Metabolomics analysis of poly(L-lactic acid)nanofibers’performance on PC12 cell differentiation

The aim of this article is to reveal the influence of aligned/random poly(L-lactic acid)(PLLA)nanofibers on PC12 cell differentiation from the perspective of metabolic ***,three materials-PLLA aligned nanofibers(PLLA AF),PLLA random nanofibers(PLLA RF)and PLLA films(control)-were prepared by electrospinning and spin *** surface morphologies were ***,the cell viability,cell morphology and neurite length of PC12 cells on the surface of the three materials were evaluated,indicating more neurites in the PLLA RF groups but the longer average neurite length in the PLLA AF ***,the metabolite profiles of PC12 cells cultured on the surface of the three nanofibers after 12 h,24 h and 36 h showed that,compared with the control,51,48 and 31 types of differential metabolites were detected at the three time points among the AF groups,respectively;and 56,45 and 41 types among the RF groups,***,the bioinformatics analysis of differential metabolites identified two pathways and three metabolites critical to PC12 cell differentiation influenced by the *** addition,the verification experiment on critical metabolites and metabolic pathways were *** integrative analysis combining cytology,metabolomics and bioinformatics approaches revealed that though both PLLA AF and RF were capable of stimulating the synthesis of neurotransmitters,the PLLA AF were more beneficial for PC12 cell differentiation,whereas the PLLA RF were less effective.

OBJECTIVE:To explore the four Qi of Pfaffia glomerata(PG)and endow this foreign folk herb with the properties of Chinese medicine,make it Chinese medicinal and localized,and could be used as a Chinese ***:The normal group,six cold herb groups,six hot herb groups,six cool herb groups,PG prescription group(PGPG),and Dangshen(Radix Codonopsis)prescription group(CPPG)were prepared with corresponding concentrations of water extracts,these herb extracts were administered by gavage to SpragueDawley rats,and the 12 h urine at night on the 29th day of the SD rats in each group were collected,Liquid Chromatograph Mass Spectrometer system was used to analyze them,the best discriminant models for the medicinal properties of cold-hot and cold-cool were set up,so as to the medicinal properties of PGPG,CPPG and PG were *** on the Progenesis QI,Human Metabolome Database,Kyoto Encyclopedia of Genes and Genomes,MetaboAnalyst 5.0 database,we enriched metabolic pathway and classification mechanism of medicinal properties of cold-cool Chinese herbs and the molecular mechanism of PG ***:We established a best model of cold-hot herbal discrimination in the positive ion mode,then the probability that PGPG was predicted cold property was 88.9%.Furthermore,a model of cold-cool herbal discrimination was established,then the probability of PGPG containing the cool property was 77.8%.In addition,typical cold and cool herbs mainly affected nine biomarkers such as tyrosine-proline,(R)-3',7-Dihydroxy-2',4'-dimethoxyisoflavane in *** regulation trend of PGPG on markers was basically as same as the cool herbs and mainly involved in regulating the two pathways of cytochrome P450 and purine ***:The results showed PGPG had a cool medicinal property as same as CPPG,and the regulation trend of PGPG on markers was consistent with cool ***,the medicine properties PG and CP should be consistent,and the Traditional Chinese Medicine p

The p38/MAPK pathway as a therapeutic target to prevent therapeutic escape of breast cancer stem cells

Cancer stem cells(CSCs)play an important role in metastasis development,tumor recurrence,and treatment resistance,and are essential for the eradication of ***,therapies fail to eradicate CSCs due to their therapeutic stress-induced cellular escape,which leads to enhanced aggressive behaviors compared with CSCs that have never been ***,the underlying mechanisms regulating the therapeutic escape remain *** this end,we established a model to isolate the therapeutic escaped CSCs(TSCSCs)from breast CSCs and performed the transcription profile to reveal the ***,we demonstrated that the behavior of therapeutic escape was regulated through the p38/MAPK signaling pathway,resulting in TSCSCs exhibiting enhanced motility and ***,blocking the p38/MAPK signaling pathway effectively reduced motility and metastasis ability both in vitro and in vivo,which were further supported by downregulated motility-related genes and epithelial-mesenchymal transition(EMT)-related proteins vimentin and *** obtained findings reveal the p38/MAPK pathway as a potential therapeutic target for TSCSCs and would provide profound implications for cancer therapy.

