Serum Adiponectin and Resistin Levels in Patients with Polycystic Ovarian Syndrome and Their Clinical Implications

In this study, serum adiponectin and resistin levels were determined in 46 patients with polycystic ovarian syndrome (PCOS), and their correlation with serum sexual hormones and insulin resistance (IR) were examined.The subjects included 26 obese patients with body mass index (BMI)>25 and 20 non-obese patients with BMI≤25, with 25 obese and 25 non-obese healthy volunteers without PCOS serving as controls.Serum adiponectin and resistin levels in all subjects were measured, and endocrinal and metabolic indices were also analysed.Our results showed that the serum adiponectin levels in both obese and non-obese PCOS groups were significantly lower than their controls, while the serum resistin levels in obese and non-obese PCOS group were significantly higher than in their controls (P<0.001).The serum adiponectin level was significantly lower and serum resistin level significantly higher in the non-obese PCOS group as compared with the obese control group (P<0.05).Serum adiponectin level was negatively correlated with FIN, HOMA-IR, LH and LH/FSH (P<0.05), but serum resistin level was positively correlated with FIN, HOMA-IR, LH and LH/FSH (P<0.05).We are led to conclude that PCOS patients have obvious IR, low serum adiponectin and high serum resistin, and adiponectin and resistin might play important roles in the pathogenesis of IR in PCOS patients.

Aim: To investigate the effect of cryopreservation on the plasma membrane integrity in the head and tail regions ofindividual sperm, and the relationship between intact cryopreserved sperm and its motility and zona-free hamster oocytepenetration rate. Methods: The eosin Y exclusion and the hypoosmotic swelling tests were combined to form a sin-gle test (HOS-EY test) to identify the spermatozoa with four types of membrane integrity. Results: After cryop-reservation, there was a marked decline in the percentage of spermatozoa with Type Ⅳ membrane integrity (head mem-brane intact/tail membrane intact), and a significant increase in those with Type Ⅰ (head membrane damaged/tail mem-brane damaged) and Type Ⅲ (head membrane damaged/tail membrane intact) membrane integrity (n = 50, P 0.05). Conclusion: (1) The HOS-EY test has the advan-tage of showing four patterns of membrane integrity in individual spermatozoon; (2) Cryopreservation causes a signifi-cant membrane rupture in the head and tail regions of spermatozoa; Type Ⅲ is the main transitional state of membranecryodamage; (3) Cryodamage to head and tail membrane may occur independently; the presence of an intact tail mem-brane does not necessarily indicate the intacmess of head membrane. (4) Intact membranes are closely related to post-thaw motility, but do not reflect the fertilizing potential.

Recurrent pregnancy loss is defined by the World Health Orgnization as the loss of three or more consecutive pregnancies before the 20th week[1].According to studies,the prevalence of this disorder in Europe and America is between 1%and 4%for women of reproductive age.Various etiologies including uterine factors,thrombophilia,endocrine,physiological,genetic,and immunological factors are considered risk factors for recurrent pregnancy loss.

To the Editor:Systemic lupus erythematosus(SLE)is an autoimmune disease that tends to affect females of childbearing age.There are nearly 4 million people in China currently suffering from SLE,which can cause multiple organ damage in its early stages and seriously affect patients’quality of life.[1-3]Menstrual disorders are common in SLE patients and vary from heavy menstrual bleeding to amenorrhea.The etiology of menstrual irregularities includes the disease activity itself,glucocorticoids(GCs)treatment,and the use of some immunosuppressants.[4]This multicenter,cross-sectional,web-based observational study investigated menstrual irregularity,pregnancy outcomes,and birth outcomes on SLE patients across China.The research was approved by the Ethics Committee of Peking University People’s Hospital,China(No.2019PHB253-01).The patients were informed that consent was implied with completion of the questionnaire,and the process was anonymous and voluntary.

