Effects of La, Ce, Y and Tb ions on Free Radical Mediated Peroxidation of Human erythrocyte Membrane

The effects of the lanthanides in various concentration on the peroxidation of 'ghost' of human erythrocyte with free radicals were studied by methods of fluorometry, spectrophotometry and electron paramagnetic resonance. It is shown that La 3+ and Ce 3+ in the range of 2×10 -4 ～2×10 -7 mol·L -1 inhibit the t BHP mediated peroxidation significantly. Tb 3+ and Y 3+ inhibit the t BHP mediated peroxidation in low concentration (<2×10 -5 mol·L -1 ), but promotes the peroxidation when the concentration is higher than 2×10 -5 mol·L -1 .

Liposome,a widely used drug delivery system(DDS),still shows several disadvantages such as dominant clearance by liver and poor target organ *** overcome the drawbacks of liposomes,we developed a novel red blood cell(RBC)-liposome combined DDS to modulate the tumor accumulation and extend the blood circulation life of the existing liposomal ***,RBCs,an ideal natural carrier DDS,were utilized to carry liposomes and avoid them undergo the fast clearance in the *** this study,liposomes could either absorbed onto RBCs’surface or fuse with RBCs’membrane by merely altering the interaction time at 37℃,while the interaction between liposome and RBCs would not affect RBCs’*** the in vivo antitumor therapeutic efficacy study,1,2-dipalmitoyl-sn-glycero-3-phosphocholine(DPPC)liposomes attached onto RBCs’surfaces exhibited lung targeting effect(via RBC-hitchhiking approach)and reduced clearance in the liver,while DPPC liposomes fused with RBCs had prolong blood circulation up to 48 h and no enrichment in any ***,20 mol%of DPPC liposomes were replaced with pH-sensitive phospholipid 1,2-dioleoyl-Sn-glycero-3-phosphoethanolamine(DOPE)as it could respond to the low pH tumor microenvironment and then accumulate in the *** DOPE attached/fusion RBCs showed partial enrichment in lung and about 5-8%tumor accumulation,which were significantly higher than(about 0.7%)the conventional liposomal ***,RBC-liposome composite DDS is able to improve the liposomal tumor accumulation and blood circulation and shows the clinical application promises of using autologous RBCs for antitumor therapy.

The effects of vanadate on human erythrocyte membrane have been investigated with stopped-flow and equilibrium fluorescence quenching techniques. The equilibrium study showed a half-quenching concentration(K 1/2 ) of 0.27 mmol.L-1. The stopped-flow experiment exhibited a fast rise (tsk1/f1 S) and a slow drop (tin 1~2 min) in fluorescence. Based on the results and that from the across membrane transport of vanadate, a kinetic model is proposed which suggests that the membrane proteins experience a series of conformational changes before and during the quenching of the intrinsic fluorescence. These changes are induced mainly by three kinds of interactions: (i) the long-distance, non-specific interaction between the vanadate and the erythrocyte membrane surface, (ii) the charge interaction between the vanadate and parts of the membrane proteins, and(iii) the binding of the vanadate to some membrane proteins.

Laser Raman spectroscopy was used to investigate the photodamage characteristics of human erythrocyte membranes sensitized by hypocrellin B (HB) at the molecular level. It brought to light that the essence of the changes of erythrocyte membranes’ functions caused by membrane protein cross linking and membrane lipid peroxidation, including increase of fluidity and ion permeability of membranes, etc., was that the orderly structure of erythrocyte membranes had been damaged by the active oxygen ( 1O 2, O 2 .- and .OH) generated by HB, including the decrease of α helix, β sheet and the increase of random coil in the main chain of membrane proteins, the decrease of mercapto groups, indole rings, p hydroxy phenyl rings, monosubstituted phenyl rings, etc. in the side chain, and the changes of the conformations of membrane lipids as well. With the increase of the irradiation time, the trans conformation of membrane lipids increased first, then decreased. On the contrary, the gauche conformation decreased first, then increased. In addition, the decrease of the intensities of the lines assigned to bending vibration of the conformation insensitive CH 2 and CH 3 of membrane proteins and lipids suggested that break of their chains had occurred.

OBJECTIVE: To show whether Danggui Sini plus Wuzhuyu Shengjiang Tang(DSWST) has any transient effect on erythrocyte deformability in normal ***: A total of 25 subjects [mean age(27.8± 1.8) years] was enrolled in this study. The study was designed as a cross-over trial in which the subjects took part for 2 d. On the first day, blood samples were collected at baseline and 1-2 h after administration of water, whereas, on the second day, instead of water, the subjects were administered DSWST after the baseline blood sampling. The blood samples collected at baseline and after the administration water or DSWST, were examined for erythrocyte ***: The elongation index increased significantly after 2 h(P = 0.009) compared to the baseline after DSWST intake. However, after water intake, there was no significant differenceobserved. When comparing the percent change of erythrocyte deformability between DSWST and water, we found that after 2 h of administration,DSWST improved erythrocyte deformability significantly compared to water(P erythrocyte deformability in normal subjects.

Objective To assess the relationships between plasma and intracellular Ca^2+ , Mg^2+ and blood cell membrane adenosine triphosphatase (ATPase) activity in normotensive and hypertensive subjects. Methods Plasma and intracellular Ca^2+ , Mg^2+ were measured with atomic absorption spectrophotometry, and red blood cell membrane Na^+ K^+ ATPase and Ca 2+ ATPase activities were determined with colorimetric method in 55 patients with essential hypertension and 32 normotensive controls. Results The results showed that the hypertensive group consistently demonstrated a significant decreased activity of ATPase studied, with significantly lower plasma Ca 2+ and higher cytosolic Ca 2+ levels when compared with those in normotensive group (P< 0.01 or P<0.05, respectively). No significant differences were found in either plasma Mg 2+ or intracellular Mg 2+ level between the two groups. Conclusions This study suggests that patients with essential hypertension have widespread depression of cell membrane Na + K + ATPase and Ca 2+ ATPase activities with plasma Ca 2+ depletion and cytosolic Ca 2+ overload, which may reflect an underlying membrane abnormality in essential hypertension. The celluar abnormalities may be related to the defective transport mechanisms that in turn may be aggravated by plasma Ca^2+ depletion.

Folding patterns of chromatin in chicken erythrocyte studied by using AFM and TEM

Four folding patterns (～15, ～30-40, ～60 and ～90-110 nm) of the higher_order structure of chromatin in chicken erythrocytes have been observed with an atomic force microscope (AFM) under near natural conditions. Results showed that the chromatin in the interphase nucleus of chicken erythrocyte was not in a disorganized form but represented a higher_order conformation. The thin sections of the chromatin in the nucleus of chicken erythrocyte were examined with a transmission electron microscope (TEM). Compared with TEM, AFM had the advantages that the samples did not need the preparation procedures such as fixing, sectioning and staining, and thus were closer to their native condition.

Weinreb first reported the results of transmission electron microscopy observation on ultrastructure of goldfish erythrocyte in 1963. In recent years, there are few reports about the ultrastructure of blood cells in fish For further understanding the fine structure of mature erythrocyte in teleosts, using goldfish (Carassius auratus) as the experimental material We made an observation of the morphology of its mature erythrocyte by means of scanning electron microscopy as well as a study on the ultrastructure by freeze etching technique. Since there are none of reports concerning the study on erythrocyte in fish by using freeze etching technique, the present note will provide here a practical method and the data of studying about ultrastructure of erythrocyte in fish.
