Protective effects of two Lactobacillus plantarum strains in hyperlipidemic mice

AIM:To investigate the effects of Lactobacillus plantarum(L.plantarum) CAI6 and L.plantarum SC4 on hyperlipidemic mice.METHODS:Male Kunming mice were fed a highcholesterol diet for 28 d to construct hyperlipidemic models.Hyperlipidemic mice and normal mice were assigned to 3 groups which were separately treated with L.plantarum CAI6,L.plantarum SC4,and physiological saline through oral gavage for 28 d.Total cholesterol(TC),triglycerides(TG),high-density lipoprotein cholesterol(HDL-C),and low-density lipoprotein cholesterol(LDL-C) levels were measured by commercially available enzyme kits.FACS Calibur flow cytometry was used to examine hepatic and renal nuclear factor-erythroid 2-related factor 2(Nrf2) expression.The morphology of livers was checked by hematoxylin and eosin staining and optical microscope observation.RESULTS:Compared with normal mice,hyperlipidemic mice possessed significantly higher TC(3.50 ± 0.43 vs 2.89 ± 0.36,P < 0.01),TG(1.76 ± 0.07 vs 1.10 ± 0.16,P < 0.01),and LDL-C(1.72 ± 0.20 vs 0.82 ± 0.10,P < 0.01) levels,resulting in an increase of atherogenic index(AI)(2.34 ± 1.60 vs 0.93 ± 0.55,P < 0.05) and LDL-C/HDL-C ratio(1.43 ± 0.12 vs 0.51 ± 0.16,P < 0.05).After treatment with L.plantarum CAI6/L.plantarum SC4,TG(1.43 ± 0.27/1.54 ± 0.10 vs 1.76 ± 0.07,P < 0.01/P < 0.05) and LDL-C(1.42 ± 0.07/1.47 ± 0.12 vs 1.72 ± 0.20,P < 0.01/P < 0.01) in hyperlipidemic mice significantly decreased.In addition,TC,HDL-C,AI,and LDL-C/HDL-C ratio were all positively changed.Meanwhile,the treatment markedly alleviated hepatic steatosis and significantly stimulated Nrf2 expression(73.79 ± 0.80/72.96 ± 1.22 vs 54.94 ± 1.84,P < 0.01/P < 0.01) in hepatocytes of hyperlipidemic mice.CONCLUSION:L.plantarum CAI6 and L.plantarum SC4 may protect against cardiovascular disease by lipid metabolism regulation and Nrf2-induced antioxidative defense in hyperlipidemic mice.

Is tumor cell specificity distinct from tumor selectivity in vivo?A quantitative NIR molecular imaging analysis of nanoliposome targeting

The significance and ability for receptor targeted nanoliposomes(tNLs)to bind to their molecular targets in solid tumors in vivo has been questioned,particularly as the efficiency of their tumor accumulation and selectivity is not always predictive of their efficacy or molecular specificity.This study presents,for the first time,in situ near-infrared(NIR)molecular imaging-based quantitation of the in vivo specificity of tNLs for their target receptors,as opposed to tumor selectivity,which includes influences of enhanced tumor permeability and retention.Results show that neither tumor delivery nor selectivity(tumor-to-normal ratio)of cetuximab and IRDye conjugated tNLs correlate with epidermal growth factor receptor(EGFR)expression in U251,U87,and 9L tumors,and in fact underrepresent their imaging-derived molecular specificity by up to 94.2%.Conversely,their in vivo specificity,which we quantify as the concentration of tNL-reported tumor EGFR provided by NIR molecular imaging,correlates positively with EGFR expression levels in vitro and ex vivo(Pearson’s r=0.92 and 0.96,respectively).This study provides a unique opportunity to address the problematic disconnect between tNL synthesis and in vivo specificity.The findings encourage their continued adoption as platforms for precision medicine,and facilitates intelligent synthesis and patient customization in order to improve safety profiles and therapeutic outcomes.

