High mobility group box 1 protein： possible pathogenic link to atrial fibrillation

Atrial fibrillation （AF） is the most common sustained dysrhythmia in clinical practice. The bulk of evidence suggests that inflammatory processes, oxidative stress and matrix metalloproteinase are associated with development of AF. However, these agents may be involved in high mobility group box 1 protein （HMGB1）. We hypothesized that HMGB1 may be a possible pathogenic link to AF. A growing body of evidence supports these hypotheses. First, the level of serum HMGB1 is significantly increased in patients with AF including paroxysmal and persistent AF. Second, HMGB1 has been identified as a new pro-inflammatory cytokine in cardiovascular diseases, along with tumor necrosis factor （TNF）-a, interleukin （IL）-6, and C-reactive protein, and there is cross-talk between HMGB1 and inflammatory cytokines. Third, oxidative stress is involved in the release of the pro-inflammatory cytokine, HMGB1, indicating there is cross-talk between oxidative stress and inflammation, and oxidative stress may reinforce the effect of inflammation on the pathogenesis of AF and inflammation may play a more important role in the pathogenesis of AF. Fourth, HMGB1 can promote matrix metalloproteinase-9 upregulation and activation. Fifth, HMGB1 receptors （receptor for advanced glycation end products, Toll-like receptor-2,4） may mediate the atrial structural remodeling or be up-regulated in patients with non-valvular AF. These results suggest that HMGB1 may participate in the pathogenesis of AF and provide a potential target for pharmacoloclical interruption of AF.

External anal sphincter electromyography in multiple system atrophy:implications for diagnosis,clinical correlations,and novel insights into prognosis

Multiple system atrophy is a sporadic,progressive,adult-onset,neurodegenerative disorder characte rized by autonomic dysfunction symptoms,parkinsonian features,and cerebellar signs in va rious combinations.An early diagnosis of multiple system atrophy is of utmost impo rtance for the proper prevention and management of its potentially fatal complications leading to the poor prognosis of these patients.The current diagnostic criteria incorporate several clinical red flags and magnetic resonance imaging marke rs supporting diagnosis of multiple system atrophy.Nonetheless,especially in the early disease stage,it can be challenging to differentiate multiple system atrophy from mimic disorders,in particular Parkinson’s disease.Electromyography of the external anal sphincter represents a useful neurophysiological tool for diffe rential diagnosis since it can provide indirect evidence of Onuf’s nucleus degeneration,which is a pathological hallmark of multiple system atrophy.However,the diagnostic value of external anal sphincter electromyography has been a matter of debate for three decades due to controve rsial reports in the literature.In this review,after a brief ove rview of the electrophysiological methodology,we first aimed to critically analyze the available knowledge on the diagnostic role of external anal sphincter electromyography.We discussed the conflicting evidence on the clinical correlations of neurogenic abnormalities found at external anal sphincter electro myography.Finally,we repo rted recent prognostic findings of a novel classification of electromyography patterns of the external anal sphincter that could pave the way toward the implementation of this neurophysiological technique for survival prediction in patients with multiple system atrophy.

Although the endothelium has a number of important functions, the term endothelial dysfunction is commonly used to describe impairment in its vasodilatory capacity. There have been numerous studies evaluating the relationship between endothelial dysfunction and cardiovascular disease, however assessment of endothelial function is perhaps still primarily thought of as a research tool and has not reached widespread clinical acceptance. In this review we explore the relationship between endothelial dysfunction and cardiovascular disease, its prognostic significance, methods of pharmacological reversal of endothelial dysfunction, and ask the question, is reversal of endothelial dysfunction still an attractive target in modern cardiology?

