Pomegranate fruit peel is rich in bioactive plant natural products, such as hydrolyzable tannins and anthocyanins. Despite their documented roles in human nutrition and fruit quality, genes involved in natural product biosynthesis have not been cloned from pomegranate and very little sequence information is available on pomegranate in the public domain. Shotgun transcriptome sequencing of pomegranate fruit peel cDNA was performed using RNA-Seq on the Illumina Genome Analyzer platform. Over 100 million raw sequence reads were obtained and assembled into 9,839 transcriptome assemblies （TAs）（〉200 bp）. Candidate genes for hydrolyzable tannin, anthocyanin, flavonoid, terpenoid and fatty acid biosynthesis and/or regulation were identified. Three lipid transfer proteins were obtained that may contribute to the previously reported IgE reactivity of pomegranate fruit extracts. In addition, 115 ssr markers were identified from the pomegranate fruit peel transcriptome and primers were designed for 77 ssr markers. The pomegranate fruit peel transcriptome set provides a valuable platform for natural product biosynthetic gene and ssr marker discovery in pomegranate. This work also demonstrates that next-generation transcriptome sequencing is an economical and effective approach for investigating natural product biosynthesis, identifying genes controlling important agronomic traits, and discovering molecular markers in non-model specialty crop species.

This study provides new insights for the hatchery released Chinese shrimp(Fenneropenaeus chinensis),including proportion,dynamic migration route,after they were released into nature for stock enhancement using a new strategy quite different than *** shrimp were sampled at 22 survey stations during two investigation voyages acrossing 74 survey stations in the Bohai Sea from July 16 to August 9 in *** 289 sampled individuals during the second voyage,totally 155 shrimps were identified as hatchery shrimp released into the Laizhou Bay at mid-May in 2015 based on finger-print of eight ssr(simple sequence repeats)markers,and the proportion of hatchery released shrimp in recapture samples were from 41.30%–85.71%in each station with an average value 53.63%,which verified a previous view point that up to 90%of autumn season Chinese shrimp landing in the Bohai Sea were composed of hatchery ***,the dynamic migration route of hatchery released shrimp revealed that part of released shrimp migrated heading northwest along the west coast of the Bohai Sea up to the Bohai Bay but just remained at the Laizhou Bay until over-wintering migration at midOctober when they initiate over-wintering *** unnatural spring season shrimp fishing model cut the throat of spawner shrimp chance to swim back to their respective spawning plants at each spring,it still no chance to clarify whether the hatchery released shrimp could replenish to the reproduce population and complete a whole life cycle as same as their natural relatives.

Purity Identification of Xinkui 19 by ssr marker Technique

In this study, by using Xinkui 19 and its parents as experimental materials, 100 pairs of ssr molecular markers were screened to obtain specific poly- morphic primers, aiming at providing an accurate and efficient method for identifying the purity of Helianthus annuus L. hybrid seeds. According to the experimen- tal results, by using polymorphic ssr primer 455, the amplified bands of female and male parents were 460 and 430 bp, respectively; by using polymorphic ssr primer 478, the amplified bands of female and male parents were 330 and 350 bp, respectively. The identification results of Xinkui 19 hybrids and its parents with ssr marker technique and field cultivation were basically consistent. ssr primers 455 and 478 could be used for rapid and effective identification of the purity of Xinkui 19 hybrid seeds.

[Objective] This study aimed to construct DNA fingerprint for hybrid rice cultivars those have been approved by Hunan Province. [Method] The primers which produced polymorphic and bright DNA bands were selected to construct the DNA molecular fingerprint map for 77 major hybrid rice cultivars approved by Hunan Province. [ Result] A total of 48 ssr primers were selected. Every cultivar had its unique fingerprint map so that the obtained data could identify differ- ent hybrid rice cultivars. [ Conclusion] This study made great contributions to the perfection of hybrid rice germplasm identification.

To select highly informative microsatellite markers （ssrs） and establish a useful genetic ssr framework for rice genotyping, 15 rice （Oryza sativa L.） cultivars including six indica varieties and nine japonica varieties were used to analyze the polymorphism information content （PIC） value of 489 ssr markers. A total of 1 296 alleles were detected by 405 polymorphic markers with an average of 3.2 per locus. The PIC value of each chromosome was ranged from 0.4039 （chromosome 2） to 0.5840 （chromosome 11）. Among the two rice subspecies, indica （0.3685-0.4952） gave a higher PIC value than japonica （0.1326-0.3164） and displayed a higher genetic diversity. Genetic diversity of indica was high on chromosome 12 （0.4952） and low on chromosome 8 （0.3685）, while that for japonica was high on chromosome 11 （0.3164） and low on chromosome 2 （0.1326）. A ssr framework including 141 highly informative markers for genotyping was selected from 199 ssr markers （PIC〉0.50）. Ninety-three ssr markers distributed on 12 chromosomes were found to be related to indica-japonica differentiation. Of these 93 pairs of ssr primers, 17 pairs were considered as core primers （all the japonica varieties have the same specific alleles, while the indica varieties have another specific alleles）, 48 pairs as the second classic primers （all the japonica or indica varieties have the same specific alleles, while the indica or japanica varieties have two or more other specific alleles ） and 28 pairs as the third classic primers （all the japonica and indica varieties have two or more alleles, but the specific alleles are different between japonica and indica）. Thirty-two ssr markers were selected to be highly informative and useful for genetic diversity analysis of japonica varieties. This work provides a lot of useful information of ssr markers for rice breeding programs, especially for genotyping, diversity analysis and genetic mapping.

