary cultured human GC cells. Western blotting was carried out to detect the protein expression. Cell Counting Kit-8(CCK-8) was used to evaluate cell proliferation. Flow cytometry analysis was performed to assess cell apoptosis. ZJW inhibited proliferation and induced apoptosis in primary DDP-resistant GC cells. Notably, the apoptosis in GC cells was mediated by inducing cofilin-1 mitochondrial translocation, down-regulating Bcl-2 and up-regulating Bax expression. Surprisingly, the level of p-AKT protein was higher in DDP-resistant GC cells than that of the DDP-sensitive GC cells, and the activation of AKT could attenuate ZJW-induced sensitivity to DDP. These data revealed that ZJW can increase the chemosensitivity in DDP-resistant primary GC cells by inducing mitochondrial apoptosis and AKT inactivation. The combining chemotherapy with ZJW may be an effective therapeutic strategy for GC chemoresistance patients.

Effects of Buyang Huanwu Tang Combined with Bone Marrow Mesenchymal Stem Cell Transplantation on the Expression of VEGF and Ki-67 in the Brain Tissue of the Cerebral Ischemia-Reperfusion Model Rat

Objective:To explore the mechanism of Buyang Huanwu Tang (补阳还五汤 Decoction Invigorating Yang for Recuperation) combined with bone marrow mesenchymal stem cells (MSCs) transplantation in protecting nerves of cerebral ischemic injury. Methods: Local cerebral ischemia-reperfusion rat model was established with modified Zea-Longa thread-occlusion method, and MSCs were injected into the caudal vein, and Buyang Huanwu Tang(补阳还五汤)was administrated. Vascular endothelial growth factor (VEGF) and Ki-67 expression in the ischemic side of the brain in the cerebral ischemic-reperfusion rat were detected with immuno-histochemical staining method. Results: VEGF and Ki-67 expressions were significantly up-regulated in the MSCs group and the combination group, with significant differences as compared with the model group and the sham operation group (P<0.05), and with the most strongest effect in the combination group. Conclusion: Buyang Huanwu Tang(补阳还五汤)combined with MSCs transplantation repairs the injured blood vessels and lesion tissues possibly by up-regulation of VEGF and Ki-67 expression.

The effects of Xingnao Jieyu capsules on post-stroke depression are similar to those of fluoxetine

The Xingnao Jieyu capsule has been shown to effectively relieve neurologic impairments and les- sen depression. It remains poorly understood whether this capsule can be used to treat post-stroke depression. Thus, in the present study, we established a rat model of post-stroke depression using left middle cerebral artery occlusions in combination of chronic unpredictable stress and solitary housing during development. Experimental rats received intragastric perfusion with 0.82, 0.41, and 0.20 g/kg Xingnao Jieyu capsules separately dissolved in 2 mL distilled water. Fluoxetine served as a positive control. The treatment was conducted over 28 days. Sugar water consumption test, open-field test, real-time fluorescent quantitative PCR and immunohistochemical staining results demonstrated that intragastric perfusion with various doses of Xingnao Jieyu capsules increased sugar water consumption, voluntary behaviors and synaptotagmin mRNA and protein expression in rats with post-stroke depression. These therapeutic effects were similar to those of fluoxetine. These results indicate that Xingnao Jieyu capsules upregulate synaptotagmin expression in hip pocampi of rats with post-stroke depression, and exert antidepressant effects.

OBJECTIVE： To investigate the inhibitory effect of Panax notoginseng saponins（PNS） on liver fibrosis and explore the underlying ***： Carbon tetrachloride（CCl4）-treated rats and hepatic stellate cells（HSCs） were used. The effect of PNS on CCl4-induced liver fibrosis was studied with histochemical and biochemical *** growth factor（TGF）-β1, α-smooth muscle actin（α-SMA）, and collagen Ι m RNA expression were determined by reverse transcriptase-polymerase chain reaction（RT-PCR）. Mean-while, the protein expression levels of α-SMA, collagen Ι, phosphorylation-Janus activated kinase signal transducer（p-Jak2） / Jak2, and phosphorylation-activator of transcription（p-Stat）3 / Stat3 were determined by immunohistochemistry and / or ***： PNS treatment significantly improved the liver function of rats as indicated by decreased serum enzymatic activities of alanine aminotransferase and aspartate aminotransferase. Histopathological results indicated that PNS alleviated liver damage and reduced the formation of fibrous septa. Moreover, PNS significantly decreased liver hydroxyproline and significantly attenuated expressions of collagen p-Stat3 / StatⅠ, α-SMA, TGF-β1, p-Jak2 / Jak2,and 3 in the rat liver fibrosis model and ***： PNS can relieve liver fibrosis by modulating Jak2/Stat3 signaling transduction pathway, which may be one of its mechanisms to suppress hepatic fibrosis.

Objective：To study the chemical composition,anticancer,anti-neuroinflammatory,and antioxidant activities of the essential oil of Patrinia scabiosaefolia（EO-PS）.Methods：Patrinia scabiosaefolia was analyzed by gas chromatography-mass *** human carcinoma cell lines,including SGC-7901,AGS,Hep G2,HT-29,HCT-8,5-FU/HCT-8,He La,and MDA-MB-231,were assessed by methylthiazolyldiphenyltetrazolium bromide（MTT） ***-neuroinflammatory activity was assessed by production of interleukin（IL）-1β and IL-6 induced by lipopolysaccharide in BV-2 cells（microglia from mice）.The antioxidant activity was evaluated with a 2,2-diphenyl-1-picrylhydrazyl（DPPH） scavenging ***：Forty-four components,representing 83.919% of the total oil,were identified in the *** major constituents were caryophyllene oxide（12.802%）,caryophyllene（6.909%）,α-caryophyllene（2.927%）,β-damascenone（3.435%）,calarene（5.621%）,and phenol（3.044%）.The MTT assay showed that the EO-PS exhibited significant dose-dependent growth inhibition in the 50–200 μg/m L dilution *** EO-PS exhibited a dose-dependent scavenging activity against the DPPH radical,with an half of maximal inhibitory concentration 1.455 mg/m ***：The EO-PS possesses a wide range of antitumor,anti-neuroinflammatory and antioxidant activities,suggesting that it may be a good candidate for further investigations of new bioactive substances.
