Pathological bone loss is caused by an imbalance between bone formation and resorption.The bone microenvironments are hypoxic,and hypoxia-inducible factor (HIF) is known to play notable roles in bone remodeling.However,the relevant functions of HIF-2α are not well understood.Here,we have shown that HIF-2α deficiency in mice enhances bone mass through its effects on the differentiation of osteoblasts and osteoclasts.In vitro analyses revealed that HIF-2α inhibits osteoblast differentiation by targeting Twist2 and stimulates RANKL-induced osteoclastogenesis via regulation of Traf6.In addition,HIF-2α appears to contribute to the crosstalk between osteoblasts and osteoclasts by directly targeting RANKL in osteoprogenitor cells.Experiments performed with osteoblast- and osteoclast-specific conditional knockout mice supported a role of HIF-2α in this crosstalk.HIF-2α deficiency alleviated ovariectomy-induced bone loss in mice,and specific inhibition of HIF-2α with ZINC04179524 significantly blocked RANKLmediated osteoclastogenesis.Collectively,our results suggest that HIF-2α functions as a catabolic regulator in bone remodeling,which is critical for the maintenance of bone homeostasis.

Copper-catalyzed intermolecular oxidative-etherification type dearomatization reaction of 2-naphthols was developed.With air as the terminal oxidant,the reaction proceeded in excellent yields under mild conditions.In addition,the reaction between two different naphthol substrates occurred smoothly.A series of multifunctionalized β-naphthalenones,important scaffold existed widely in natural products and biologically active molecules,were synthesized efficiently.

Summary of main observation and conclusion An effective approach relying on a Lewis acid-or Brφnsted acid-assisted gold(Ⅰ)-catalyzed glycosylation has been reported in the synthesis of a panel of the representative natural spirostanol saponins,namely polyphyllin D (1),polyphyllin V (2),dioscin (3),formosanin C (4),and a derivative of polyphyllin D bearing a terminal azide group (5).This approach highlights the engagement of low Ioadings of Ph3AuPOTf (≤ 0.5mol%) in the presence of Ga(OTf)3,In(OTf)3,or HOTf (~10 mol%) as a co-catalyst,at practical reaction rates as an alternative to the conventional ~10 mol%Ioadings of the gold(Ⅰ) catalyst in the glycosylation.Polyphyllin D (1) was obtained in 41% overall yield over six steps compared to the maximum 30% yield in previous syntheses,where conventional donors and promoters were used.By exploiting a regioselective rhamnosylation,a "one-pot" approach was adopted to assemble 1 and 5,thus further strengthening the efficiency of the gold(Ⅰ)-catalyzed glycosylation.

Growing evidence suggests close associations between periodontitis and atherosclerosis.To further understand the pathological relationships of these associations,we developed periodontitis with ligature placement around maxillary molars or ligature placement in conjunction with Porphyromonas gingivalis lipopolysaccharide injection at the ligature sites (ligature/P.g.LPS) in Apolipoprotein E knock out mice and studied the atherogenesis process in these animals.The mice were fed with high fat diet for 11 weeks and sacrificed for analyzing periodontitis,systemic inflammation,and atherosclerosis.Controls did not develop periodontitis or systemic inflammation and had minimal lipid deposition in the aortas,but mice receiving ligature or ligature/P.g.LPS showed severe periodontitis,systemic inflammation,and aortic plaque formation.The aortic plaque contained abundant macrophages and cells expressing both endothelial and mesenchymal cell markers.The severity of periodontitis was slightly higher in mice receiving ligature/P.g.LPS than ligature alone,and the magnitude of systemic inflammation and aortic plaque formation were also notably greater in the mice with ligature/P.g.LPS.These observations indicate that the development of atherosclerosis is due to systemic inflammation caused by severe periodontitis.In vitro,P.g.LPS enhanced the secretion of pro-inflammatory cytokines from macrophages and increased the adhesion of monocytes to endothelial cells by upregulating the expression of adhesion molecules from endothelial cells.Moreover,secretory proteins,such as TNF-α,from macrophages induced endothelial–mesenchymal transitions of the endothelial cells.Taken together,systemic inflammation induced by severe periodontitis might exacerbate atherosclerosis via,in part,causing aberrant functions of vascular endothelial cells and the activation of macrophages in mice.

This paper deals with the output regulation problem for a class of SISO infinite dimensional systems with an uncertain exosystem.For these systems,a concept of relative degree is firstly introduced and used to construct a transformation which leads to the canonical form of output feedback systems.Then,based on this canonical form,by means of an internal model and a recursive adaptive control,the authors obtain an adaptive regulator which solves the problem.It should be pointed out that the proposed regulator is finite dimensional while it is usually infinite dimensional in existing literatures.

Bismuth(Bi)-doped photonic materials, which exhibit broadband near-infrared(NIR) luminescence(1000–1600 nm), are evolving into interesting gain media. However, the traditional methods have shown their limitations in enhancing Bi NIR emission, especially in the microregion. Consequently, the typical NIR emission has seldom been achieved in Bi-doped waveguides, which highly restricts the application of Bi-activated materials.Here, superbroadband Bi NIR emission is induced in situ instantly in the grating region by a femtosecond(fs)laser inside borosilicate glasses. A series of structural and spectroscopic characterizations are summoned to probe the generation mechanism. And we show how this novel NIR emission in the grating region can be enhanced significantly and erased reversibly. Furthermore, we successfully demonstrate Bi-activated optical waveguides.These results present new insights into Bi-doped materials and push the development of broadband waveguide amplification.

The dynamic linear state feedback control problem is addressed for a class of nonlinear systems subject to time-delay.First,using the dynamic change of coordinates,the problem of global state feedback stabilization is solved for a class of time-delay systems under a type of nonhomogeneous growth conditions.With the aid of an appropriate Lyapunov-Krasovskii functional and the adaptive strategy used in coordinates,the closed-loop system can be globally asymptotically stabilized by the dynamic linear state feedback controller.The growth condition in perturbations are more general than that in the existing results.The correctness of the theoretical results are illustrated with an academic simulation example.

This paper investigates the observabihty of free Boolean networks by using the semi-tensor product method,and presents some new results.First,the concept of observability for free Boolean networks is proposed,based on which and the algebraic form of Boolean networks,a kind of observabihty matrix is constructed.Second,by the observability matrix,a new necessary and sufficient condition is given for the observability of Boolean networks.Third,the concept of observabihty index for observable Boolean networks is defined,and an algorithm is established to calculate the observability index.Finally,a practical example of D.Melanogaster segmentation polarity gene networks is studied to support our new results.The study of the illustrative example shows that the new results obtained in this paper are very effective in investigating the observability of free Boolean networks.
