Occurrence of viable but non-culturable(VBNC)pathogenic bacteria in tap water of public places

Viable but non-culturable(VBNC)bacteria have been detected in source water and effluent of drinking water treatment processes,leading to significant underestimation of viable cell *** information exists on VBNC bacteria in tap water,particularly in public *** address this gap,a comprehensive nine-month study was conducted in a major city in south-eastern China,using culture-based and quantitative PCR with propidium monoazide(PMA)dye ***-five samples were collected from five representative public places(railway station,campus,hospital,shopping mall,and institution).The findings revealed that culturable bacteria represented only 0–17.51%of the viable 16S rRNA genes,suggesting that the majority of viable bacteria existed in an uncultured or VBNC ***,opportunistic pathogens such as Escherichia coli,Enterococcus faecalis,Pseudomonas aeruginosa,Salmonella sp.,and Shigella *** primarily detected as VBNC cells,with concentrations ranging from 1.03×10^(0)to 3.01×10^(3),1.20×10^(0)to 1.42×10^(2),1.32×10^(0)to 8.82×10^(0),1.00×10^(0)to 6.71×10^(1),and 2.07×10^(0)to 1.93×10^(2)cell equivalent/100 mL,*** *** was observed in tap water after prolonged stagnation,indicating potential risks associated with bacterial *** and temporal factors accounted for 17.1%and 26.0%,respectively,of the variation in tap water community structure during the sampling period,as revealed by 16S rRNA amplicon *** study provides quantitative insights into the occurrence of VBNC bacteria in tap water and highlights the need for more sensitive monitoring methods and microbial control techniques to enhance tap water safety in public locations.

Cellular senescence is the results of aging and age-related diseases,and the development of anti-aging methods may improve health and extend *** natural flavonol fisetin has been shown to antagonize senescence in vitro and increases longevity in vivo,but has poor water solubility and limited *** this study,a food-grade and senescent cell-targeted delivery system for fisetin was developed based on whey protein isolate-galactooligosaccharides(WPI-GOS)Maillard conjugate,which could recognize senescence associatedβ-galactosidase in senescent *** fisetin nanoparticles possessed a high encapsulation efficiency,excellent dispersibility in water,good storage stability and well ***,they could effectively accumulate and retain in senescent cells with excellent senescent cell-targeting efficacy,and inhibit the oxidative stress-induced cellular senescence in ***,this novel nanoparticle system based on WPI-GOS Maillard conjugate showed promise to deliver hydrophobic bioactive ingredients like fisetin to senescent cells to improve their bioavailability and anti-senescence effect.

Diet mediates the gut microbiome composition and assembly processes in infant and toddler populations

Infancy and toddlerhood are critical phases of life， as the gut microbiota is established here， which influences current and future health. During this period， the microbiota was relatively less stable and highly responsive to environmental factors. Therefore， it is important to understand how dietary factors affect this complex stage of microbial assembly. The effect of feeding practices (breast milk/formula) on microbial colonization in early infancy has been actively studied; however， studies on the effect of diet on the gut microbiota during the complementary feeding period are sparse. The introduction of complementary foods provides abundant new dietary compounds for the gut microbiota， which induces a shift in gut microbiota and metabolism from milk-adapted toward a more mature and diverse adult-like community. Herein， we discuss the impact of dietary nutrients (carbohydrates， proteins， fats， vitamins， and minerals) on microbiome of infants and toddlers. Furthermore， this review summarizes the effects of complementary feeding patterns， specific foods (such as cereals; legumes and nuts; vegetables and fruits; meats; dairy products)， food additives， and malnutrition (undernutrition or overnutrition) on gut microbiota of this populations. These findings might deepen our comprehension of the complex interactions between diets and the development and establishment of the gut microbiota. This may facilitate the tailoring of interventions aimed at promoting beneficial modifications within the gut microbial community. Furthermore， the insights gained could inform the design and implementation of safe and efficacious complementary feeding practices.

