Stretch-induced human myometrial cytokines enhance immune cell recruitment via endothelial activation

Spontaneous term labour is associated with amplified inflammatory events in the myometrium including cytokine production and leukocyte infiltration; however, potential mechanisms regulating such events are not fully understood. We hypothesized that mechanical Stretch of the uterine wall by the growing fetus facilitates peripheral leukocyte extravasation into the term myometrium through the release of various cytokines by uterine myocytes. Human myometrial cells （hTERT-HM） were subjected to static mechanical Stretch; Stretch-conditioned media was collected and analysed using 48-plex Luminex assay and ELISA. Effect of Stretch-conditioned media on cell adhesion molecule expression of human uterine microvascular endothelial cells （UtMVEC-Myo） was detected by quantitative polymerase chain reaction （qPCR） and flow cytometry; functional assays testing leukocyte-endothelial interactions： adhesion of leukocytes to endothelial cells and transendothelial migration of calcein-labelled primary human neutrophils as well as migration of THP-1 monocytic cells were assessed by fluorometry. The current in vitrostudy demonstrated that mechanical Stretch （i） directly induces secretion of multiple cytokines and chemokines by hTERT-HM cells （IL-6, CXCL8, CXCL1, migration inhibitory factor （MIF）, VEGF, G-CSF, IL-12p70, bFGF and platelet-derived growth factor subunit B （PDGF-bb）, P〈0.05）; Stretch-induced cytokines （ii） enhance leukocyte adhesion to the endothelium of the surrounding uterine microvasculature by （iii） inducing the expression of endothelial cell adhesion molecules and （iv） directing the transendothelial migration of peripheral leukocytes. （vi） Chemokine-neutralizing antibodies and broad-spectrum chemokine inhibitor block leukocyte migration. Our data provide a proof of mechanical regulation for leukocyte recruitment from the uterine blood vessels to the myometrium, suggesting a putative mechanism for the leukocyte infiltrate into the uterus during labour and postpartum involution.

将ICCOS换流原理应用于Stretch meat grinder电路中的电感储能型脉冲电源新型拓扑具有有效提高关断电流和降低系统成本的优势。为构建电气性能和非电性能最优的电感储能型脉冲电源系统,对基于ICCOS的Stretch meat grinder电路的逆流回路的分析和优化设计是必要的。基于ICCOS的Stretch meat grinder电路的理论分析,结合初步实验结果,本文提出了ICCOS逆流回路的基本设计约束,通过逆流电容值及其预充电压的优化取值,使逆流能量最小。仿真分析表明,ICCOS最小逆流能量和所要关断的电流大小直接相关,且受到一次直流电源的影响,而与所关断的电感初始储能大小无关。

Lamin A/C Modulate Apoptosis of Rat Vascular Smooth Muscle Cells During Cyclic Stretch Application

Objective The apoptosis of vascular smooth muscle cells(VSMCs)influenced by abnormal cyclic Stretch is crucial for vascular remodeling during hypertension.We explored that the causes of mechano-responsive lamin A/C changingin aonormai cyclic stretcn and its roles in VSMC apoptosis.Methods and results Our previous vascular proteomics study revealed that LaminA/C is mechano-sensitive molecule.When VSMCs are subjected to cyclic Stretch,the expression of LaminA/C is significantly changed which participates dysfunctions of VSMCs during hypertension.However,the molecular mechanism involved in regulation of LaminA/C expression and the role of LaminA/C in the VSMC apoptosis during cyclic Stretch application are still unclear.In the present study,VSMCs were subjected to different amplitudes of cyclic steetch in vitro:5%cyclic Stretch(physiological strain)or 15%cyclic Stretch(pathological strain).The expression of 2 different selective cleavage isomers of LaminA/C,i.e.LaminA and LaminC,and the apoptosis of VSMCs were detected.The results showed that compared with 5%group,15%cyclic Stretch significantly decreased the expression of LaminA and LaminC,and promoted the apoptosis of VSMCs.Using specific small interfering RNA(siRNA)transfection which targets on LMNA the encoding gene of LaminA/C,the expression of LaminA and LaminC in VSMCs was significantly decreased,and the apoptosis was significantly increased.In order to study the molecular mechanism involved in cyclic Stretch regulating the expression of LaminA/C,we focused on the microRNA(miR).Bioinformatics analysis showed that the 3’untranslated region(3’UTR)of LMNA has two potential binding sites to miR-124-3p.Double luciferase reported system revealed that both sites have binding abilities to miR-124-3p.Under static condition,miR-124-3p inhibitor significantly up-regulated the expression levels of LaminA and LaminC,while the miR-124-3p mimics significantly down-regulated them.RT-PCR results showed that 15%cyclic Stretch significantly up-regulated the expression of miR-12

