Blautia has attracted attention because of its potential efficacy in ameliorating host energy metabolism and *** study aims to investigate the influences of Blautia producta D4 on colitis induced by dextran sulfate sodium(DSS)and to reveal the underlying *** showed that *** D4 intervention significantly relieved body weight loss,and suppressed the elevation of pro-inflammatory cytokines(including interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),and interleukin-1β(IL-1β))and excessive oxidative stress(myeloperoxidease(MPO)activity,superoxide dismutase(SOD)activity,glutathione peroxidase(GSH-Px)activity,and malondialdehyde(MDA)level)in colitis ***,the concentrations of tight junction proteins(occludin,claudin-1,and ZO-1)related to the intestinal barrier were obviously elevated,and colitis-related TLR4/NF-κB pathway activation was remarkably inhibited after *** D4 *** intestinal microbial disorder was evidently ameliorated by increasing the relative abundance of Clostridium sensu stricto 1,Bifidobacterium,GCA-900066225,Enterorhabdus,and reducing the relative abundance of Lachnospiraceae NK4A136 *** conclusion,oral administration of *** D4 could ameliorate DSS-induced colitis by suppressing inflammatory responses,maintaining the intestinal barrier,inhibiting TLR4/NF-κB pathway,and regulating intestinal microbiota *** results are conducive to accelerate the development of *** D4 as a functional probiotic for colitis.

Inflammatory bowel disease(IBD)is a complex relapsing inflammatory disease in the gut and is driven by complicated host-gut microbiome *** commensals have shown different functions in IBD prevention and *** gain a mechanistic understanding of how different commensals affect intestinal inflammation,we compared the protective effects of 6 probiotics(belonging to the genera Akkermansia,Bifidobacterium,Clostridium,and Enterococcus)on dextran sulfate sodium(DSS)-induced colitis in mice with or without gut ***-inflammatory properties(ratio of interleukin(IL)-10 and IL-12)of these strains were also evaluated in an in vitro mesenteric lymph nodes(MLN)co-culture *** showed that 4 probiotics(belonging to the species Bifidobacterium breve,Bifidobacterium bifidum,and Enterococcus faecalis)can alleviate colitis in normal *** probiotic strains differed in regulating the intestinal microbiota,cytokines(IL-10,IL-1βand interferon(IFN)-γ),and tight junction function(Zonulin-1 and Occludin).By constrast,Akkermansia muciniphila AH39 and Clostridium butyricum FHuNHHMY49T1 were not ***,*** JSNJJNM2 with high anti-inflammatory potential in the MLN model could relieve colitis symptoms in antibiotic cocktail(Abx)-treated ***,*** FJSWX25M1induced low levels of cytokines in vitro and showed no beneficial ***,we provided insight into the clinical application of probiotics in IBD treatment.

Gut microbiome is indispensable for maintaining normal brain ***,gut microbiota plays a causal role in sleep deprivation(SD)-induced cognitive *** this study,neurobehavioral effects of the Bifidobacterium breve strain(CCFM1025)were assessed in sleep-deprived ***1025 improved the body weight and food and water intake of the *** also alleviated SD-induced cognitive behavioural abnormalities(in the novel object recognition test),but did not show beneficial effects on mood-and spatial memory-related ***1025 significantly altered the gut microbial composition and genome *** microbial metabolites that may regulate sleep function were also identified,such as isovaleric acid andγ-aminobutyric acid in the gut and purine metabolites in the *** metabolites may participate in gutbrain communication by acting on the striatal melatonin system,for example to increase melatonin levels,and by regulating the expression of circadian clock genes such as those encoding the adenosine A2A receptor and period circadian regulator ***,administration of probiotics alleviated cognitive impairment and circadian rhythm disturbance induced by SD via modulation of gut microbiome and its *** findings may help guide the treatment of insomnia or other sleep disorders via dietary strategies.

Milk fat globule membrane(MFGM),which contains abundant glycoproteins and phospholipids,exerts beneficial effects on intestinal health and *** aim of this study was to evaluate the protective effects and possible underlying mechanisms of MFGM on cow’s milk allergy(CMA)in aβ-lactoglobulin(BLG)-induced allergic mice *** was supplemented to allergic mice induced by BLG at a dose of 400 mg/kg body *** demonstrated that MFGM alleviated food allergy symptoms,decreased serum levels of lipopolysaccharide,pro-inflammatory cytokines,immunoglobulin(Ig)E,Ig G1,and Th2 cytokines including interleukin(IL)-4,while increased serum levels of Th1 cytokines including interferon-γand regulatory T cells(Tregs)cytokines including IL-10 and transforming growth factor-β.MFGM modulated gut microbiota and enhanced intestinal barrier of BLG-allergic mice,as evidenced by decreased relative abundance of Desulfobacterota,Rikenellaceae,Lachnospiraceae,and Desulfovibrionaceae,while increased relative abundance of Bacteroidetes,Lactobacillaceae and Muribaculaceae,and enhanced expressions of tight junction proteins including Occludin,Claudin-1 and zonula ***,MFGM increased fecal short-chain fatty acids(SCFAs)levels,which elevated G protein-coupled receptor(GPR)43 and GPR109A *** increased expressions of GPR43 and GPR109A induced CD103+dendritic cells accumulation and promoted Tregs differentiation in mesenteric lymph node to a certain *** summary,MFGM alleviated CMA in a BLG-induced allergic mice model through enhancing intestinal barrier and promoting Tregs differentiation,which may be correlated with SCFAs-mediated activation of *** findings suggest that MFGM may be useful as a promising functional ingredient against CMA.

