urotensin ii RECEPTOR IN THE RAT AIRWAY SMOOTH MUSCLE AND ITS EFFECT ON THE RAT AIRWAY SMOOTH MUSCLE CELLS PROLIFERATION

Objective. To investigate the characteristics of urotensin ii (U ii) receptor in the rat airway smooth muscle and the effect and signal transduction pathway of U ii on the proliferation of airway smooth muscle cells. Methods. Using 125I Uii binding assay to measure the Bmax and Kd of U ii receptor. Using the 3H TdR incorporation to determine the effect of U ii on the proliferation of airway smooth muscle cells and its signal transduction pathway. Using Fura 2/AM to measure the effect of U ii on the cytosolic free calcium concentration. Results. 1. 125I Uii binding increased with the time and reached saturation at 45min. The Bmax was (11.36±0.37)fmol/mg pr and Kd was (4.46±0.61)nmol/L. 2. U ii increased 3H TdR incorporation of the airway smooth muscle cells in a dose dependent manner. 3. H7, PD98059 and nicardipine, inhibitors of PKC, MAPK, calcium channel, respectively, significantly inhibited U ii stimulated 3H TdR incorporation of airway smooth muscle cells. W7, inhibitor of CaM PK, had no effect. 4. Cyclosporin A, inhibitor of CaN, inhibited 3H TdR incorporation of the airway smooth muscle cells induced by U ii in a dose dependent manner. 5. U ii promoted cytosolic free calcium concentration increase by 18%. Conclusions. 1. There was U ii receptor in the rat airway smooth muscle. 2. The effect of U ii stimulated 3H TdR incorporation of airway smooth muscle cells was mediated by such signal transduction pathway as Ca2+, PKC, MAPK and CaN, etc.

Aim:To study whether urotensin ii（Uii）,a potent vasoconstrictive peptide,isinvolved in the development of cardiac hypertrophy and fibrogenesis of ratsinduced by isoproterenol（ISO）.Methods:Thirty male Wistar rats were randomlydivided into 3 *** 1 was the healthy control group,group 2 was the ISOgroup,and group 3 was the ISO+Uii *** groups 2 and 3,ISO(5 mg-kg-1.d-1)wasgiven（sc）once daily for 7 *** 3 was also given Uii in the first day [3 nmol/kg（5μg/kg）,iv],followed by sc（1.5 μg/kg）twice *** 1 received 0.9% *** receptor（UT）mRNA expression was determined by *** contents ofUii and angiotensin ii（Ang ii）were determined by *** vitro,the effects of Uii on DNA/collagen synthesis of cardiac fibroblasts were deter-mined by [3H]thymidine/[3H]proline ***:The ratio of heartweight/body weight,plasma lactate dehydrogenase activity,myocardialmalondialdehyde and hydroxyproline concentration increased significantly in theISO group,as well as UT mRNA expression,plasma and cardiac Uii and ventricu-lar Ang ii,compared with the control group（Pii+ISO co-treatment significantly increasedthe changes of biochemical markers of injury and the degree of cardiac hypertro-phy and *** vitro,5×10～9-5×10-7mol/L Uii stimulated[3H]thymidine/[3H]proline incorporation into cardiac fibroblasts in a dose-dependent manner（Pii was involved in the development ofcardiac fibrosis and hypertrophy by synergistic effects with ISO.

＜正＞Central urotensinⅡ（U-ii） may participate in the regulation of cardiovascular functions by stimulating sympathy ***,their central mechanism remained *** studies have shown that brain reactive oxygen species（ROS） mediate the sympatho-excitaory *** present study,we tested the hypothesis that ROS mediate central cardiocvascular effects of *** were completed in Wistar-Kyoto（WKY） rats and spontaneously hypertensive rats（SHR）.The following observations were made:（1） Expressions of U-ii receptors of rostral ventrl medulla （rVLM ） and nucleus tractus solitarii（NTS） were increased in SHR rats compared with WKY rats.（2） Doubling Immunoflurescence analysis showed that U-ii receptor and NADPH oxidase subunit gp91phox were co-localization in the rVLM and NTS.（3） Intracerbroventricular U-ii significantly increased mean arterial pressure（MAP） and heart rate（HR）. Tempol（a superoxide dismutase mimetic） or Apocinin（NADPH oxidase inhibitor） pretreatment eliminated these effects of U-ii.（4） Brain superoxide levels were enhanced in U-ii-treated rats compared with CSF-treated *** results indicate that superoxide mediates central cardiovascular effects of U-ii and provide evidence for a novel relationship between U-ii and ROS.

Increasing evidence has clarified that Hydrogen sulfide(HS) performs a wide range of physiological and pathological *** is formed from cysteine by cystathionineβ-synthase(CBS) and cystathionineγ-Iyase(CSE), and it is produced mainly by CBS in *** much less is known about its roles than about its two counterparts, NO and CO,it is currently considered to be the third gaseous *** present study,we test the distribution of CBS in the rostral ventrolateral medulla(RVLM) and the effects of HS on regulating central blood *** were completed in Wistar-Kyoto(WKY) rats and spontaneous hypertensive rats(SHR).The following observations were made: (1) Expressions of CBS of RVLM were increased in SHR rats compared with WKY rats.(2) Doubling immunoflurescence analysis showed that U-ii receptor and CBS were co-localized in the RVLM.(3) Introcerbroventricular NaHS by low concentration(200-400 mol,10μl) significantly decreased mean arterial pressure(MAP),while high concentration(1-2 mmol,10μl) could increase mean arterial pressure(MAP).These results indicate that HS produced by CBS in brain may regulate blood pressure and have some relationship with urotensin ii(U-ii),a peptide can regulate cardiovascular functions by stimulating sympathy pathway.
