Background prostate specific membrane antigen(PSMA)can facilitate the growth,migration,and invasion of the LNCaP prostate cancer cell lines,but the underlying molecular mechanisms have not yet been clearly ***,we investigated whether PSMA serves as a novel regulator of the phosphatidylinositol 3-kinase(PI3K)/Akt signaling by employing PSMA knockdown model and PI3K pharmacological inhibitor(LY294002)in LNCaP prostate cancer *** PSMA knockdown had been stably established by transfecting with lentivirus-mediated siRNA in our previous ***,LNCaP cells were divided into interference,non-interference,and blank *** first testified the efficacy of PSMA knockdown in our LNCaP cell ***,we compared the expression of PSMA and total/activated Akt by Western blotting in the above three groups with or without LY294002 ***,immunocytochemistry was performed to confirm the changes of activated Akt(p-Akt,Ser473)in ***,cell proliferation,migration,and cell cycle were measured by CCK-8 assay,Transwell analysis,and Flow cytometry *** After PSMA knockdown,the level of p-Akt(Ser473)but not of total-Akt(Akt1/2)was significantly decreased when compared with the non-interference and blank ***,LY294002 administration significantly reduced the expression of p-Akt(Ser473)in all the three *** results of immunocytochemistry further confirmed that PSMA knockdown or LY294002 treatment was associated with p-Akt(Ser473)*** of cell proliferation,migration,and survival were also observed upon PSMA knockdown and LY294002 *** Taken together,our results reveal that PI3K/Akt signaling pathway inhibition may serve as a novel molecular mechanism in LNCaP prostate cancer cells of PSMA knockdown and suggest that Akt(Ser473)may play a critical role as a downstream signaling target effector of PSMA in this cellular model.

Head-to-head comparison of PSMA PET/CT and mpMRI for detecting biochemical recurrence of prostate cancer: A systematic review and meta-analysis

Objectives This study aimed to evaluate the performance of prostate-specific membrane antigen positron emission tomography/computed tomography(PSMA PET/CT)in comparison to multiparametric magnetic resonance imaging(mpMRI)for detecting biochemical recurrence of prostate cancer(PCa).Materials and methods We conducted a comprehensive search for articles published in PubMed,Web of Science,Embase,and the Cochrane Library,spanning the inception of the database until October 26,2022,which included head-to-head comparisons of PSMA PET/CT and mpMRI for assessing the biochemical recurrence of *** A total of 5 studies including 228 patients were *** overall positivity rates of PSMA PET/CT and mpMRI for detecting biochemical recurrence of PCa after final treatment were 0.68(95%confidence interval[CI],0.52–0.89)and 0.56(95%CI,0.36–0.88),*** positivity rates of PSMA PET/CT and mpMRI for detecting local recurrence,lymph node metastasis,and bone metastases were 0.37(95%CI,0.30–0.47)and 0.38(95%CI,0.22–0.67),0.44(95%CI,0.35–0.56)and 0.25(95%CI,0.17–0.35),and 0.19(95%CI,0.11–0.31)and 0.12(95%CI,0.05–0.25),*** with mpMRI,PSMA PET/CT exhibited a higher positivity rate for detecting biochemical recurrence and lymph node metastases,and no significant difference in the positivity rate of local recurrence was observed between these 2 imaging *** Compared with mpMRI,PSMA PET/CT appears to have a higher positivity rate for detecting biochemical recurrence of *** both imaging methods showed similar positivity rates of detecting local recurrence,PSMA PET/CT outperformed PSMA PET/CT in detecting lymph node involvement and overall recurrence.

Anti-Tumor Efficacy of Gene Vaccine Expressing PSMA

OBJECTIVE To observe anti-tumor effects of PVAX-PSMA gene vaccine. METHODS The PSMA gene was inserted into a mammalian expression vector, PVAX-1, to construct the DNA vaccine candidate, and was then used to vaccinate C57BL/6 mice. Animals vaccinated with PVAX-1 and NaC1 were used as controls. Anti-PSMA antibody was detected in sera of the animals. The proliferation and cytotoxicity of the spleen cells were observed. The immunized mice were inoculated with RM-1 cells. The mice were inoculated with RM-1 cells, and then the mice were immunized. The anti-tumor efficacy of the gene vaccine was evaluated by the ratio of tumor formation, tumor volume, tumor mass before and after gene vaccination and evaluated by survival rate of the immunized mice. RESULTS High level of anti-PSMA antibody was induced in the PVAX-PSMA group. The splenocytes from PVAX-PSMA group were stimulated to produce strong proliferation responses and significant cytotoxic T-cells （CTL） activity. After the mice were immunized with PVAX-PSMA gene, tumor occurrence was decreased, and the growth velocity of tumor was markedly reduced, resulting in prolonged tumor-free time （P 〈 0.05）. CONCLUSION PVAX-PSMA gene vaccine has significant anti- tumor effects and provides an experimental basis for primary prevention and immunotherapy of prostate cancer.

In this study, prostate-specific membrane antigen（PSMA）-targeted and core-crosslinked micelles were developed based on prostate cancer-binding peptide（PCP） modified glycol chitosan-lipoic acid（PGCLA） conjugate. The degree of substitution was 5.2 PCP groups and 10.7 lipoic acid groups per 100 sugar residues of glycol chitosan in PGC-LA copolymer. Docetaxel（DTX） was chosen as a model anti-tumor drug. The DTX-loaded micelles were prepared by an o/w method, and core-crosslinked micelles were further constructed by using a catalytic amount of dithiothreitol. The mean diameter of DTX-loaded core-crosslinked PGC-LA（DTX-PGC-LA/cc） micelles was 397 nm determined by dynamic light scattering（DLS）. In vitro DTX released from core-crosslinked micelles was slower than that from non-crosslinked counterpart. Blank micelles exhibited good biocompatibility. Additionally, cellular uptake and cytotoxcity of PCP-modified micelles were higher than those of micelles without PCP in PSMA-positive LNCaP cells. Importantly, DTXPGC-LA/cc demonstrated the stronger anti-tumor efficacy against LNCaP tumor xenograft models than DTX injection and other DTX-loaded micelles. Taken together, this study provides a potential way in developing actively targeted and core-crosslinked micelles for hydrophobic drug delivery in cancer therapy.