Aflatoxin B_(1)(AFB_(1))is a naturally-occurring mycotoxin and recognized as the most toxic foodborne toxin,particularly causing damages to *** podocytes are terminally differentiated epithelial ***_(1)induces podocyte inflammation,proteinuria and renal *** the mechanism of AFB_(1)-induced podocyte inflammation and murine kidney dysfunction,we detected that AFB_(1)increased ubiquitindependent degradation of the transcription factor RelA through enhanced interaction of RelA with E3 ubiquitin ligase tripartite motif containing 7(TRIM7)in mouse podocyte clone-5(MPC-5)and mouse *** of RelA resulted in decreasing microRNA-9(miR-9)and activating the chemokine receptor 4(CXCR4),thioredoxin interacting protein(TXNIP),and NOD-like receptor pyrin domain-containing 3(NLRP3)signaling axis(CXCR4/TXNIP/NLRP3 pathway),leading to podocyte *** also determined that downregulation of miR-9 led to CXCR4 expression and the downstream TXNIP/NLRP3 pathway *** of miR-9 or deletion of CXCR4 suppressed AFB_(1)-induced CXCR4/TXNIP/NLRP3 pathway,resulting in alleviating podocyte inflammation and kidney *** findings indicated that ubiquitin-dependent proteolysis of RelA,downregulation of miR-9,and activation of CXCR4/TXNIP/NLRP3 pathway played an essential role in AFB_(1)-induced glomerular podocyte *** study revealed a novel mechanism,via RelA,for the control of AFB_(1)’s nephrotoxicity,leading to an effective protection of food safety and public health.

Insufficient sleep can produce a multitude of deleterious repercussions on various domains of human ***,the walnut(Juglans mandshurica)confers numerous salutary biological activities pertaining to ***,the sedative and hypnotic capacities of walnut's functional constituents remain *** this investigation,we analyzed the sedative and hypnotic components of the walnut Diaphragma juglandis fructus and innovatively discovered a compound,defined as 3-hydroxy-4-iminobutyric acid(HIBA),which disrupts motor activity and enhances sleep duration by regulating the neurotransmitters(GABA,DA,etc.)within the brain and serum of ***,a metabolomics approach of the serum,basal ganglia,hypothalamus,and hippocampus as well as the gut microbiota was undertaken to unravel the underlying molecular mechanisms of sleep *** data reveal that HIBA can regulate the metabolism of basal ganglia(sphingolipids,acylcarnitines,etc.),possibly in relation to HIBA's influence on the gut microbiome(Muribaculum,Bacteroides,Lactobacillus,etc.).Therefore,we introduce a novel natural product,HIBA,and explicate the modulation of sleep promotion in mice based on the microbiota-gut-brain *** study contributes fresh insights toward natural product-based sleep research.

Sea cucumber gut tetrapeptide ameliorates alcoholic gastric damage via restoring mitochondrial dynamics

This paper aimed to explore the mechanism of tetrapeptide VTPY in improving alcoholic gastric injury. VTPY has the potential to enhance the growth and movement of normal GES-1 cells. Following ethanol-induced impairment， VTPY effectively improved migration of GES-1 and HUVEC cells， enhanced angiogenesis， eliminated cellular and mitochondrial ROS， inhibited excessive mitochondrial division， enhanced F-actin polymerization and mitochondrial respiratory capacity. To counteract excessive mitochondrial fission， VTPY primarily restores the mitochondria dynamics by reducing the expression of Drp1 and Fis1， while increasing Mfn2. Further studies utilizing inhibitors clarifies that the inhibition of excessive mitochondrial fission can markedly reduce F-actin depolymerization， consequently enhancing cell migration. Additionally， VTPY can inhibit the apoptosis pathway by maintaining potential of mitochondrial membrane， preventing the release of mitochondrial cytochrome c， bolstering the levels of Bcl-XL， while reducing the levels of Bax and cleaved-caspase3. Further investigations using inhibitors demonstrates that excessive mitochondrial fission could activate apoptotic pathway. However， VTPY counteracts this effect and enhance cells viability. Further evidence suggests that VTPY effectively improves ulcer index and pathologic changes， relieves inflammation， enhances the balance of oxidation and anti-oxidation， promotes angiogenesis， improves the expression of mitochondrial dynamics factors， blocks apoptotic pathway， and subsequently ameliorates gastric damage in mice through Fis-1/Bcl-2 pathway.