Purity Identification of Xinshikui 6 Using ssr marker Technique

[Objective] ssr molecular marker technique was used to determine the purity of sunflower seed with the aim to provide accurate, convenient method for the identification of the purity of hybrid seeds in production and processing. [Method] With the DNA of Xinshikui 6 and its parents as template, about 100 pairs of ssr molecular markers were screened after DNA extraction, PCR amplification and electrophoresis production. [Results] ssr polymorphic primer marker 532 produced a specific band of 469 bp in the female parent, and a specific band of 451 bp in the male parent; primer marker 574 produced a specific band of 364 bp in the female parent, and a specific band of 384 bp in the male parent. The indoor molecular purity identification and field purity identification were consistent with each other. The primer marker 532 and 574 could be obtained from the ssr molecular marker method to distinguish the male parent, female parent and hybrid of Xinshikui 6, and both of the 2 primer markers can effectively identify the purities of the hybrid seeds of Xinshikui 6, as well as the authenticity of the seeds. [Conclusion] The proposed method was simple, fast, accurate to operate with the advantages of high reproducibility, and it had become the major method in the identification of sunflower varieties.

Genetic diversity in relation to sharp eyespot resistance was investigated among 76 wheat cultivars from home and abroad using 61 wheat ssr markers.A total of 321 allelic variants were detected,the polymorphism information content of ssr markers ranged from 0 to 0.86,with an average of 0.59,meaning that 76 resources had high *** clusters were identified based on 0.76 of genetic *** three genomes were ranked as B＞D＞A based on ssr alleles per locus and PIC *** varieties were more polymorphic than large-scale planted varieties and local *** information generated in this study will assist in the selection of parental lines in order to increase the efficiency of breeding efforts for sharp eyespot resistance.

The genetic diversity of 118 accessions of broomcom millet （Panicum miliaceum L.）, collected from various ecological areas, was analyzed. Using 46 ssr （Simple Sequence Repeat） polymorphic markers from rice, wheat, oat and barley, a total of 226 alleles were found, which exhibited moderate level of diversity. The number of alleles per primer ranged from two to nine, with an average of 4.91. The range of polymorphism information content （PIC） was 0.2844）.980 （average, 0.793）. The expected heterozygosity （He） varied from 0.346 to 0.989, with an average of 0.834. The average coefficient of the genetic similarity of ssr markers among the 118 accessions was 0.609, and it ranged from 0.461 to 0.851. The UPGMA （Unweight Pair Group Method with Arithmetic Mean） clustering analysis at the genetic similarity value of 0.609 grouped the 118 accessions into five groups. Mantel test meant that geographical origin and genetic distance presented positive correlation. The clustering results were consistent with known information on ecological growing areas. The genetic similarity coefficient of the accessions in the Loess Plateau ecotype was significantly lower than those in the other ecotypes. It indicates that the highest level of genetic diversity occurred in the Loess Plateau, which is probably the original site of Panicum miliaceum.

Genetic diversity of 62 Sichuan wheat landraces accessions of China was investigated by agronomic traits and ssr markers. The landrace population showed the characters of higher tiller capability and more kernels/spike, especially tiller no./plant of six accessions was over 40 and kernels/spike of three accessions was more than 70. A total of 547 alleles in 124 polymorphic loci were detected with an average of 4.76 alleles per locus by 114 ssr markers. Parameters analysis indicated that the genetic diversity ranked as genome A 〉 genome B 〉 genome D, and the homoeologous groups ranked as 5〉4〉3〉1〉2〉7〉6 based on genetic richness （Ri）. Furthermore, chromosomes 2A, 1B and 3D had more diversity than that of chromosomes 4A, 7A and 6B. The variation of ssr loci on chromosomes 1B, 2A, 2D, 3B, and 4B implied that, in the past, different selective pressures might have acted on different chromosome regions of these landraces. Our results suggested that Sichuan common wheat landraces is a useful genetic resource for genetic research and wheat improvement.

Analysis of an Applied Core Collection of Adzuki Bean Germplasm by Using ssr markers

Genetic diversity of 158 accessions of an applied core collection of adzuki bean （Vigna angularis） and 18 wild genotypes were assessed by using 85 microsatellite markers. With an average of 5.81 alleles per locus, 493 alleles were detected, and their distribution frequencies lower than 5% accounted for 73.02% of the total number. The distributions of alleles between the cultivated and the wild adzuki bean germplasm are different, with a higher allelic diversity in the wild germplasm than that of the cultivated ones. An obvious genetic differentiation was also observed between the wild and the cultivated adzuki beans, and ssr markers may be useful in study identification and classification of them. Among cultivated adzuki bean, the genetic similarity coefficient varied from 0.366 to 0.939. Genetic structure analysis can clearly separate the wild genotypes from the cultivated adzuki bean, and also can divide the cultivated ones into different populations, as these populations are closely agreeable with the ecological regions where they originally grow. The results of this study will be useful in arranging local breeding programs, especially in the aspect of parental combinations or identification of progenies. These ssr markers can also provide important information to explain the genetic relationship between the cultivated and wild adzuki beans, and to accelerate the wild gene resources in broadening the gene pool in breeding program.