Independent and joint association of physical activity and sedentary behavior on all-cause mortality

Backgrounds:Physical activity(PA)and sedentary behavior(SB)have been associated with mortality,while the joint association with mortality is rarely reported among Chinese population.We aimed to examine the independent and joint association of PA and SB with all-cause mortality in southern China.Methods:A cohort of 12,608 China Hypertension Survey participants aged≥35 years were enrolled in 2013 to 2014,with a follow-up period of 5.4 years.Baseline self-reported PA and SB were collected via the questionnaire.Kaplan–Meier curves(log-rank test)and Cox proportional hazards regression were performed to evaluate the associations of PA and SB on all-cause mortality.Results:A total of 11,744 eligible participants were included in the analysis.Over an average of 5.4 years of follow-up,796 deaths occurred.The risk of all-cause mortality was lower among participants with high PA than those with low to moderate level(5.2%vs.8.9%;hazards ratio[HR]:0.75,95%confidence interval[CI]:0.61–0.87).Participants with SB≥6 h had a higher risk of all-cause mortality than those with SB<6 h(7.8%vs.6.0%;HR:1.37,95%CI:1.17–1.61).Participants with prolonged SB(≥6 h)and inadequate PA(low to moderate)had a higher risk of all-cause mortality compared to those with SB<6 h and high PA(11.2%vs.4.9%;HR:1.67,95%CI:1.35–2.06).Even in the participants with high PA,prolonged SB(≥6 h)was still associated with the higher risk of all-cause mortality compared with SB<6 h(7.0%vs.4.9%;HR:1.33,95%CI:1.12–1.56).Conclusions:Among Chinese population,PA and SB have a joint association with the risk of all-cause mortality.Participants with inadequate PA and prolonged SB had the highest risk of all-cause mortality compared with others.

EFFECTS OF CERTAIN VASOACTIVE PEPTIDES ON PATHOGENESIS OF VASCULAR RESTENOSIS

Objective.To investigate the effects of several vasoactive peptides on the development of arterial restenosis after balloon angioplasty. Methods. In rat aortic artery restenosis model produced by denudation of aortic endothelia,we observed changes of endothelin(ET),angiotensin II(AII),calcitonin gene-related peptide(CGRP)and adrenomedullin(Adm)in plasma and aorta with radioimmunoassay and expression of hypertension-related gene(HRG-1)with semi-quantitative RT-PCR,and studied the effects of these peptides on intimal hyperplasia,intima/media ratio and MAPK activities of aortic artery after angioplasty respectively. Furthermore,in cultured cells,we studied the effects of these peptides on vascular smooth muscle cell(VSMC) proliferation and expression of HRG-1 of VSMC from spontaneously hypertensive rats(SHR)and Wistar-Kyoto(WKY)rats with 3H-TdR incorporation and RT-PCR respectively. Results. After angioplasty,the levels of ET and AII in plasma and aorta significantly increased,accompanied with VSMC proliferation and neointima hyperplasia. On day 10 after angioplasty,the levels of ET in plasma and aorta increased by 69% and 124% respectively,compared with sham group(P<0.01);and the level of aortic AII increased by 80%(P< 0.01). Antiserum against ET or inhibitors of angiotensin converting enzyme(ACE)could significantly inhibit the proliferation of VSMC and neointima formation. Compared with the sham group,on day 3 after angioplasty,the CGRP levels in plasma and aorta increased by 64% and 89% respectively(P< 0.01)and the Adm levels in plasma and tissue increased by 129% and 102% respectively(P< 0.01). On day 10,intravenous administration of CGRP significantly inhibited the proliferation of VSMC and neointima forma-tion induced by balloon aortic injury(by 66% and 79% respectively,P< 0.01). In addition,ET and AII attenuated the expression of HRG-1 in aorta and stimulated mitogen-activated protein kinase(MAPK)activity,while CGRP and Adm potentiated the expression of HRG-1 and inhibited MAPK.Conclusions. Postnatal heart maturation Is the basis of normal cardiac function and provides critical insights into heart repair and regenerative medicine.While static snapshots of the maturing heart have provided much Insight into its molecular signatures,few key events during postnatal cardiomyocyte maturation have been uncovered.Here,we report that cardiomyocytes(CMs)experience epige-netic and transcriptional decline of cardiac gene expression immediately after birth,leading to a transi-tion state of CMs at postnatal day 7(P7)that was essential for CM subtype specification during heart maturation.Large-scale single-cell analysis and genetic lineage tracing confirm the presence of transition state CMs at P7 bridging immature state and mature states.Silencing of key transcription factor JUN In P1-hearts significantly repressed CM transition,resulting in per-turbed CM subtype proportions and reduced cardiac function in mature hearts.In addition,transplantation of P7-CMs into infarcted hearts exhibited cardiac repair potential superior to P1-CMs.Collectively,our data uncover CM state transition as a key event in postnatal heart maturation,which not only provides insights into molecular foundations of heart maturation,but also opens an avenue for manipulation of cardiomyocyte fate in disease and regenerative medicine.