Natural killer(NK)cells are thought to play a key role in the successful establishment of a pregnancy by facilitating immunological adaptation of the semi-allogeneic developing embryo.The aim of this study was to explore the cell number,immunophenotypic characteristics,and activities of peripheral blood NK cells in women with repeated implantation failure(RIF).Peripheral blood was obtained from 27 women with RIF and 11 healthy,fertile controls during the middle luteal phase of the menstrual cycle.CD3^-CD56^+NK cells were quantified and analyzed by flow cytometry for the expression of cytolytic molecules(granzyme B,granulysin,and perforin)as well as cell surface receptors responsible for NK cell activation or inhibition(NKG2D,NKp30,NKp46,CD158a,CD158b).NK cytotoxicity was measured at three effector-to-target cell ratios.Women with RIF and fertile controls did not differ significantly in the percentage of circulating CD3CD56t NK cells,or in the proportions of these cells that expressed granzyme B,granulysin,or perforin.The two groups also did not differ significantly in the proportions of NK cells expressing the receptors NKG2D,NKp30,NKp46,CD158a or CD158b.General linear model analysis showed that NK cytotoxicity increased with effector-to-target cell ratio.However,NK cytotoxicity did not differ significantly between patients with RIF and fertile controls.These results suggest that RIF is not associated with significant alterations in the number or function of peripheral blood NK cells.

Infertility is experienced by 8%–12%of adults in their reproductive period globally and has become a prevalent concern.Besides routine therapeutic methods,stem cells are rapidly being examined as viable alternative therapies in regenerative medicine and translational investigation.Remarkable progress has been made in understanding the biology and purpose of stem cells.The affected pluripotent stem cells(iPSCs)and mesenchymal stem cells(MSCs)are further studied for their possible use in reproductive medicine,particularly for infertility induced by premature ovarian insufficiency and azoospermia.Accordingly,this study discusses current developments in the use of some kinds of MSCs such as adipose-derived stem cells,bone marrow stromal cells,umbilical cord MSCs,and menstrual blood MSCs.These methods have been used to manage ovarian and uterine disorders,and each technique presents a novel method for the therapy of infertility.

To the Editor:Infertility affects 15%to 20%of couples worldwide,and assisted reproductive technology(ART)is the most effective treatment strategy.In addition to female age,many other factors,such as antral follicle count(AFC),basal follicle-stimulating hormone(FSH)level,and anti-Mullerian hormone(AMH)level,collectively known as biomarkers for ovarian reserve tests(ORTs),are widely used to predict ovarian responses and assisted-pregnancy outcomes.[1]Among them,AMH and FSH levels were discordant in 20%to 43%of women undergoing ART treatment,and AMH was proven to be superior in predicting live birth rate(LBR).[2]

Piwi-interacting RNAs(piRNAs)is a novel class of non-coding RNAs.However,changes in piRNA expression profiles in recurrent spontaneous abortion(RSA)have not yet been investigated.The aim of this study was to identify differentially expressed piRNAs in deciduas of RSA patients.Decidua tissues were collected by curettage from recruited RSA patients and normal early pregnant(NEP)women with their informed consent.Small RNA sequencing was used to evaluate the differences in piRNA expression profiles between RSA and NEP.The present results demonstrated that the counts of total piRNA reads in RSA samples were increased compared with those in NEP samples(0.21%vs.0.11%).Differential expression analysis identified 29 upregulated piRNAs and 18 downregulated piRNAs in RSA samples.RT-qPCR further confirmed that the expression levels of uniq-109625,uniq-89328,uniq-50651 and uniq-4569 were decreased in 8 RSA tissues,compared with 13 NEP tissues.Otherwise,pi-22628 and uniq-173406 were increased in 8 RSA tissues.Based on GO term and KEGG pathway analysis,we speculate that these piRNAs regulate RSA by targeting extracellular matrix component pathway,cell adhesion pathway and focal adhesion pathway.PiRNAs may be involved in RSA pathogenesis by target genes function on adhesion and extracellular matrix component.
