The rapid development of biophotonics and biomedical sciences makes a high demand on photonic structures to be interfaced with biological systems that are capable of manipulating light at small scales for sensitive detection of biological signals and precise imaging of cellular ***,conventional photonic structures based on artificial materials(either inorganic or toxic organic)inevitably show incompatibility and invasiveness when interfacing with biological *** design of biophotonic probes from the abundant natural materials,particularly biological entities such as virus,cells and tissues,with the capability of multifunctional light manipulation at target sites greatly increases the biocompatibility and minimizes the invasiveness to biological *** this review,advances in biophotonic probes for bio-detection and imaging are *** emphatically and systematically describe biological entities-based photonic probes that offer appropriate optical properties,biocompatibility,and biodegradability with different optical functions from light generation,to light transportation and light *** representative biophotonic probes,i.e.,biological lasers,cell-based biophotonic waveguides and biomicrolenses,are reviewed with applications for bio-detection and ***,perspectives on future opportunities and potential improvements of biophotonic probes are also provided.

Ubiquitination, a key and extensive posttranslational modification of proteins, has profound effects on a variety of physiological and pathological processes. The inherent complexity of ubiquitin conjugates makes it highly challenging to study the functional and structural mechanisms of ubiquitination. To address these challenges, accesses to sufficient poly-ubiquitin chains or ubiquitinated proteins are urgently needed. Over the last decade, synthetic protein chemists have developed several novel peptide ligation methods for the preparation of ubiquitin conjugates with precise control over the atomic structure. In this review, we summarize the recent breakthroughs and potential challenges in the chemical synthesis and semi-synthesis of ubiquitin conjugates with respect to the preparation of poly-ubiquitin-based proteins and ubiquitin-based probes.

Polarity is an integral microenvironment parameter in biological systems closely associated with a multitude of cellular *** polarity variations accompany the initiation and development of pathophysiological ***,monitoring the abnormal polarity is of scientific and practical *** state-of-the-art monitoring techniques are primarily based on fluorescence imaging which relies on a single emission intensity and may cause inaccurate detection due to heterogeneous accumulation of the ***,we report carbon dots(CDs)with ultra-sensitive responses to *** CDs exhibit two linear relationships:one between fluorescence intensity and polarity and the other between polarity and the maximum emission *** emission spectrum is an intrinsic property of the probes,independent of the excitation intensity or probe *** features enable two-color imaging/quantitation of polarity changes in lipid droplets(LDs)and in the cytoplasm via in situ emission *** probes reveal the polarity heterogeneity in LDs which can be applied to make a distinction between cancer and normal cells,and reveal the polarity homogeneity in cytoplasm.

Enzymes are macromolecular biological catalysts which can accelerate chemical reactions in living organisms. Almost all the physiological metabolism activities in the cell need enzymes to sustain life via rapid catalysis. Currently, medical research has proved that abnormal enzyme activity is associated with numerous diseases, such as Parkinson’s disease(PD), Alzheimer’s disease(AD) and cancers. On the other hand, early diagnosis of those diseases is of great significance to improve the survival rate and cure *** the current diagnostic tools, two-photon fluorescent probes(TPFPs) are developing rapidly due to their unique advantages, such as higher spatial resolution, deeper imaging depth, and lower ***, the design and synthesis of two-photon(TP) small molecule enzymatic probes have broad prospects for early diagnosis and treatment of diseases. As of now, scientists have developed many TP small molecule enzymatic probes. This review aims to summarize the TP small molecule enzymatic probes and expound the reaction mechanism.

We report on the fabrication of fluorescent and multicolor probes for Zn^2＋ ions and temperature from a mixture of three types of fluorophore-labeled responsive block copolymers in aqueous media. Quinoline-based Zn^2＋-recognizing fluorescent mono- mer ZQMA, red-emitting rhodamine B-based monomer RhBEA, and blue-emitting coumarin derivative Coum-OH, were syn- thesized first. A ZQMA-labeled well-defined double hydrophilic block copolymer （DHBC）, PEG-b-P（MEO2MA-co-ZQMA）, was synthesized via reversible addition-fragmentation chain-transfer （RAFT） polymerization of 2-（2-methoxyethoxy）ethyl methacrylate （MEOzMA） and ZQMA by utilizing a PEG-based macroRAFT agent. Following similar procedures, PEG-b-P（St-co-RhBEA） amphiphilic diblock copolymer and PEG-b-P（MEOzMA-co-Coum） DHBC were also synthesized, where P（St-co-RhBEA） was a RhBEA-labeled polystyrene （PS） block. At room temperature in aqueous solution, almost non- fluorescent PEG-b-P（MEOzMA-co-ZQMA） can effectively bind Zn2＋ ions, leading to prominent green fluorescence enhance- ment due to the coordination of ZQMA with Zn^2＋ ions. However, by mixing red-emitting PEG-b-P（St-co-RhBEA） and blue-emitting PEG-b-P（MEO2MA-co-Coum） with PEG-b-P（MEO2MA-co-ZQMA） at an appropriate ratio, three color transitions could be observed. In the absence of Zn^2＋ ions, a mixed pink fluorescent originating from Coum and RhBEA was ob- served; upon the addition of a certain amount of Zn^2＋ ions, the green fluorescence enhanced dramatically, leading to a white fluorescence readout. By further increasing the amount of Zn2＋ ions, the green fluorescence further enhanced and overwhelmed the blue and red emissions, leading to a green-dominant mixed-fluorescence emission. In addition, upon increasing the temperature, the fluorescence of Coum decreased considerably due to the fluorescence-resonance energy transfer （FRET） between Coum and ZQMA moieties. In this way, a ratiometric fluorescent thermometer can be constructed.

Organic phosphate biomolecules(OPBs) are indispensable components of eukaryotes and prokaryotes,such as acting as the fundamental components of cell membranes and important substrates for nucleic acids. They play pivotal roles in various biological processes, such as energy conservation, metabolism,and signal modulation. Due to the difficulty of detection caused by variety OPBs, investigation of their respective physiological effects in organisms has been restrained by the lack of efficient tools. Many small fluorescent probes have been employed for selective detection and monitoring of OPBs in vitro or in vivo due to the advantages of tailored properties, biodegradability and in situ high temporal and spatial resolution imaging. In this review, we summarize the recent advances in fluorescent probes for OPBs,such as nucleotides, NAD(P)H, FAD/FMN and PS. Importantly, we describe their identification mechanisms in detail and discuss the general strategies for these OPBs probe designs, which provide new insights and ideas for the future probe designs.

Hydrogen peroxide(H2 O2), as important products of oxygen metabolism, plays an important role in many biological processes, such as immune responses and cellular signal transduction. However, abnormal production of H2 O2 can damage cellular biomolecules, which was closely associated with many ***, it is urgent to monitor the level change of H2 O2 in living cells, particularly at subcellular *** this end, a wide variety of H2 O2 fluorescent probes have been designed, developed and applied for imaging of H2 O2 in subcellular levels. In this review, we highlight the representative cases of H2 O2 fluorescent probes with mitochondria, nuclei and lysosomes-targetable ability. The review contains organelle target strategies, structures, fluorescence behavior and biological applications of these probes.