Failure investigation on high velocity impact deformation of boron carbide(B_(4)C) reinforced fiber metal laminates of titanium/glass fiber reinforced polymer

High velocity ballistic impact deformation behaviour of Titanium/GFRP Fiber Metal Laminates(FML)has been explored.Both single and multiple projectiles impact conditions were considered.Ti/GFRP FML targets were fabricated with addition of 5%and 10%weight percentage of boron carbide(B_(4)C)particles.Mechanical properties of Ti/GFRP FML targets were determined as per ASTM standards.High velocity ballistic experiments were conducted using armour Piercing Projectile(APP)of diameter 7.62 mm and velocity ranging between 350 and 450 m/s.Depth of penetration of the projectile into the target was measured.The deformation behaviour of Ti/GFRP targets with and without the presence of ceramic powder(B_(4)C)was investigated.“Ductile hole growth”failure mode was observed for pure GFRP target when subjected to single projectile impact whereas“plugging”failure mode was noted for Ti/GFRP targets.The presence of B_(4)C(5%by weight)particles has significantly improved the ballistic resistance of the Ti/GFRP FML target by offering frictional resistance to the projectile penetration.Further addition(10%by weight)of B_(4)C has reduced the ballistic performance due to agglomeration.None of the targets showed‘brittle cracking’or‘fragmentation’failures.When compared to the published results of Aluminium(Al 1100/GFRP and Al 6061/GFRP)FMLs,Ti/GFRP FML showed lesser DoP which increases its potential application to aerospace industry.

采用搅拌摩擦焊(FSW)工艺制备AZ31B镁合金焊接接头,并在不同条件下进行热处理。研究AZ31B镁合金焊后热处理(PWHT)不同区域的抗弹行为,使用7.62 mm×39 mm穿甲弹,冲击速度为(430±20)m/s。分析热处理前后搅拌摩擦焊接头的显微硬度。结果表明,PWHT工艺(250°C,1 h)能提高热处理后搅拌摩擦焊接头的显微硬度。扫描电镜(SEM)结果显示,热处理使α-Mg晶粒细化,形成细小的析出相,使β-Mg17Al12相溶解到Mg基体中。通过穿深(DOP)试验评估PWHT不同区域的抗弹行为。热处理后接头母材区(BMZ)的DOP值较小。抗弹试验后对弹坑周边3个区域的横截面进行SEM表征,观察到绝热剪切带(ASB)的形成。

Since the carbon dioxide(CO_(2))capture using solid sorbent is a reversible reaction,the solid sorbent can be regenerated by the desorption process.Therefore,the desorption process is one of the key important processes for the CO_(2)capture system.Traditionally,most of the literature studies focus on the desorption of solid sorbent under an N_(2)atmosphere.However,the desorption process of the solid sorbent is inappropriate in the real system because the system will need another process to separate CO_(2)and nitrogen(N_(2))after the desorption process.This study focused on the CO_(2)desorption of potassium carbonate supported on gamma-alumina(K2CO3/γ-Al_(2)O_(3))in a wet fluidized bed under a steam atmosphere by using the multiphase computational fluid dynamics(CFD)simulation.The effects of water thickness and dry restitution coefficient on CO_(2)desorption rate were investigated to provide a realistic particle collision behavior and to explore their effects on CO_(2)desorption phenomena.Moreover,the effect of steam velocity on the hydrodynamic behaviors of fluidization which on CO_(2)desorption rate was studied.The simulated results demonstrated that all the parameters,water thickness,dry restitution coefficient,and steam velocity had significantly affected system hydrodynamics and CO_(2)desorption rate in the wet fluidization desorption process.Furthermore,the effect of desorption temperature on CO_(2)desorption rate was evaluated for finding the appropriate temperature for CO_(2)desorption process of K2CO3/γ-Al_(2)O_(3).The results showed that the appropriate desorption temperature for CO_(2)desorption under steam atmosphere was the temperature over 150℃.

