Encapsulated ferric pyrophosphate improves intestinal inflammation and gut flora disorder in suckling rats with iron deficiency anemia

The prevalence of iron deficiency anemia （IDA） remains high in infants， resulting in growth retardation， neurodevelopmental impairment， immunodeficiency and other irreversible injuries. Efficient and safe iron supplementation for infants has been the goal of recent research. This study aims to investigate the effect of encapsulated ferric pyrophosphate （FePP） on intestinal inflammation and gut microbiota in IDA suckling rats. Newborn Sprague Dawley rats were gavaged with low and high doses of FePP and FeSO4（2 and 10 mg Fe/kg BW， respectively） during postnatal days 2-14. Results showed that FePP supplementation was as effective as FeSO4in promoting growth， alleviating anemia and restoring body iron levels. Both low and high doses of FePP could significantly down-regulate the expression of pro-inflammatory cytokines in the colon to the level similar to that in the Ctrl group （P > 0.05）. However， the high dose of FeSO4did not show a down-regulation effect. Compared with the Ctrl group， IDA caused a disturbance of gut microbiota composition in suckling rats， and FePP could restore this dysbiosis. Besides， FePP was more beneficial than FeSO4in increasing the abundance of beneficial bacteria such as Bacteroides and Akkermansia. Spearman's correlation analysis showed a correlation between gut microbiota and biochemical indicators such as iron status， pro-inflammatory cytokine expression， and oxidative stress level. Overall， these findings suggested that FePP could effectively improve IDA， and is more effective than FeSO4in alleviating intestinal inflammation and regulating gut microbiota， which provides a basis for the application of new iron fortificant in infant formula.

Dry-fermented sausages are a good source of bioactive peptides,whose stability against gastrointestinal(GI)digestion determines their *** study focused on evaluating the effect of peptide extracts from sausages fermented with Staphylococcus simulans QB7 during in vitro simulated GI digestion,including peptide profiles and antioxidant and anti-inflammatory *** present in sausages were degraded during digestion,with molecular weight reduced from>12 kDa to<1.5 ***,the content of amino acids increased from 381.15 to 527.07 mg/g,especially tyrosine being found only after GI *** anti-inflammatory activities were increased after GI digestion,however,the changes in antioxidant activities were the opposite.A total number of 255,252 and 386 peptide sequences were identified in undigested,peptic-digested and GI-digested samples,***,BIOPEP-UWM and admetSAR were used to further predict the functional properties and intestinal absorption of the identified peptide sequences from GI ***,18 peptides were discovered to possess either antioxidant or anti-inflammatory capacities.

foodborne pathogens are the leading causes of human diseases that result in hundreds of thousands of deaths annually. Clostridium perfringens （C. perfringens） is a notorious foodborne pathogen of global significance that is known to produce a large repertoire of toxic factors， several of which can severely destroy the gut lining to lead to abdominal pain， diarrhea， and even death. However， like many other foodborne pathogens， there is no specific treatment for infections caused by C. perfringens and rehydration therapy is largely ineffective at reducing symptoms and preventing transmission. Given this， there is an urgent need to discover new therapeutic targets that can be harnessed to treat C. perfringens infection. We recently discovered that a highly toxic factor produced by C. perfringens named perfringolysin O （PFO） activates the innate immune sensor NLRP3. We characterized that PFO is the specific agonist of NLRP3 inflammasome and mechanistically， the intracellular entrance of PFO is required for active assembly of NLRP3 inflammasome. Further， PFO targets the mitochondria resulting in mitochondrial destabilization and ROS release. Mito-TEMPO scavenging mtROS specifically inhibits the level of caspase-1 p20 and IL-1β in both in vitro and in vivo models. Additionally， 5-hydroxydecanoate （5-HD）， a selective mitochondrial ATP-sensitive potassium channel (KATP) inhibitor， could relieve PFO-induced NLRP3 inflammasome activation as well as mitochondrial destabilization， indicating that potassium ion flux is necessary for mitochondrial damage upstream of NLRP3 inflammasome activation. We expect that the information gathered from this study will develop insights into features of the immune response that can be targeted for the treatment of food poisoning and other foodborne pathogenic infections.

Approximately 2.5% of the global population experiences allergic reactions to seafood， making it one of the most prevalent and life-threatening allergies. Seafood allergy can lead to the disruption of the intestinal barrier， possibly due to abnormalities in intestinal glycosylation. In this study， the mechanisms underlying seafood allergy were explored through the lens of intestinal glycobiology. Mice were first sensitized with tropomyosin， resulting in significant increases in allergy symptom scores and specific antibody and Th2 cytokine levels. Intestinal damage was confirmed by histopathology assessments and levels of diamine oxidase and claudin-1. Moreover， alterations in glycosylated proteins within the jejunum were analyzed using high-throughput mass spectrometry and the pGlyco3.0 search engine， yielding 2283 glycosylation peptides corresponding to 655 unique glycosylation sites on 399 proteins in precision N-glycoproteomics. Differential expression and enrichment analyses revealed that differentially expressed glycoproteins play significant roles in the extracellular matrix (ECM)-receptor interaction pathway and focal adhesion pathway. In conclusion， tropomyosin sensitization leads to intestinal glycome changes， accompanied by remodeling of the intestinal ECM. Our research establishes an essential theoretical basis for targeting the intestinal glycome and ECM remodeling in a precise and fine-tuned manner for the treatment of food allergies.

Sea buckthorn (Hippophae rhamnoides L.) is a natural homologous substance of medicine and food. Polysaccharide， as one of its primary active components， has very superior biological activity and can be used as a dietary supplement for functional foods， with good commercial prospects. Although initial progress has been made in the study of sea buckthorn polysaccharides， related studies have been fragmented and lacked systematic and generalization. This manuscript presents a critical analysis and systematic summary of the extraction and purification methods， structural characterization and physicochemical properties， biological activity and potential mechanisms， and structure-activity relationships of sea buckthorn polysaccharides. Accumulating evidence has indicated that sea buckthorn polysaccharides， which were widely prepared by water extraction and column chromatography purifications， exhibited exhibit superior biological activities in vitro and in vivo， including antioxidant， immunomodulatory， anti-inflammatory， hepatorenal protective， antibacterial， antiviral， and prebiotic activities. After analysis， it was concluded that there is a correlation between the relevant activities of sea buckthorn polysaccharides and that the structure of sea buckthorn polysaccharides has a great influence on their biological activity. We reviewed the challenges and limitations of sea buckthorn polysaccharides， summarized the critical aspects， and provided suggestions for potential breakthroughs in the research and application of sea buckthorn polysaccharide.
