Clinical significance,molecular characterization,and immune microenvironment analysis of coagulation-related genes in clear cell renal cell carcinoma

Background:Numerous studies have revealed a tight connection between tumor development and the coagulation system.However,the effects of coagulation on the prognosis and tumor microenvironment(TME)of clear cell renal cell carcinoma(ccRCC)remain poorly understood.Methods:We employed the consensus clustering method to characterize distinct molecular subtypes associated with coagulation patterns.Subsequently,we examined variations in the overall survival(OS),genomic profiles,and TME characteristics between these subtypes.To develop a prognostic coagulation-related risk score(CRRS)model,we utilized the least absolute shrinkage and selection operator Cox regression and stepwise multivariate Cox regression analyses.We also created a nomogram to aid in the clinical application of the risk score,evaluating the relationships between the CRRS and the immune microenvironment,responsiveness to immunotherapy,and targeted treatment.The clinical significance of PLAUR and its biological function in ccRCC were also further analyzed.Results:There were significant differences in clinical features,prognostic stratification,genomic variation,and TME characteristics between the two coagulation-related subtypes.We established and validated a CRRS using six coagulation-related genes that can be employed as an effective indicator of risk stratification and prognosis estimation for ccRCC patients.Significant variations in survival outcomes were observed between the high-and low-risk groups.The nomogram was proficient in predicting the 1-,3-,and 5-year OS.Additionally,the CRRS emerged as a novel tool for evaluating the clinical effectiveness of immunotherapy and targeted treatments in ccRCC.Moreover,we confirmed upregulated PLAUR expression in ccRCC samples that was significantly correlated with poor patient prognosis.PLAUR knockdown notably inhibited ccRCC cell proliferation and migration.Conclusion:Our data suggested that CRRS may be employed as a reliable predictive biomarker that can provide therapeutic benefits for immunotherapy and

In this study,samples were taken from three locations,upstream to downstream,along the central route project of the China South to North Water Diversion(SNWD)scheme in summer and winter.These were used to reveal the variations of dissolved organic matter(DOM)during the water transfer process,and the effects of these variations on drinking water treatment and disinfection by-products formation potential(DBPs-FP).The results showed that polysaccharides accumulate in summer and reduce in winter with flow distance,which has an important effect on the overall properties of DOM,as well as on the performance of coagulation,ultrafiltration,and the formation of DBPs.Humic substances,and their hydrophilic content,also increased in summer and decreased in winter with flow distance.In contrast,the concentration of small organic substances(MW⩽1000 Da)increased in both summer and winter with flow distance,which affected both nanofiltration(NF)membrane fouling and DBPs-FP.The results provide a useful case study of spatial and temporal changes in raw water DOM during long distance water transfer and their impact on the treatment and quality of drinking water from the SNWD.

Background:Hemodialysis(HD)per se is a risk factor for thrombosis.Considering the growing body of evidence on blood-flow restriction(BFR)exercise in HD patients,identification of possible risk factors related to the prothrombotic agent D-dimer is required for the safety and feasibility of this training model.The aim of the present study was to identify risk factors associated with higher D-dimer levels and to determine the acute effect of resistance exercise(RE)with BFR on this molecule.Methods:Two hundred and six HD patients volunteered for this study(all with a glomerular filtration rate of70 years old),and HD vintage.Furthermore,RE+BFR significantly increases D-dimer after 4 h.Patients with borderline baseline D-dimer levels(400-490 ng/mL)displayed increased risk of elevating D-dimer over the normal range(-500 ng/mL).Conclusion:These results identified factors associated with a heightened prothrombotic state and may assist in the screening process for HD patients who wish to undergo RE+BFR.D-dimer and/or other fibrinolysis factors should be assessed at baseline and throughout the protocol as a precautionary measure to maximize safety during RE+BFR.

