Interleukin-18 polymorphism as an inflammatory index in metabolic syndrome: A preliminary study

AIMTo assess circulatory levels of interleukin-18 (IL-18) and determine whether the presence of IL-18 promoter polymorphism influences metabolic syndrome phenotypes. METHODSThis study recruited one hundred and eighty individuals divided into three groups with sixty subjects each as: Normal weight (18.0-22.9 kg/m2), overweight (23.0-25.9 kg/m2) and obese (> 26.0 kg/m2) according to South Asian criteria of BMI. Fasting blood glucose (FBG), Lipid profile, insulin, IL-18 and tumor necrosis factor (TNF)α were measured using ELISA kits, whereas low density lipoprotein (LDL)-cholesterol, insulin resistance (HOMA-IR) and insulin sensitivity (QUICKI) were calculated. The body fat percentage (BF) was measured through bioelectrical impedance analysis; waist and hip circumference were measured. Genotyping of IL-18 -607 C/A polymorphism was performed by using tetra-primer amplification refractory mutation system. Student t test, One-way analysis of variance, Hardy-Weinberg equilibrium, Pearson’s χ2 test and Pearson’s correlation were used, where a P value < 0.05 was considered significant. RESULTSIn an aged matched study, obese subjects showed higher levels of FBG, cholesterol, triglycerides and LDL levels as compared to normal weight (P < 0.001). Highest levels of IL-18 and TNF levels were also seen in obese subjects (IL-18: 58.87 ± 8.59 ng/L) (TNF: 4581.93 ± 2132.05 pg/mL). The percentage of IL-18 -607 A/A polymorphism was higher in overweight and obese subjects vs normal weight subjects (P < 0.001). Moreover, subjects with AA genotype had a higher BF, insulin resistance, TNFα and IL-18 levels when compared with subjects with AC (heterozygous) or CC (wild type) genotypes. However, we did not find any difference in the lipid profile between three subgroups. CONCLUSIONThis preliminary data suggests that IL-18 polymorphism affects IL-18 levels that might cause low grade inflammation, further exacerbated by increased TNFα. All these incr

Since plant oils are believed to be better than animal fats for cerebrovascular and cardiovascular diseases, the effects of various plant oils and trans-fat on blood lipid profiles and ischemic stroke were investigated. Sprague- Dawley rats were fed a diet containing the oils or trans-fat, and then body weights, blood lipids, and effects on brain infarction and physical dysfunction induced by middle cerebral artery occlusion （MCAO） were analyzed. All the oils and trans-fat, except perilla oil, significantly increased body fats and body weight gain. Sesame oil and trans-fat specifically increased blood cholesterols and triglycerides, respectively, while perilla oil decreased both cholesterols and triglycerides. Perilla oil not only attenuated cerebral infarction, but also restored locomotor activity and rota-rod performances of MCAO rats. It is suggested that perilla oil among oils and fats could be the first choice to reduce the risk of metabolic syndrome and ischemic stroke.

Objective: Aim of this study was to investigate the changes in body composition and some biochemical parameters of women at different age and body mass index (BMI) groups and to evaluate their relations with to cardiometabolic risk factors. Methods: The study was created by retrospective research of the file records of total 800 women who got into nutritional counseling between 2002 - 2007 years at Cerrahpasa Medical Faculty. Participants were grouped as 10 - 19, 20 - 29, 30 - 39, 40 - 49, 50 - 59, ≥60 years according to ages and were divided into 4 groups as normal, overweight, obese and morbidly obese according to body mass index. Data were statistically analysed in SPSS for Windows computer program with ANOVA and Tukey HSD tests. Findings: There are significant differences between different age groups in weight, BMI, waist and hip circumferences, percentage of body water, fat and fat-free mass, basal metabolic rate and blood glucose, insulin, triglyceride, cholesterol, HDL, LDL levels and HOMA-IR values. HOMA-IR values were the highest at age group 1 and the lowest at age group 2. Although they were in the same BMI group;their waist circumference, basal metabolic rate and percent of body fat showed significant increases with advancing age. Conclusion: The findings of our study show that it will be useful to use standard values generated for each age group according to BMI levels in predicting increased cardiovascular and metabolic risks together with advancing age. While the lowest rate of obesity according to body fat percent (>30%) was found at the ages in between 20 - 29 (58.4%), it is noteworthy that the obesity rate with respect to their body fat percent has been noticed at the ages of 10 to 19 (2.3%). Comparing with the other groups, significantly higher body heights found at the ages in between 10 - 19, could possibly predicate that the new generations are having better nutrition and overhaul. Natural killer T cells(NKT cells) are innate-like T cells that acquire effector functions while developing in the thymus, polarize into three distinct functional subsets viz. NKT1, NKT2 and NKT17 cells that produce interferon(IFN)-γ, interleukin(IL)-4 and IL-17, respectively. However, there has been no unique surface markers that define each subsets, forcing investigators to use intracellular staining of transcription factors and cytokines in combination of surface markers to distinguish among these subsets. Intracellular staining, however, causes apoptosis and prevents subsequent utilization of NKT cells in functional in vitro and in vivo assays that require viable cells. This limitation has significantly impeded understanding the specific properties of each subset and their interactions with each other. Therefore, there has been fervent efforts to find a specific markers for each NKT cell subset. We have recently identified that syndecan-1(SDC-1; CD138) as a specific surface marker of NKT17 cells. This discovery now allows visualization of NKT17 in situ and study of their peripheral tissue distribution, characteristics of their TCR and viable sorting for in vitro and in vivo analysis. In addition, it lays the ground working for investigating significance of SDC-1 expression on this particular subset in regulating their roles in host defense and glucose metabolism.