The high intraspecies heterogeneity of Baciillus coagulans leads to significant phenotypic differences among different ***,6 *** strains were tested in the present study using an irritable bowel syndrome(IBS)animal model to determine whether the IBS-alleviating effects of *** strains are *** results of this study showed that the ingestion of *** GBI-30,6086,and *** CCFM1041 significantly alleviated IBS symptoms in *** contrast,other *** strains showed no or limited alleviating effects on IBS *** to our experimental results,the two main common features of these strains were as follows:1)The resistance of vegetative cells to bile salts,and 2)ability to synthesize specific lipids and secondary *** strains based on these two indicators may greatly reduce costs and provide a basis for mining new functional *** *** results also suggest that administration of *** could significantly regulate microbiota dysbiosis in animal ***,the close relationships between the gut microbiota,gut microbiota metabolites,and IBS were further confirmed in this study.

Mucin 2 (MUC2) is a critical component of the intestinal mucus barrier. Lactic acid bacteria (LAB) strains can improve mucosal homeostasis. In this study， we determined the expression of Muc2 induced by dead bacteria and cell-free conditioned medium (CM) of 50 LAB strains in the human goblet cell line， LS174T. Dead bacteria or CM of LAB affected the Muc2 expression in a species- and strain-specific manner under homeostasis. Next， LAB strains with different regulatory abilities were selected， gavaged into mice， and exposed to dextran sodium sulfate (DSS) after 1 week. Different LAB strains inhibited intestinal injury to different degrees， with Lactobacillus acidophilus FCQHC4L1 exerting the most potent effect. FCQHC4L1 significantly decreased the secretion of pro-inflammatory factors， promoted the expression and secretion of mucin， and inhibited colitis development. This strain also regulated the gut microbiota and increased the secretion of butyric acid. Moreover， CM of FCQHC4L1 inhibited endoplasmic reticulum (ER) stress and ameliorated the abnormal expression of MUC2 by suppressing the activation of the GRP78/ATF6 and GRP78/IRE1/XBP1 signaling pathways. Our results highlight the potential of FCQHC4L1 as a therapeutic agent for strengthening the mucus barrier and improving the gut health.

The gut microbiota-brain axis has emerged as a novel target for Alzheimer's disease(AD),a neurodegenerative disease characterised by behavioural and cognitive ***,most previous microbiome-based intervention studies have focused on single factors and yielded only modest cognitive ***,we proposed a multidomain intervention strategy that combined Bifidobacterium breve treatment with environmental enrichment(EE)*** this study,we found that compared with EE or *** treatment alone,*** intervention combined with EE amplified its neuroprotective effects on AD mice,as reflected by improved cognition,inhibited neuroinflammation and enhanced synaptic ***,using microbiome and metabolome profiling,we found that the combination of *** and EE treatment restored AD-related gut microbiota dysbiosis and reversed microbial metabolite ***,by integrating behavioural and neurological data with metabolomic profiles,we revealed that the underlying mechanism may involve the modulation of microbiota-derived glutamine metabolism via gut-brain ***,combined *** intervention with EE treatment can alleviate AD-related cognitive impairment and improve brain function by regulating glutamine metabolism of the gut *** findings provide a promising multidomain intervention strategy,with a combination of dietary microbiome-based and lifestyle-targeted interventions,to promote brain function and delay the progression of AD.

Insufficient sleep can produce a multitude of deleterious repercussions on various domains of human ***,the walnut(Juglans mandshurica)confers numerous salutary biological activities pertaining to ***,the sedative and hypnotic capacities of walnut's functional constituents remain *** this investigation,we analyzed the sedative and hypnotic components of the walnut Diaphragma juglandis fructus and innovatively discovered a compound,defined as 3-hydroxy-4-iminobutyric acid(HIBA),which disrupts motor activity and enhances sleep duration by regulating the neurotransmitters(GABA,DA,etc.)within the brain and serum of ***,a metabolomics approach of the serum,basal ganglia,hypothalamus,and hippocampus as well as the gut microbiota was undertaken to unravel the underlying molecular mechanisms of sleep *** data reveal that HIBA can regulate the metabolism of basal ganglia(sphingolipids,acylcarnitines,etc.),possibly in relation to HIBA's influence on the gut microbiome(Muribaculum,Bacteroides,Lactobacillus,etc.).Therefore,we introduce a novel natural product,HIBA,and explicate the modulation of sleep promotion in mice based on the microbiota-gut-brain *** study contributes fresh insights toward natural product-based sleep research.

Infancy and toddlerhood are critical phases of life， as the gut microbiota is established here， which influences current and future health. During this period， the microbiota was relatively less stable and highly responsive to environmental factors. Therefore， it is important to understand how dietary factors affect this complex stage of microbial assembly. The effect of feeding practices (breast milk/formula) on microbial colonization in early infancy has been actively studied; however， studies on the effect of diet on the gut microbiota during the complementary feeding period are sparse. The introduction of complementary foods provides abundant new dietary compounds for the gut microbiota， which induces a shift in gut microbiota and metabolism from milk-adapted toward a more mature and diverse adult-like community. Herein， we discuss the impact of dietary nutrients (carbohydrates， proteins， fats， vitamins， and minerals) on microbiome of infants and toddlers. Furthermore， this review summarizes the effects of complementary feeding patterns， specific foods (such as cereals; legumes and nuts; vegetables and fruits; meats; dairy products)， food additives， and malnutrition (undernutrition or overnutrition) on gut microbiota of this populations. These findings might deepen our comprehension of the complex interactions between diets and the development and establishment of the gut microbiota. This may facilitate the tailoring of interventions aimed at promoting beneficial modifications within the gut microbial community. Furthermore， the insights gained could inform the design and implementation of safe and efficacious complementary feeding practices.