MicroRNAs 103 and 107： potential molecular links between diabetes and cancerand cancer

The prevalence of type 2 diabetes （T2DM）, which is characterized by insulin resistance, is increasingdramatically worldwide, and it has become a major public health issue. T2DM and cancer are common diseases that are frequently diagnosed in the same individual and the relationship of them appears to be complex. Epidemiologic data have shown T2DM is significantly associated with cancer, but potential biologic links between the two diseases are not fully understood. MicroRNAs （miRNAs） are endogenously expressed RNAs consisting of 20-24 nucleotides that influence the expression of hundreds of genes that involved in numerous biological processes, including organ development, lipid metabolism, differentiation, apoptosis, and brain morphogenesis. MiRNAs are now shown to be stably expressed in serum,~ plasma,2 or blood.3＇4 Moreover, the unique expression patterns of these circulating miRNAs are correlated with specific human diseases,5 including various types of cancer and diabetes. Therefore, miRNAs in serum or plasma are emerging as novel blood-based fingerprints for the detection of human cancers with diabetes, especially at an early stage. Accumulating studies have association between T2DM demonstrated a significant and tumorigenesis. One of the mechanisms underlying is insulin resistance, which contributes to the development of T2DM and cancer. High expression of miRNAs 103 and 107 （miR-103/107） leads to insulin resistance by down regulating caveolin-1, which is the direct target gene of miR-103/1076 and also a critical regulator of insulin receptor. Downregulation of caveolin-1 is susceptible to be tumorigenesis.7 Emerging evidence suggests the important role of miR-103/107 in the development and progression of breast and colorectal cancer patients,8＇9 which of them are associated with diabetes, yet their roles in linking diabetes and cancer are only now beginning to be recognized. Roles of mushroom polysaccharides in chronic disease management

Chronic diseases have drawn much attention as the primary cause of death and disability. In exploring novel sideeffect-free agents against chronic diseases, significant efforts have been devoted to mushroom polysaccharides due to their diverse biological activities. This work reviewed the structural features, biological performances and molecular mechanisms of mushroom polysaccharides in managing cancers, diabetes mellitus and cardiovascular diseases. The potentials of mushroom polysaccharides against chronic diseases highly depend on their structural features, including monosaccharide composition, molecular weight, the type and configuration of glycosidic bonds, degree of branching, the type of substituent pattern and chain conformation. Regarding their working mechanisms, shared and diseasespecific pathways were found. The three chronic diseases shared the regulation of specific signalling pathways and the adjustment of gut microbiota. In addition, the roles of transcription factors, receptors, enzymes, hormones and other functional proteins involved in the molecular mechanisms of mushroom polysaccharides against chronic diseases are first elaborated herein. The present review describes the state of the art of mushroom polysaccharides in treating chronic diseases and addresses the perspectives, and will further promote research on this topic.

MicroRNAs: a novel promising therapeutic target for cerebral ischemia/reperfusion injury?

To determine the molecular mechanism of cerebral ischemia/reperfusion injury, we examined the micro RNA（mi RNA） expression profile in rat cortex after focal cerebral ischemia/reperfusion injury using mi RNA microarrays and bioinformatic tools to systematically analyze Gene Ontology（GO） function classifications, as well as the signaling pathways of genes targeted by these differentially expressed mi RNAs. Our results show significantly changed mi RNA expression profiles in the reperfusion period after focal cerebral ischemia, with a total of 15 mi RNAs up-regulated and 44 mi RNAs down-regulated. Target genes of these differentially expressed mi RNAs were mainly involved in metabolic and cellular processes, which were identified as hub nodes of a mi RNA-GO-network. The most correlated pathways included D-glutamine and D-glutamate metabolism, the renin-angiotensin system, peroxisomes, the PPAR signaling pathway, SNARE interactions in vesicular transport, and the calcium signaling pathway. Our study suggests that mi RNAs play an important role in the pathological process of cerebral ischemia/reperfusion injury. Understanding mi RNA expression and function may shed light on the molecular mechanism of cerebral ischemia/reperfusion injury.