The concentration of biomarkers in sweat can be used to evaluate human health,making efficient sweat sensing a focus of research.While flow channel design is often used to detect sweat velocity,it is rarely incorporated into the sensing of biomarkers,limiting the richness of sensing results.In this study,we report a time sequential sensing scheme for uric acid in sweat through a sequential design of Tesla valve channels.Graphene electrodes for detecting uric acid and directional Tesla valve flow channels were fabricated using laser engraving technology to realize time sequential sensing.The performance of the channels was verified through simulation.The time sequential detection of uric acid concentration in sweat can help researchers improve the establishment of human health management systems through flexible wearable devices.

The developmental origin,anatomical location,and other factors contribute to aortic heterogeneity in a physiological state.On this basis,vascular diseases occur at different ratios based on position specificity,even with the same risk factor.However,the continuous intersegmental aortic profile has been rarely reported at the single-cell level.To reveal aortic heterogeneity,we identified 15 cell subtypes from five continuous aortic segments by marker genes and functional definitions.

A potential impact of A Disintegrin and Metalloproteinase Domain-Like Protein Decysin-1(ADAMDEC1)on clear cell renal cell carcinoma propagation

Clear cell renal cell carcinoma(KIRC)is the most common and aggressivemalignancy subtype of renal neoplasm that arises from proximal convoluted tubules.It is characterized by poor clinical outcomes and high mortality of patients due to the lack of specific biomarkers for varying stages of the disease and no effective treatment.Proteases are associated with the development of several malignant tumors in humans by their ability to degrade extracellular matrices,facilitating metastasis.Herein,differentially expressed genes in KIRC cases compared to healthy kidneys were screened out from the Gene Expression Profiling Interactive Analysis(GEPIA)database.This data was applied to determine the most elevated protease in KIRC and as a result,A Disintegrin and Metalloproteinase Domain-Like Protein Decysin-1(ADAMDEC1)was selected.This expression pattern was exclusive for KIRC and not observed for papillary and chromophobe renal cell carcinomas,in which ADAMDEC1 was at the same level in tumors and non-cancer specimens.Furthermore,the ADAMDEC1 significant increase was detected in the fourteen other human malignancies compared to healthy samples,which suggested its strong involvement in cancer development.Next,GEPIA and Pathology Atlas correlated ADAMDEC1 high expression with more advanced tumor grade and shorter survival of KIRC patients.Xena Functional Genomics Explorer presented that ADAMDEC1 could be hypermethylated in some tumor cases and one somatic mutation in the gene sequence was detected.Finally,a Search Tool for the Retrieval of Interacting Genes/Proteins;STRING base was utilized to predict the interactions of ADAMDEC1 with other molecules and construct the signaling network.In summary,ADAMDEC1 showed the tremendous potential to be the predictive marker for the KIRC and its development.Therefore,this review with data analysis can be a good base for further in vitro and in vivo research that experimentally can confirm the ADAMDEC1 as prognostic biomarkers and therapeutic target of KIRC.
