Background The World Health Organization recommends the use of Schisto point-of-care circulating cathodic anti-gens(Schisto POC-CCA)for screening of schistosoma mansoni as it offers better sensitivity than ***,there are limitation facing the use of this method including timely availability of the test *** aim of this study was to determine the reliability of dried urine spot(DUS)method for collection of urine and detection of *** using Schisto POC-CCA cassettes in a resource-limited *** A cross-sectional study was conducted between October and November 2022 among 250 primary school children in Sengerema District,northwestern *** CCA was detected in filter paper-based DUSs,liquid urine using DUS Schisto POC-CCA(index),and direct urine Schisto POC-CCA(comparator)methods *** eggs in stool were detected using duplicate Kato-Katz(KK)*** measures of accuracy were com-puted and compared between the index and comparator *** strength of agreement between inter-raters precisions was tested using Cohen's kappa(k).Results This study revealed *** prevalence rates of 28.8%,54.0%and 50.8%by duplicate KK,direct urine Schisto POC-CCA and DUS Schisto POC-CCA methods *** mean intensity of infection among infected participants Was 86.3 eggs per gram of stool(EPG)ranging from 12.0 EPG to 824.0 *** sensitivity of DUS Schisto POC-CCA and direct urine Schisto POC-CCA Was 94.44%(95%CI:89.15-99.74%)and 97.22%(95%CI:93.43-100.00%)*** DUS Schisto POC-CCA method had slightly higher specificity(66.85%)than direct urine Schisto POC-CCA method(63.48%).The accuracy of the DUS Schisto POC-CCA Was found to be slightly high(74.80%,95%CI:68.94-79.06%)compared to that of direct urine Schisto POC-CCA(73.20%,95%CI:67.25-78.59%).There was good agreement between two laboratory technologists who performed the DUS Schisto POC-CCA method on similar samples(k=0.80,95%CI:0.59-0.95).Con

Background:Mass drug administration of praziquantel is the World Health Organization’s endorsed control strategy for schistosomiasis.A decade of annual treatments across sub-Saharan Africa has resulted in significant reductions of infection prevalence and intensity levels,although‘hotspots’*** drug treatments place strong selective pressures on parasites,which may affect life-history traits that impact transmission *** drug treatment responses and the evolution of such traits can help inform on how to minimise the risk of drug resistance developing,maximise sustainable control programme success,and improve diagnostic ***:We performed a four-generation schistosoma mansoni praziquantel selection experiment in mice and *** used three *** lines:a praziquantel-resistant isolate(R),a praziquantel-susceptible isolate(S),and a coinfected line(RS),under three treatment regimens:untreated,25 mg/kg praziquantel,or 50 mg/kg *** traits,including parasite adult-worm establishment,survival,reproduction(fecundity),and associated morbidity,were recorded in mice across all four *** variables were tested in a series of generalized linear mixed effects models to determine which factors had a significant influence on parasite life-history traits in definitive hosts under different selection ***:Praziquantel pressure significantly reduced adult-worm burdens across all generations and isolates,including within ***,previous drug treatment resulted in an increase in adult-worm establishment with increasing generation from P1 to *** highest worm numbers were in the co-infected RS *** treatment decreased adult-worm burden,but had a larger negative impact on the mean daily number of miracidia,a proxy for fecundity,across all three parasite ***:Our predicted cost of resistance was not supported by the traits we measured within the murine host.

Melatonin and schistosomal antigens ameliorate the anti-oxidative and biochemical response to schistosoma mansoni infection in hamster

The present study was designed to investigate the potential protective effect of melatonin as an antioxidant separately or in combination with antigens （cerearial; CAP or soluble worm; SWAP） against schistosoma mansoni infection in hamsters. Each hamster was sensitized with an initial immunization of 0.6 ml of the extracted antigen （30μg protein/mL）. After four days, a second injection of 0.4 mL was given （20μg protein/mL）. Then, each hamster was exposed to 260 ± 20 S. mansoni cercariae followed with melatonin treatment （3.5 mg/kg） for thirty days from the 1st day of post infection. Levels of lipid peroxidation （LPO） products, eatalase （CAT） activity, hepatic glutathione （GSH） and biochemical changes in the liver and kidneys functions were investigated. The results revealed a high significant increasing of LPO and decreasing of CAT and GSH in liver of infected hamsters. Biochemical observations showed severe damage in the liver enzyme activities and increasing cholesterol level in infected animals. Melatonin co-treatment with antigen to the infeeted-hamster attenuated the increase of LPO and restored the activity of CAT and levels of hepatic GSH. Also, the biochemical damages in the liver and kidneys funetions were reduced. The present study suggests that melatonin may be useful in combating free radical-induced damage due to infection toxicity. The immunization with previous antigens resulted in a remarkable improvement on the liver enzyme activities, which were increased after infection. Thus, vaccination of hamsters with antigens （both CAP and SWAP） and melatonin treatment has more potent effect on the enhancement of antioxidant and biochemical of S. mansoni infected-hamster than each treatment separately. Immunization of the hamster with SWAP followed by melatonin was the best way among the other regime treatments to improve the biochemical and antioxidant parameters of the infected-hamsters.

AIM:To investigate the therapeutic effect of schistosoma mansoni(***) soluble worm proteins on gastrointestinal motility disturbances during experimental colitis in mice. METHODS:Colitis was induced by intrarectal injection of trinitrobenzene sulphate(TNBS) and 6 h later,mice were treated ip with *** *** were performed 5 d after TNBS *** quantified using validated inflammation parameters. AIM： To investigate the potential of bone marrow mononuclear cells （BM-MCs） in the regeneration of hepatic lesions induced by schistosoma mansoni （***） chronic infection. METHODS： Female mice chronically infected with *** were treated with BM-MCs obtained from male green fluorescent protein （GFP） transgenic mice by intravenous or intralobular injections. Control mice received injections of saline in similar conditions, Enzyme-linked immunosorbent assay （ELISA） assay for transforming growth factor-beta （TGF-β）, polymerase chain reaction （PCR） for GFP DNA, immunofluorescence and morphometric studies were performed. RESULTS： Transplanted GFP^＋ cells migrated to granuloma areas and reduced the percentage of liver fibrosis. The presence of donor-derived cells was confirmed by Fluorescence in situ hybridization （FISH） analysis for detection of cells bearing Y chromosome and by PCR analysis for detection of GFP DNA. The levels of TGF-β, a cytokine associated with fibrosis deposition, in liver fragments of mice submitted to therapy were reduced. The number of oval cells in liver sections of ***-infected mice increased 3-4 fold after transplantation. A partial recovery in albumin expression, which is decreased upon infection with ***, was found in livers of infected mice after cellular therapy. CONCLUSION： In conclusion, transplanted BMCs migrate to and reduce the damage of chronic fibrotic liver lesions caused by ***.