Effects of Short-Term Mechanical Stretch on the Proliferation and Osteogenic Differentiation of Mesenchymal Stem Cell and TRPC1 Expression

Objective Mechanical Stretch regulates mesenchymal stem cell(MSC)function,which much more emphasis has been placed its prolonged effect on lineage differentiation,especially osteogenic differentiation.In contrast,there are few reports about its short term effect on MSC proliferation.In the present study,effects of short-term mechanical Stretch on the proliferation and osteogenic differentiation of mesenchymal stem cell proliferation were investigated.In addition,the Stretchinfluenced expression of transient receptor potential cation channel,subfamily C,member 1(TRPC1)was also investigated due to its mechanosensitivity and positive correlation with MSC proliferation.Methods MSCs,harvested from rat bone marrow,were seeded on collagen l-coated silicone chamber and exposed to mechanical Stretch with various magnitude(0%,5%,10%and 15%)or various duration(2 h,6 h,12 h and 24 h).Cell proliferation was examined by cell counting kit-8(CCK-8)assay and cell cycle analysis.The gene and protein expression of two makers for osteogenic differentiation,collagen I and Cbfα1,and TRPC1 were determined by RT-PCR and western blotting,respectively.BMSC were harvested,and total RNA was isolated with Trizol reagent.A 2μg portion of total RNA was synthesized to cDNA according to the manufacturer s instructions.cDNA was used as a template for each PCR amplification.BMSC were solubilized in RIPA lysis buffer on ice for 30 min.Phenylmethane sulfonyl fluoride(PMSF)was added to avoid proteolysis.Equal portions of the cell lysates were separated on 10%sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE)and transferred to polyvinylidene fluoride(PVDF)membranes.The membranes were incubated with primary antibodies to TRPC1 and GAPDH at4℃overnight to identify the specific proteins.The PVDF membranes were washed with TBST three times and incubated with a horseradish peroxidase(HRP)-conjugated secondary antibody.Immunoreactive bands were visualized using an enhanced chemiluminescent(ECL)system.Results The OD value for the three stretc

Influence of Stretch and temperature on the energy density of dielectric elastomer generators

Application of dielectric elastomers (DE) has remarkably increased in mechatronics because they are suitable candidates for energy harvesting due to their low cost,light weight,and high energy density.The dielectric elastomer generators (DEGs) exhibit high performance regardless of the applications scale.However,functioning as a generator,a DE may lose its efficiency due to several failure modes including material rupture,loss of tension (LT),electrical breakdown (EB),and electromechanical instability (EMI).The failure modes confine the area of allowable states for generation process.Dielectric constant and dielectric strength of such elastomers depend on the amount of applied deformation and also working temperature,which are often ignored in theoretical simulations.In this paper,variations of the above-mentioned parameters are considered in mechanical and electrical modellings to investigate their effects on energy density and efficiency of generators.Obtained results show that,ignoring the variations of material dielectric constant and dielectric strength leads to overestimation of the specific energy.Furthermore,it is shown that,for an acrylic-based generator,the specific energy sharply decreases with temperature rise.