Laser cut hole matrices in novel armour plate steel for appliqué battlefield vehicle protection

During this research, experimental rolled homogeneous armour steel was cast, annealed and laser cut to form an appliqué plate. This Martensitic–Bainitic microstructure steel grade was used to test a novel means of engineering lightweight armour. It was determined that a laser cutting speed of 1200 mm/min produced optimum hole formations with limited distortion. The array of holes acts as a double-edged solution, in that they provide weight saving of 45%, providing a protective advantage and increasing the surface area. Data collected were used to generate laser cut-edge hole projections in order to identify the optimum cutting speed, edge condition, cost and deformation performance. These parameters resulted in the generation of a surface, with less stress raising features. This can result in a distribution of stress across the wider surface. Provided that appropriate process parameters are used to generate laser cut edges, then the hardness properties of the surface can be controlled. This is due to compressive residual stresses produced in the near edge region as a result of metallurgical transformations. This way the traverse cutting speed parameter can be adjusted to alter critical surface characteristics and microstructural properties in close proximity to the cut-edge. A relationship was identified between the width of the laser HAZ and the hardness of the cut edge. It is the thickness of the HAZ that is affected by the laser process parameters which can be manipulated with adjusting the traverse cutting speed.

It is well-known that optimizing the wheel system of lunar rovers is essential.However,this is a difficult task due to the complex terrain of the moon and limited resources onboard lunar rovers.In this study,an experimental prototype was set up to analyze the existing mechanical design of a lunar rover and improve its performance.First,a new vane-telescopic walking wheel was proposed for the lunar rover with a positive and negative quadrangle suspension,considering the complex terrain of the moon.Next,the performance was optimized under the limitations of preserving the slope passage and minimizing power consumption.This was achieved via analysis of the wheel force during movement.Finally,the effectiveness of the proposed method was demonstrated by several simulation experiments.The newly designed wheel can protrude on demand and reduce energy consumption;it can be used as a reference for lunar rover development engineering in China.

Analytical and Numerical Modeling for Flexible Pipes

The unbonded flexible pipe of eight layers, in which all the layers except the carcass layer are assumed to have isotropic properties, has been analyzed. Specifically, the carcass layer shows the orthotropic characteristics. The effective elastic moduli of the carcass layer have been developed in terms of the influence of deformation to stiffness. With consideration of the effective elastic moduli, the structure can be properly analyzed. Also the relative movements of tendons and relative displacements of wires in helical armour layer have been investigated. A three-dimensional nonlinear finite element model has been presented to predict the response of flexible pipes under axial force and torque. Further, the friction and contact of interlayer have been considered. Comparison between the finite element model and experimental results obtained in literature has been given and discussed, which might provide practical and technical support for the application of unbonded flexible pipes.

Printability——A key issue in extrusion-based bioprinting

Three-dimensional(3D) extrusion-based bioprinting is widely used in tissue engineering and regenerative medicine to create cell-incorporated constructs or scaffolds based on the extrusion technique.One critical issue in 3D extrusion-based bioprinting is printability or the capability to form and maintain reproducible 3D scaffolds from bioink(a mixture of biomaterials and cells).Research shows that printability can be affected by many factors or parameters,including those associated with the bioink,printing process,and scaffold design,but these are far from certain.This review highlights recent developments in the printability assessment of extrusion-based bioprinting with a focus on the definition of printability,printability measurements and characterization,and printability-affecting factors.Key issues and challenges related to printability are also identified and discussed,along with approaches or strategies for improving printability in extrusion-based bioprinting.

Characterization of localized corrosion pathways in 2195-T8 Al-Li alloys exposed to acidic solution

The corrosion properties of aluminum-lithium(Al-Li) alloys, which are potential materials used to construct for tanks of liquid rockets or missiles, are essential for safe propellant storage and transport. In order to manifest the corrosion resistance of the 2195 Al-Li alloy in practical propellant tanks filled with N2O4, the alloy was soaked in 30% nitric acid solution, an accelerating corrosion environment, to test its corrosion behavior. Scanning electron microscopy(SEM) and transmission electron microscopy(TEM)were used to characterize microstructure and corrosion morphology of the alloy. Focused ion beam(FIB),combined with SEM, was used to demonstrate localized corrosion features and the propagation of corrosion pathways beneath the alloy surface. It was found that the corrosion network was formed with most intergranular corrosion and sparse intragranular corrosion. Additionally, the distribution and number of intermetallic particles influenced the localized corrosion degree and the direction of corrosion pathways. Aggregated particles made corrosion pathways continuously and caused more severe corrosion. The results from this work were valid and useful to corrosion prevention and protection for storage safety on propellant tanks in N_(2)O_(4).
