PoWRKY71 is involved in Paeonia ostii resistance to drought stress by directly regulating light-harvesting chlorophyll a/b-binding 151 gene

Although the functions of WRKY transcription factors in drought resistance are well known,their regulatory mechanisms in response to drought by stabilising photosynthesis remain ***,a differentially expressed PoWRKY71 gene that was highly expressed in drought-treated Paeonia ostii leaves was identified through transcriptome ***71 positively responded to drought stress with significantly enhanced expression patterns and overexpressing PoWRKY71 in tobacco greatly improved plant tolerance to drought stress,whereas silencing PoWRKY71 in *** resulted in a drought-intolerant ***,lower chlorophyll contents,photosynthesis,and inhibited expression of photosynthesis-related light-harvesting chlorophyll a/b-binding 151(CAB151)gene were found in PoWRKY71-silenced ***,a homologous system indicated that drought treatment increased PoCAB151 promoter *** assays revealed that PoWRKY71 directly bound on the W-box element of PoCAB151 promoter and activated its *** addition,PoCAB151 overexpressing plants demonstrated increased drought tolerance,together with significantly higher chlorophyll contents and photosynthesis,whereas these indices were dramatically lower in PoCAB151-silenced *** above results indicated that PoWRKY71 activated the expression of PoCAB151,thus stabilising photosynthesis via regulating chloroplast homeostasis and chlorophyll content in *** under drought *** study reveals a novel drought-resistancemechanism in plants and provides a feasible strategy for improving plant drought resistance via stabilising photosynthesis.

MRI can reveal metabolic changes in lily bulbs in vivo during dormancy release

The factors influencing dormancy release in lily bulbs strongly affect commercialization success, but the mechanism of dormancy release is still unclear. Magnetic resonance imaging (MRI) can detect changes in morphology and water status in a living plant bulb and aid in investigating release factors. To evaluate whether MRI could be used to detect intra-bulb metabolic changes during the dormant period in Oriental Lilies (Lilium 'Sorbonne'), a series of MRI and sugar concentration measurements were performed weekly on bulbs stored for 11 weeks at 4°C. The image quality of intra-bulb structure obtained using T 1-weighted imaging was superior to that obtained using T 2 -weighted imaging and had a higher signal-to-noise ratio (0.97±0.01). Magnetization transfer ratio values for the bud and basal plate declined during the first eight weeks of cold storage (P>0.05), and were well correlated with concentration of soluble sugar in the bud (R 2 =0.95) and basal plate (R 2 =0.93). Thus, MRI can serve as a valuable tool for observation and analysis of dynamic morphological and metabolic changes in vivo during dormancy release. This information is potentially useful as a guide in the improvement of horticultural product quality.

Endogenous auxin is an important regulator of in vivo organ development,but its role in in vitro organogenesis is *** has been observed that the basal end of epicotyl cuttings of juvenile citrus seedlings produces fewer shoots than the apical ***,we report that elevated endogenous auxin levels in the basal end of citrus epicotyl cuttings are inhibitory for in vitro shoot *** transgenic citrus plants expressing an auxin-inducible GUS reporter gene,we have observed elevated levels of auxin at the basal end of stem cuttings that are mediated by polar auxin *** endogenous auxin or blocking polar auxin transport enhances shoot *** auxin transport inhibitor,N-1-naphthylphthalamic acid(NPA),can also enhance shoot organogenesis independent of its action on polar auxin ***,we demonstrate that the promotional effects of depleting endogenous auxin or blocking polar auxin transport on shoot organogenesis are *** study thus provides meaningful insights into possible roles of endogenous auxin and polar auxin transport,as well as auxin–cytokinin interactions,in in vitro shoot ***,our results may also provide practical strategies for improving in vitro shoot organogenesis for citrus and many other plant species.

GREEN INFRASTRUCTURE PLANNING ON CAMPUS CASE STUDIES FROM UNIVERSITY OF MASSACHUSETTS-AMHERST

College campuses pride themselves in being leaders in promoting innovation in both technical and theoretical aspects of all fields of study, including sustainability. The drive for sustainability in higher education has become institutionalized in many college campuses in the form of offices of sustainability, as well as membership in the American Association for Sustainability in Higher Education (AASHE). In parallel, but not always in concert, are more grass-roots efforts by students to promote sustainability through recycling programs, urban agriculture, and similar initiatives. Students are often frustrated between the classroom lessons they learn about sustainability and the slow adoption of sustainable practices in the “real-world,” including on campus. In order to address this issue, faculty have begun to engage students in the sustainability challenges facing their own campuses. The Journal of Green Building showcases these projects in the New Directions in Teaching and Research section of the journal. As part of this series, this article will highlight a course at the University of Massachusetts-Amherst in which students engage in green infrastructure planning projects for the campus.