Cyclic mechanical Stretch regulates MMPs and TIMPs expression in corneal fibroblasts

Laser in situ keratomileusis(LASIK)is a relatively new ophthalmic procedure in refractive surgery.It involves the use of a microkeratome to create a thin corneal flap followed by excimer laser ablation of the corneal stroma and repositioning of the flap.However,complications are bound to occur when a surgical procedure is performed with increasing frequency.In some cases,serious complications such as keratectasia occurred.Risk factors for corneal keratectasia include a thin cornea at baseline,thick corneal flap,excessive ablation,irregular corneal thickness,diverse ablation rates,preexisting keratoconus or form fruste keratoconus,and high intraocular pressure(IOP)~[1].It is evident from incisional refractive surgery that the cornea is not mechanically inert.The mechanical environment of the central corneal tissue is also altered un-

Effects of pulmonary Stretch reflex on lung injury in rabbits with acute respiratory distress syndrome

BACKGROUND： Pulmonary Stretch reflex plays an important role in regulation of respiratorymovement. This study aimed to evaluate the effect of pulmonary Stretch reflex on lung injury inrabbits with acute respiratory distress syndrome （ARDS）.METHODS： ARDS rabbits were given intratracheal infusion of hydrochloric acid and ventilatedwith neurally adjusted ventilatory assistance （NAVA） with a tidal volume （VT） of 6 mL/kg and theelectrical activity of diaphragm （EAdi）-determined positive end expiratory pressure. After isolation ofthe bilateral vagus nerve trunk, the rabbits were randomized into two groups： sham operation （SHAM）group （n=5） and bilateral vagotomy （VAG） group （n=5）. Gas exchange and respiratory mechanicswere detected at baseline, after lung injury and 1, 2, and 3 hours after ventilation respectively.Pulmonary permeability index, pathological changes and infl ammatory response were also measured.RESULTS： Compared with the SHAM group, PaO2/FiO2 in the VAG group decreased signifi cantly2 and 3 hours after ventilation （P〈0.05）. There was no significant difference in PaCO2 betweenthe SHAM and VAG groups （P〉0.05）, and the VAG group had a high VT, peak pressure （Ppeak）,and mean pressure （Pm） compared with the SHAM group 1, 2, 3 hours after ventilation （P〈0.05）.Compared to the SHAM group, dead space fraction （VD/VT） and respiratory system elastance （Ers）in the VAG group increased （P〈0.05） and static pulmonary compliance （Cst） decreased markedly（P〈0.05） after ventilation for 3 hours. Lung wet/dry weight ratio （W/D）（8.4±1.2 vs. 6.6±1.0）, lung injuryscore （6.3±1.8 vs. 3.8±1.3）, tumor necrosis factor-# （TNF-#）（779±372 pg/mL vs. 355±130 pg/mL）and interleukin-8 （IL-8）（169±21 pg/mL vs. 118±17 pg/mL） increased significantly in the VAG groupcompared with the SHAM group （P〈0.05）.CONCLUSION： Lung injury is aggravated after bilateral vagotomy, demonstrating thatpulmonary Stretch refl ex may have protective effect on the lung.

Finite element simulation of Stretch forging using a mesh condensation method

In order to reduce the computation time of finite element simulations of Stretch forging process,a mesh condensation method is presented and applied to a three-dimensional rigid-viscoplastic finite element program.In this method,a conventional mesh for the whole zone of a workpiece is condensed to a computational mesh for the active deformation zone.Two vital problems are solved,which are automatic construction of the computational mesh and treatment of interfaces between the deformation zone and the rigid zone.The mesh condensation method is compared with conventional finite element method by simulations of a six-bite Stretch forging process.Some simulation results including forging load,temperature distribution and effective strain distribution are illustrated.The efficiency and accuracy of this method are verified.
