Weather models are essential tools for checking of the effect of the weather elements in terms of their effect on the production of the crop. This research is an attempt to see the effect of only two variables i.e., temperature and rainfall for the division Faisalabad (semitropical region of Pakistan).The model fitted is of the linear form:the values of a,b, c have been found. The expected yield has been calculated by using the aridity indices (X1 and X2 ) and the result in the form of coefficient of determination R2 has been found equal to 0.166. The significance of the regression coefficient has been tested, which shows that the contribution to the yield from aridity index at germination and that at ripening is *** wheat yields are the results of a wide variety of variables, most of which show varying degree of relationship with one another, some positive and some negative in terms of output. These variables may be technology, fertilizers, pesticides, epidemics, kinds of seeds used, market price of crop and the area under cultivation etc, which can be the source of variation in the wheat yield. Since rainfall during germination and temperature at the ripening periods are the necessary factors for the yield of wheat, for this purpose these parameters have been studied in order to their contribution.

The ability to screen larger populations with fewer replicates and non-destructive measurements is one advantage of high-throughput phenotyping(HTP)over traditinal phenotyping *** this study,two wheat accessions were grown in a controlled-environment with a moderate drought imposed from stem elongation to ***-green-blue(RGB)imaging was performed on 17 of the 22 d following the start of drought *** measurements from all plants were performed at the conclusion of the *** effect of line was signifcant for shoot dry matter,spike dry matter,root dry matter,and tller number,while the water treatment was significant on shoot dry matter and root dry *** temporal,non-destructive nature of HTP allowed the drought treatment to be significantly differentiated from the well-watered treatment after 6 d in a line from Argentina and 9 d in a line from *** difference of 3 d indicated an increased degree of drought tolerance in the line from ***,HTP from the final day of imaging accurately predicted reference plant height(r=1),shoot dry matter(r=0.95)and tller number(r=0.91).This experiment ilustrates the potential of HTP and its use in modeling plant growth and development.

Brassinosteroid(BR)is a vital plant hormone that regulates plant growth *** RESISTANT1(BZR1)is a key transcription factor in BR signaling,and its nucleocytoplasmic localization is crucial for BR ***,the mechanisms that regulate BzR1 nucleocytoplasmic distribution and thus the homeostasis of BR signaling remain largely *** vacuole is the largest organelle in mature plantcells and plays a key role in maintenance of cell ular pH,storage of intracellular substances,and transport *** this study,weuncovered anovel mechanismof BR signaling homeostasis regulatedbythe vacuolar H+-ATPase(V-ATPase)and BZR1 feedback *** results revealed that the vha-a2 vha-a3 mutant(vha2,lacking V-ATPase activity)exhibits enhanced BR signaling with increased total amount of BZR1,nuclearlocalized BZR1,and the ratio of BZR1/phosphorylated BZR1 in the *** biochemical assays revealed that VHA-a2 and VHA-a3 of V-ATPase interact with the BZR1 protein through a domain that is conserved across multiple ***-a2 and VHA-a3 negatively regulate BR signaling by interacting with BzR1 and promoting its retention in the ***,a series of molecular analyses demonstrated that nuclear-localized BZR1 could bind directlyto specific motifs in the promoters of VHA-a2 andVHAa3topromote their *** together,these results suggest that V-ATPase and BzR1 mayforma feedback regulatory loop to maintain thehomeostasis of BR signaling in Arabidopsis,providing new insights into vacuole-mediated regulation of hormone signaling.

In order to generate transgenic donor cells for nuclear transfer, bovine fetal fibroblasts were isolated in vitro and transfected with the eukaryotic expression vector pSRA-EGFP-Ipr1. The mouse Ipr1 gene and human SR-A promoter were successfully cloned and then used to construct this macrophage-specific eukaryotic expression vector. Bovine fetal fibroblasts in stable primary culture (4th passage) were transfected with pSRA-EGFP-Ipr1 by electroporation. Fluorescence from GFP was observed after 24h. Transgenic cells were selected using G418 and the resultant monoclones were picked and expanded. The transgenic cells, at the 9 th passage, were evaluated by PCR and flow cytometry. The inserted Ipr1 was confirmed by PCR, indicating stable integration of the transgene into the genome and cells had normal karyotypes and very good appearance, which indicate no deleterious result of the transgenesis. In conclusion, the cells obtained could be used as donor cells for nuclear transfer for further research of transgenic cattle.

High auxin stimulates callus through SDG8-mediated histone H3K36 methylation in Arabidopsis

Callus induction,which results in fate transition in plant cells,is considered as the first and key step for plant *** process can be stimulated in different tissues by a callus-inducing medium(CIM),which contains a high concentration of phytohormone *** a few key regulators for callus induction have been identified,the multiple aspects of the regulatory mechanism driven by high levels of auxin still need further ***,we find that high auxin induces callus through a H3 K36 histone methylation-dependent mechanism,which requires the methyltransferase SET DOMAIN GROUP 8(SDG8).During callus induction,the increased auxin accumulates SDG8 expression through a TIR1/AFBs-based transcriptional ***8 then deposits H3 K36 me3 modifications on the loci of callus-related genes,including a master regulator WOX5 and the cell proliferation-related genes,such as *** epigenetic regulation in turn is required for the transcriptional activation of these genes during callus *** findings suggest that the massive transcriptional reprogramming for cell fate transition by auxin during callus formation requires epigenetic modifications including SDG8-mediated histone H3 K36 *** results provide insight into the coordination between auxin signaling and epigenetic regulation during fundamental processes in plant development.

As one of the most important crops in the world,rice(Oryza sativa)is a model plant for metabolome *** many studies have focused on the analysis of specific tissues,the changes in metab-olite abundance across the entire life cycle have not yet been *** this study,combining both tar-geted and nontargeted metabolite profiling methods,a total of 825 annotated metabolites were quantified in rice samples from different tissues covering the entire life *** contents of metabolites in different tissues of rice were significantly different,with various metabolites accumulating in the plumule and radicle during seed *** these data with transcriptome data obtained from the same time period,we constructed the Rice Metabolic Regulation *** metabolites and co-expressed genes were further divided into 12 clusters according to their accumulation patterns,with members within each cluster displaying a uniform and clear pattern of abundance across *** this dataset,we established a comprehensive metabolic profile of the rice life cycle and used two independent strategies to identify novel transcription factors-namely the use of known regulatory genes as bait to screen for new networks underlying lignin metabolism and the unbiased identification of new glycerophospholipid metabolism regulators on the basis of tissue *** study thus demonstrates how guilt-by-association analysis of metabolome and transcriptome data spanning the entire life cycle in cereal crops provides novel resources and tools to aid in understanding the mechanisms underlying important agro-nomic traits.