Weizmannia coagulans spores alleviates DSS-induced ulcerative colitis model by modulation of gut flora， metabolites and suppressing TLR4/MyD88/NF-κB pathway

Ulcerative colitis (UC) is a chronic intestinal inflammatory disease characterized by a complex pathogenesis. Weizmannia coagulans has emerged as a potential probiotic for treating intestinal disorders. This study aimed to assess the therapeutic impact of Weizmannia coagulans BC99 on mice with DSS-induced UC and to elucidate its underlying mechanism of action. Our findings revealed that BC99 administration ameliorated symptoms associated with DSS-induced UC mice， as evidenced by reduced disease activity indexes， reversal of weight loss， and normalization of colon length. Furthermore， BC99 treatment also protected the integrity of the intestinal barrier through maintaining the antioxidant activity and the expression of tight junction proteins (ZO-1 and occluding)， and regulating the inflammatory cytokines in DSS-induced UC mice. Additionally， BC99 supplementation enhanced the production of short-chain fatty acids (SCFAs) through the proliferation of SCFA-producing bacteria， including Bidobacterium， Blautia and Faecallbaculum. Notably， the NF-κB signaling pathway was found to be closely related to BC99 treatment in DSS-induced UC mice. The positive protein expression the mRNA expression of TLR4， MyD88 and p65 in colon tissue were all detected in BC99-treated groups， which indicating that BC99 could alleviate UC symptoms by inhibiting TLR4/MyD88/NF-κB signaling pathway. Metabolomics further confirms the previous results. Collectively， these findings provide basic support for the Weizmannia coagulans as a functional food additive or a promising therapeutic agent for the effective management of UC.

Effects of Using Multivitamins as Dietary Supplements on Cognitive Function in Older Adults: a Systematic Review and Meta-analysis

Multivitamins were widely used health supplements that replenished essential nutrients in the human body. Despite their popularity， the impact of multivitamins on the cognitive function of older adults remained unclear and contentious. This study offered a comprehensive review and meta-analysis of research published until June 2024， analyzing the effects of multivitamins on various cognitive functions in individuals aged 65 and older. We included ten randomized controlled trials encompassing 13，600 participants from multiple databases. These studies evaluated the impact of multivitamins on reasoning， memory， learning， visual perception， idea production， cognitive speed， psychomotor abilities， and higher cognitive functions. Our meta-analysis revealed that multivitamins significantly enhanced delayed free recall (SMD = 0.09， 95% CI: [0.05， 0.13]， p < 0.0001). However， they had no substantial effects on immediate free recall (SMD = 0.85， 95% CI: [-0.18， 1.90]， p = 0.11)， idea production (SMD = -0.00， 95% CI: [-0.04， 0.03]， p = 0.86)， or cognitive speed (SMD=0.34， 95% CI: [-0.07， 0.74]， p = 0.11). Thus， while multivitamins facilitated delayed free recall， they did not significantly improve other cognitive functions in older adults.

Curcumin delivery nanoparticles based on Maillard reaction of Haematococcus pluvialis protein/galactose for alleviating acute alcoholic liver damage

The aim of this study is to investigate the feasibility of Maillard reaction products of Haematococcus pluvialis protein and galactose(HPP-GAL)for improving the bioactivities of curcumin(CUR)for alleviating alcoholic liver *** was embedded into HPP-GAL nanoparticles by the self-assembly of hydrogen bonding and hydrophobic interaction with the particle size around 200 ***-GAL enhanced the encapsulation efficiency and loading amount of CUR with the value of(89.21±0.33)%and(0.500±0.004)%,*** stabilities of CUR under strong acid,salt ion stability and ultraviolet irradiation conditions were improved by the ***-GAL-CUR nanoparticles exhibited excellent concentration-dependent in vitro antioxidant activities including DPPH and ABTS scavenging rates,and better protective effect on CUR against gastric acid environment as well as longer release of CUR in simulated intestinal *** addition,the HPPGAL-CUR delivery system possessed liver targeting property due to the existence of GAL,which could effectively alleviate the alcohol-induced liver damage and the inflammation indexes by inhibiting the oxidative ***,HPP-GAL-CUR nanoparticles might be a potential candidate system for the prevention of alcoholic liver damage in the future.