Environmental influence in the forested area toward human health: incorporating the ecological environment into art psychotherapy

This study on the development of a psychotherapy program based on the relationship between forests and human health focused on actively considering the natural ecological environment. This study categorized and compared an art psychotherapy program that simply moved to an outdoor space and a forest-art therapy program that actively utilized the forest environment as a medium. The characteristics of the natural environment, such as openness, change, and diversity, shortened the amount of time participants took to develop a rapport and open up and played a vital role in recovering mental health. After a bold attempt at integrating forest environment and art psychotherapy by going beyond outdoor art therapy, there were significant results pertaining to improvements in mental disorders in today’s society, including stress vulnerabilities, depression, anxiety, and aggression. The research results verified that the developed forest-art therapy method had greater efficacy in relation to both the Stress Vulnerability-Interpersonal Sensitivity Scale and the Stress Vulnerability-Self-Regulation Scale.

The feasibility of using soil seed bank for natural regeneration of degraded sandy grasslands

Desertification in degraded grasslands is manifested through the development of bare sandy patches,which eventually lead to habitat *** ability of these bare sandy patches to regenerate naturally through in-situ soil seed banks is not well *** fill this knowledge gap,we randomly selected 24 bare sandy patches with areas ranging from 19 to 898 m^(2) in a desertified grassland of the Horqin sandy land,Northern China to determine whether soil seed bank can be used for natural regeneration of bare sandy *** composition and density of soil seed bank as well as aboveground vegetation composition,abundance and coverage were *** then determined their relationships with in-situ habitat *** observations showed that the studied area had low soil seed bank density and species richness,as well as depauperate soil seed bank ***,local soil seed bank was not able to provide sufficient seed source for natural *** was indicated by the relationships between aboveground vegetation,soil seed bank and the in-situ habitat *** bare patches with an area between 300 m^(2) and 900 m^(2),increase the soil seed bank density and species richness should be the main restoration *** bare patches with a small area of less than 50 m^(2),restoration of vegetation density should be the main *** data highlighted that different extents of desertification,indicated by different bare patches,are requiring distinct restoration measures.

Application of Cydia pomonella expressed sequence tags： Identification and expression of three general odorant binding proteins in codling moth

The codling moth, Cydia pomonella, is one of the most important pests of pome fruits in the world, yet the molecular genetics and the physiology of this insect remain poorly understood. A combined assembly of 8？341 expressed sequence tags was generated from Roche 454 GS-FLX sequencing of eight tissue-specific cDNA libraries. Putative chemosensory proteins （12） and odorant binding proteins （OBPs）（18） were annotated, which included three putative general OBP （GOBP）, one more than typically reported for other Lepidoptera. To further characterize CpomGOBPs, we cloned cDNA copies of their transcripts and determined their expression patterns in various tissues. Cloning and sequencing of the 698？nt transcript for CpomGOBP1 resulted in the prediction of a 163 amino acid coding region, and subsequent RT-PCR indicated that the transcripts were mainly expressed in antennae and mouthparts. The 1？289 nt （160 amino acid） CpomGOBP2 and the novel 702 nt （169 amino acid） CpomGOBP3 transcripts are mainly expressed in antennae, mouthparts, and female abdomen tips. These results indicate that next generation sequencing is useful for the identification of novel transcripts of interest, and that codling moth expresses a transcript encoding for a new member of the GOBP subfamily.

Isolation and Functional Analysis of the Regulation of Branching by Isopentenyl Transferase Gene CmIPT1 in <i>Chrysanthemum morifolium </i>cv. ‘Jinba’

Today’s chrysanthemums are highly evolved flowering plants and they are considered as one of the most important ornamental cut flowers. In this research an isopentenyl transferase gene named CmIPT1 was isolated from Chrysanthemum morifolium cv. ‘Jinba’ using RACE and RT-PCR methods. The full cDNA sequence of CmIPT1 was 873 bp which encoded a deduced protein of 290 amino acids. It contained GxxGxGKS which is a conserved sequence of the typical domain of IPT family. The phylogenetic tree analysis of CmIPT1 in Chrysanthemum morifolium cv. ‘Jinba’ shows that it has the closest relationship with CcIPT1 in Cynara cardunculus var. scolymus. Expression of CmIPT1 was higher in stems and apex, whereas it was lower in leaves and roots. And the overexpression of CmIPT1 obviously increased the number of rosette branches in Arabidopsis. Here, in our study, we showed that CmIPT1 is a positive regulator of branch development in Chrysanthemum and may play a key role in regulating lateral branch formation of Chrysanthemum plants.

Gene editing in plants:progress and challenges

The clustered regularly interspaced short palindromic repeat(CRISPR)-associated protein 9(Cas9)genome editing system is a powerful tool for targeted gene modifications in a wide range of species,including plants. Over the last few years, this system has revolutionized the way scientists perform genetic studies and crop breeding, due to its simplicity, flexibility, consistency and high efficiency. Considerable progress has been made in optimizing CRISPR/Cas9 systems in plants, particularly for targeted gene mutagenesis. However, there are still a number of important challenges ahead, including methods for the efficient delivery of CRISPR and other editing tools to most plants, and more effective strategies for sequence knock-ins and replacements. We provide our viewpoint on the goals, potential concerns and future challenges for the development and application of plant genome editing tools.
