Diet and monensin influence the temporal dynamics of the rumen microbiome in stocker and finishing cattle

The effect of diet supplemented with vegetable oils and/or monensin on the vaccenic acid production in continuous culture fermenters

Studies have shown that supplementing ruminant diets with vegetable oils modulated the rumen biohydrogenation and increased polyunsaturated fatty acid in their products. These positive values are often accompanied by a marginal loss of supplemented unsaturated fatty acids and rise in the concentrations of saturated fatty acids. This study were carried out mainly to investigate the effect of supplementing diets with sunflower oil, olive oil with or without monensin on the production and accumulation of vaccenic acid(VA) in continuous culture fermenters as a long term in vitro rumen simulation *** dual-flow continuous culture fermenters were used in an 8 replication experiment lasted 10 days each(first 7 days for adaptation and last 3 days for samples collection). Supplementing diets with plant oils and monensin in the present experiment increased VA and conjugated linoleic acids(P > 0.05) in ruminal cultures. The results suggest that supplementing diets with both olive oil and sunflower oil and monensin increased VA accumulation compared to plant oils supplemented alone without affecting the rumen dry matter and organic matter digestibility.

一、概述 1951年Berger,J.等人在美国第一次分离到了三个多醚类抗生素,但由于毒性高无临床使用价值并未引起重视,其后经十几年时间无人问津。1967年美国Eli Lilly公司***等人报导在肉桂地链霉菌(*** ATCC15413)的培养物中分离到一个新的多醚化合物莫能菌素,发现它在鸡体内有显著的抗球虫(Ei-meria tenella)的活性,因而能降低死亡率并能促进饲料利用率。同年***等建立了它具有五环的单羧酸多醚化合物的结构,此结构的新颖及具有抗球虫的特效引起各国对多醚化合物筛选以极大的兴趣,从1968年起新多醚抗生素的发现接踊而至,从而开始归列成一大类(polyether antibi-otics)。莫能菌素在农业上具有杀虫杀螨效果,并对豆类白粉病在低浓度下即显示很好的活性。除抗球虫外还可防治猪的赤痢病,能大大提高牛对饲料的利用率,促进生长。近年来又用于增进心血管功能及治疗休克、充血

本文通过建立一种静态接口装置,采用柱后衍生技术联用超临界色谱在线测定monensin化合物在超临界CO中的溶解度及其变化规律。使用香兰素作为衍生剂,与乙醇、硫酸配置衍生化溶液,在线与monensin化合物进行柱后衍生反应,DAD检测波长为520 nm。在温度40～55℃和压力10～27.5 MPa的范围内获得monensin化合物溶解度。其中,40℃时monensin在超临界CO中的最大溶解度为1.55×10(A为88.4%,B为11.6%);55℃时的最大溶解度为1.82×10(A为89.6%,B为10.4%)。通过超临界色谱和超临界质谱对monensin化合物进行在线对比分析发现,超临界色谱根据出峰保留时间能够确定monensin A和B两个组分;而超临界质谱根据TIC和XIC确定monensin A、B、C三种组分。TIC与XIC分析结果确定monensin A、B、C的分子量(monensin A 692.4 m/z,monensin B 678.4m/z,monensin C 706.4 m/z)及停留时间4.99 min、5.41 min和5.64 min。

Pre-column Derivatization-High Performance Liquid Chromatography for the Detection of monensin in Livestock and Poultry Meat

[Objectives]A method for the detection of monensin in poultry and livestock meat by pre-column derivatization-high performance liquid chromatography was established.[Methods]The sample was extracted with chloroform,derivatized with trichloroacetic acid and 2,4-dinitrophenylhydrazine,and centrifuged to obtain a purified solution.A C18 chromatographic column(4.6 mm×150 mm,5μm)was used for separation with(1.5%)acetic acid water∶methanol(volume ratio)=1∶9 as the mobile phase using a DAD detector for detection,and the external standard method was adopted for peak area quantification.[Results]monensin had good linearity in the concentration range of 5.00-200 mg/L,with the linear correlation coefficient r 2>0.999;the detection limit was 5.00 mg/kg;the relative standard deviation was smaller than 10%;and the recoveries of standard addition experiment were in the range of 75%-110%.[Conclusions]The method has the advantages of simple pretreatment operation,good derivatization effect and fast detection speed,and is suitable for detecting monensin in poultry and livestock meat.

Live-Cell Imaging of Dual-Labeled Golgi Stacks in Tobacco BY-2 Cells Reveals Similar Behaviors for Different Cisternae during Movement and Brefeldin A Treatment

In plant cells, the Golgi apparatus consists of numerous stacks that, in turn, are composed of several flattened cisternae with a clear cis-to-trans polarity. During normal functioning within living cells, this unusual organelle displays a wide range of dynamic behaviors such as whole stack motility, constant membrane flux through the cisternae, and Golgi enzyme recycling through the ER. In order to further investigate various aspects of Golgi stack dynamics and integrity, we co-expressed pairs of established Golgi markers in tobacco BY-2 cells to distinguish sub-compartments of the Golgi during monensin treatments, movement, and brefeldin A （BFA）-induced disassembly. A combination of cis and trans markers revealed that Golgi stacks remain intact as they move through the cytoplasm. The Golgi stack orientation during these movements showed a slight preference for the cis side moving ahead, but trans cisternae were also found at the leading edge. During BFA treatments, the different sub-compartments of about half of the observed stacks fused with the ER sequentially; however, no consistent order could be detected. In contrast, the ionophore monensin resulted in swelling of trans cisternae while medial and particularly cis cisternae were mostly unaffected. Our results thus demonstrate a re- markable equivalence of the different cisternae with respect to movement and BFA-induced fusion with the ER. In addi- tion, we propose that a combination of dual-label fluorescence microscopy and drug treatments can provide a simple alternative approach to the determination of protein localization to specific Golgi sub-compartments.
