Neuron progenitors in the leptomeninges derived from the ventricular zone during development

摘      要:Objective Meninges which surround the center nerve system consist of pia mater, arachnoid membrane and dura mater. Pervious research showed that these membranes work not only for a simple protection but also to keep the brain condition. It is reported that neural progenitors can be activated in the cortical surface adjacent to the injured region and leptomeninges(include the pia mater and arachnoid membrane), and the cells sorted from the leptomeninges can form neurosphere-like clusters with selfrenewal activity in vitro. All these studies indicated that leptomeninges may be a novel stem cell niche. Some researchers thought that the microvascular pericytes may be the potential sources of neural progenitors. However, here we report that the neural progenitors may be derived from the ventricular zone as other nevour system. Methods The existence of potential neural progenitors in the leptomeninges was identified by different progenitor makers and Brd U label. And the origin of these neural progenitors was shown by in utero electroporation. Results(1) The ENKergic neurons born at E11.5 can be found near the leptomeninges of spinal cord during development stages;(2) Granule cells in the leptomeninges express or transcient express PV, GAD67 and Tbr2 during development;(3) 24 h after electroporation, the GFP-positive cells mainly located near the SVZ, but near the surface of the neocortex, there are also some GFP-positive cells. Conclusion Neural progenitors in the leptomeninges derived from the ventricular zone during development.