Non-alcoholic fatty liver disease(NAFLD)is closely related to insulin resistance,type 2 diabetes mellitus,and obesity.It is nowadays considered a multisystem disease with a strong association with cardiovascular disease and arterial hypertension,which interfere with changes in the coagulation system.coagulation disorders are common in patients with hepatic impairment and are dependent on the degree of liver damage.Patients with NAFLD may have preserved overall hemostatic profile,but many studies suggest a trend toward a procoagulant state.Hypercoagulable state in NAFLD patients may even induce progression of hepatic injury.Endothelial dysfunction is present in the systemic and portal vein circulation in NAFLD patients,and platelets are being recognized as modulators of liver diseases through various mechanisms.Through a literature review,we discuss possible disorders in the coagulation cascade and fibrinolysis,endothelial dysfunction,and platelet abnormalities in patients with NAFLD.Considering the processes and mechanisms involved in the hemostatic abnormalities associated with NAFLD,directly related to liver disease or indirectly related through inflam-matory processes and metabolic disorders,several potential therapeutic targets have been identified and reviewed here.

Impaired coagulation,liver dysfunction and COVID-19:Discovering an intriguing relationship

Coronavirus disease 2019(COVID-19)is,at present,one of the most relevant global health problems.In the literature hepatic alterations have been described in COVID-19 patients,and they are mainly represented by worsening of underlying chronic liver disease leading to hepatic decompensation and liver failure with higher mortality.Several potential mechanisms used by severe acute respiratory syndrome coronavirus 2(SARS-Co V-2)to cause liver damage have been hypothesized.COVID-19 primary liver injury is less common than secondary liver injury.Most of the available data demonstrate how liver damage in SARSCo V-2 infection is likely due to systemic inflammation,and it is less likely mediated by a cytopathic effect directed on liver cells.Moreover,liver alterations could be caused by hypoxic injury and drugs(antibiotics and non-steroidal antiinflammatory drugs,remdesivir,tocilizumab,tofacitinib and dexamethasone).SARS-Co V-2 infection can induce multiple vascular district atherothrombosis by affecting simultaneously cerebral,coronary and peripheral vascular beds.Data in the literature highlight how the virus triggers an exaggerated immune response,which added to the cytopathic effect of the virus can induce endothelial damage and a prothrombotic dysregulation of hemostasis.This leads to a higher incidence of symptomatic and confirmed venous thrombosis and of pulmonary embolisms,especially in central,lobar or segmental pulmonary arteries,in COVID-19.There are currently fewer data for arterial thrombosis,while myocardial injury was identified in 7%-17%of patients hospitalized with SARS-Co V-2 infection and 22%-31%in the intensive care unit setting.Available data also revealed a higher occurrence of stroke and more serious forms of peripheral arterial disease in COVID-19 patients.Hemostasis dysregulation is observed during the COVID-19 course.Lower platelet count,mildly increased prothrombin time and increased Ddimer are typical laboratory features of patients with severe SARS-Co V-2 infection,described as“COVID-19 associa

Enhanced cross-flow filtration with flat-sheet ceramic membranes by titanium-based coagulation for membrane fouling control

Application of ceramic membrane(CM)with outstanding characteristics,such as high flux and chemical-resistance,is inevitably restricted by membrane fouling.coagulation was an economical and effective technology for membrane fouling control.This study investigated the filtration performance of ceramic membrane enhanced by the emerging titanium-based coagulant(polytitanium chloride,PTC).Particular attention was paid to the simulation of ceramic membrane fouling using four widely used mathematical models.Results show that filtration of the PTC-coagulated effluent using flat-sheet ceramic membrane achieved the removal of organic matter up to 78.0%.Permeate flux of ceramic membrane filtration reached 600 L/(m2$h),which was 10-fold higher than that observed with conventional polyaluminum chloride(PAC)case.For PTC,fouling of the ceramic membrane was attributed to the formation of cake layer,whereas for PAC,standard filtration/intermediate filtration(blocking of membrane pores)was also a key fouling mechanism.To sum up,cross-flow filtration with flat-sheet ceramic membranes could be significantly enhanced by titanium-based coagulation to produce both high-quality filtrate and high-permeation flux.

