Objective Meninges which surround the center nerve system consist of pia mater, arachnoid membrane and dura mater. Pervious research showed that these membranes work not only for a simple protection but also to keep the brain condition. It is reported that neural progenitors can be activated in the cortical surface adjacent to the injured region and leptomeninges（include the pia mater and arachnoid membrane）, and the cells sorted from the leptomeninges can form neurosphere-like clusters with selfrenewal activity in vitro. All these studies indicated that leptomeninges may be a novel stem cell niche. Some researchers thought that the microvascular pericytes may be the potential sources of neural progenitors. However, here we report that the neural progenitors may be derived from the ventricular zone as other nevour system. Methods The existence of potential neural progenitors in the leptomeninges was identified by different progenitor makers and Brd U label. And the origin of these neural progenitors was shown by in utero electroporation. Results（1） The ENKergic neurons born at E11.5 can be found near the leptomeninges of spinal cord during development stages;（2） Granule cells in the leptomeninges express or transcient express PV, GAD67 and Tbr2 during development;（3） 24 h after electroporation, the GFP-positive cells mainly located near the SVZ, but near the surface of the neocortex, there are also some GFP-positive cells. Conclusion Neural progenitors in the leptomeninges derived from the ventricular zone during development.

Heterozygous loss-of-function variants of FOXP4 are associated with neurodevelopmental disorders(NDDs)that exhibit delayed speech development,intellectual disability,and congenital *** etiology of NDDs is *** we found that FOXP4 and N-cadherin are expressed in the nuclei and apical end-feet of radial glial cells(RGCs),respectively,in the mouse neocortex during early *** or dominant-negative inhibition of Foxp4 abolishes the apical condensation of N-cadherin in RGCs and the integrity of neuroepithelium in the ventricular zone(VZ).Inhibition of Foxp4 leads to impeded radial migration of cortical neurons and ectopic neurogenesis from the proliferating *** ectopic differentiation and deficient migration disappear when N-cadherin is over-expressed in *** data indicate that Foxp4 is essential for N-cadherin-based adherens junctions,the loss of which leads to periventricular *** hypothesize that FOXP4 variant-associated NDDs may be caused by disruption of the adherens junctions and malformation of the cerebral cortex.

Perspectives on the role of Pannexin 1 in neural precursor cell biology

We recently reported that targeted deletion of Pannexin 1 in neural precursor cells of the ventricular zone impairs the maintenance of these cells in healthy and stroke-injured brain. Here we frame this exciting new finding in the context of our previous studies on Pannexin 1 in neural precursors as well as the close rela- tionship between Pannexin 1 and purinergic receptors established by other groups. Moreover, we identify important gaps in our understanding of Pannexin 1 in neural precursor cell biology in terms of the under- lying molecular mechanisms and functional/behavioural outcomes.