Tumor vascular infarction has been regarded as a promising therapy in tumor treatment.However,due to the limited activity of designed coagulation agents,resulting from the challenges of inducing sustained and complete thrombosis and associated selectivity issues,the traditional vascular infarction strategy consequently results in treatment failure.Here,we report a photo-initiated coagulation activation and fibrinolysis inhibition approach to achieve synergetic tumor vascular infarction via gold nanorods(AuNRs)-based nanosystem for the co-delivery of coagulation initiator thrombin and fibrinolysis inhibitor tranexamic acid.With a photo-initiated cascade reaction,thrombin and tranexamic acid could be released from the nanosystem to activate coagulation and inhibit the fibrinolysis within tumor vessels.By integrating these two physiological reactions,fortified clots were formed,and tumor vessels were occluded steadily,affecting the metabolism of the tumor cells,thereby inhibiting tumor growth.This strategy might open an alternative path for the advancement of the current tumor vascular infarction strategy.

coagulation enhanced electrokinetic settling of mature fine oil sands tailings

The mature fine oil sand tailings(MFT) remain as slurry in the tailings pond for long time. The dewatering and consolidation of MFT for sustainable management is an important task for the mining industry. The objective of this study is to accelerate electrokinetic settling of MFT solids in suspensions in presence of optimal coagulant. In the first phase, optimal coagulant and coagulant dosage for settling of suspension are identified, i.e., ferric chloride at 350 mg/L. It is found that the chemical treatment is not much effective; the final solid content of the sediment is only 6.48% from an initial of 5%. In the second phase,combined coagulation and electrokinetic treatment is carried out to enhance the settling effect. The direct electric current is applied in continuous and intermittent modes on MFT suspensions placed in electrokinetic cell. The results show the final solid content reaches 23.74% under the combined application of350 mg/L ferric chloride and 218.75 V/m applied voltage gradient in the continuous mode. The intermittent current mode with 40% save in power consumption produces a settled sediment having 20.84% final solid content.

coagulation and fibrinolytic activity in patients with acute cerebral infarction

Objective To measure the concentration of D-dimer (DD), tissue plasminogen activator (t-PA), plasminogen activator inhibitor-1 (PAI-1) and plasminogen (PLG) activity in plasma and cerebrospinal fluid in patients with acute cerebral infarction and to investigate their clinical significance.Methods The concentrations of D-dimer, t-PA, and PAI-1 in plasma and cerebrospinal fluid in patients were measured by enzyme-linked immunosorbent assay (ELISA). The PLG biological activity was detected using the chromophore method. The results were compared with those of the controls.Results The concentrations of D-dimer, t-PA and PAI-1 in cerebrospinal fluid and plasma in patients with acute cerebral infarction were much higher than those of normal subjects (P<0.01). Conversely, the level of PLG activity was significantly lower in the patients than in the controls (P<0.01).Conclusion Hypercoagulability and secondary hyperfibrinolysis exist in patients with acute cerebral infarction.

The influence of various additives on coagulation process at different dosing point:From a perspective of structure properties

Structure properties of flocs(size,fractal dimension(D_(f)),etc.)have a high impact on coagulation efficiency.In this work,the influences of three different additives(ferric salt(Fe),phosphate(P),and citric acid(CA))on coagulation process/efficiency were investigated.Results showed that a small amount of extra Fe can facilitate the growth of Al flocs by providing more'active sites'.Although zeta potential and D_(f) showed a limited change,the average floc size increased apparently and the increment was more obvious when Fe was added after the formation of the flocs.In contrast,P addition during the rapid mixing period will decrease the final average floc size,while the influence is less significant when P was added after the growth of the flocs.In terms of CA,a more striking negative effect on the growth ability of the flocs was observed compared to P.The strong complexing/coordination interactions between CA and aluminum hydroxide is the main reason behind the influence.CA also significantly decreased the D_(f) value of the flocs compared to P,and D_(f) showed a comparatively higher decrease when P or CA was added during the rapid mixing stage compared to the addition after the flocs formation.These results indicated that the addition of CA or P during the rapid mixing stage'inactivated'or occupied more'active sites'on the preliminarily formed Al NPs during the hydrolysis process,and therefore presented stronger impact on the morphology/size of the formed flocs.
